摘要:
AbstractThe nuclear DNA of eukaryotes is organized into a series of loops each topologically anchored by elements of the nuclear matrix. Evidence is reviewed which indicates that the anchorage points of the loops are formed by transcriptionally active genes and that individual loops function as replicons. The data suggests a specific model for coupling of DNA replication and transcription in eukaryotes.

AbstractCollagen is the most abundant vertebrate protein and forms a stable fibrous architecture in connective tissues, such as bone, cartilage, skin and tendon. Much recent research has been directed towards an understanding of the molecular and cellular mechanisms involved in the synthesis and degradation of collagen, because a change in the normal balance, or an increased destruction of collagen, can cause loss of function of specialized tissues. This short review attempts to summarize present knowledge about the proteolytic destruction of collagen and the exciting new work on the factors that are emerging as important controlling agents. An understanding of the regulatory processes is yielding some important clues regarding the levels at which they can fail in pathological conditions and we speculate about evolving strategies for preventing uncontrolled resorption.

AbstractThe interconversion of key regulatory proteins between phosphorylated and dephosphorylated forms is an extremely versatile mechanism for reversible altering their activities, and in mammalian cells may be almost as common as allosteric regulation. It is now evident that protein phosphorylation is the basis of a complex network of interlocking systems which allow a variety of hormones and other extracellular signals, acting through just a few second messengers to coordinate biochemical functions.

AbstractThe two theories of pancreatic enzyme secretion, those of exocytosis and transmembrane flow, are described. Data thought to support the theory of transmembrane flow of single molecules from pancreatic acinar cells are first reviewed, and the conditions which could allow these data to be explained by the theory of exocytosis of enzyme quanta, i.e. secretory granules, are then discussed.The evidence suggesting short‐term modulation of the composition of pancreatic juice is also considered, and its possible explanations at the organ and cellular level are discussed, in the light of suggested theories on enzyme secretion pathway
