AbstractXenoantiserum to Sézary cell leukemia cells (ASS) was developed by immunizing rabbits with those cells and was absorbed with human red cells, liver, tonsil B cells, and cultured Raji cells. This reagent reacted by immunofluorescence with virtually all human thymus and T cells. In the thymus, medullary cells reacted more strongly with ASS than did cortical thymocytes. When immunoprecipitates that formed between ASS and125I‐labeled lymphocyte surface glycoproteins were analyzed by sodium dodecyl sulfate‐polyacrylamide gel electrophoresis, it was found that ASS precipitated a 72K molecular weight (MW) glycoprotein from T cells but not from B cells. On the one hand, it was shown by functional studies that T cells sensitive to the cytotoxic effect of ASS contained T cells that could aid the immunoglobulin synthesis of B cells induced by pokeweed mitogen. On the other hand, suppressor T cells induced by concanavalin A resided in those cells rather resistant to its cytotoxic effect. AbstractTo study the possible immunologic role of hand mirror lymphocytes (HML) in the control of Epstein‐Barr virus (EBV)‐infected B lymphocytes in patients with infectious mononucleosis (IM), we studied the in vitro interactions of these cells by time‐lapse video light microscopy. Peripheral blood T lymphocytes isolated from three patients in the early recovery phase of IM were mixed with their autologous EBV‐infected B cells. Motile lymphocytes with their characteristic hand mirror shape were observed to attach by their uropods to the B cells. The HML remained attached for variable periods ranging from 45‐75 min. Following detachment, B cells that were in contact with HML underwent lysis. Mixtures of T cells from healthy donors and EBV‐infected cell lines exhibited only rare uropod formation with no attachment or lysis of B cells. AbstractThe factors responsible for the switch from γ‐to β‐globin synthesis, which occurs in man in the perinatal period, are still unknown. In this study the relationship of fetal maturity (as measured by gestation age and fetal weight) to changing rates of globin synthesis was analyzed. The relationship of rates of erythropoiesis to the switch was also analyzed. Rates of globin chain synthesis were studied in blood from fetuses from 10‐41 wk gestation, in neonates up to 6 mo of age, and in a series of twins from 29‐40 wk gestation. The β/γ‐globin synthesis ratio in fetal blood remained relatively constant until 30‐34 wk gestation when an increase in β‐globin production was observed. Adult levels of globin synthesis were present in neonates at 6‐9 wk after birth. The γ/α‐globin synthesis ratio showed a reciprocal change, falling after 30 wk gestation. Globin synthesis ratios in fetuses and neonates from 32‐41 wk post‐conception showed a significant correlation with age (β/γ: p<0.001; β/α: p<0.01; γ/α: p<0.05), and there were significant correlations between the β/α and β/γ synthesis ratios and weight (p<0.05 for both ratios). AbstractThis study was undertaken to investigate the safety and efficacy of nonaspirin, nonsteroidal anti‐inflammatory drugs (NSAIDs) in controlling the pain and stiffness of hemophilic arthropathy. Data were collected from eight adult hemophilic patients in successive double‐blind controlled studies using choline magnesium trisalicylate (CMT) and ibuprofen (IPF). In seven patients, neither drug consistently affected bleeding times, platelet aggregation studies, frequency of joint hemorrhages, or frequency of factor infusions. In one patient, platelet aggregation appeared to be affected by CMT and bleeding time became markedly prolonged after IPF therapy. Three patients taking CMT and four patients taking IPF reported greater relief of pain and stiffness with the drugs than with placebos. AbstractThirteen cases of acute monocytic leukemia (AMoL) were studied morphologically, cytochemically, and immunologically in an attempt to determine similarities and/or differences between acute monocytic and lymphocytic leukemias. Single class surface immunoglobulins (IgG kappa) were seen in six cases. These markers are thought to represent serum immunoglobulins attached to the leukemic monocytes via the receptors for the Fc portion of immunoglobulin and are not synthesized by the cells as is the case with B lymphocytes. Receptors for complement were also noted. It appears that surface immunoglobulins and receptors for the Fc portion of the immunoglobulin and complement, which are found on several classes of normal lymphocytes and monocytes, may also be found on leukemic monocytes. AbstractA cell fraction enriched in immature granulocytes was obtained after fractionation of human bone marrow by a three‐step procedure. This fraction sediments at an average density of 1.073 and has a high content of myelocytes and metamyelocytes. It showed the highest specific uptake of etiocholanolone when compared to other bone marrow cell fractions. Autoradiographic studies demonstrated a higher number of silver grains over myelocytes and metamyelocytes than in other cell types. AbstractLight and electron microscopy of neutrophils from chronic neutrophilic leukemia (CNL) did not reveal differences from normal mature neutrophils. However, functional characterization of CNL cells showed marked differences when compared to normal cells. CNL neutrophils were much less viable in suboptimal conditions. Their survival was further reduced by autologous serum and was corrected by normal human serm. CNL cells showed very active phagocytosis, but their bactericidal activity was reduced in suboptimal conditions. The total content of lysozyme and β‐glucuronidase was lower in CNL cells compared to normal neutrophils, but the release of these enzymes from stimulated cells was much higher than normal. This observation is compatible with a marked lysosomal lability. Cells from the patients' peripheral blood and bone marrow showed excessive growth in CFU‐C assays. AbstractKinetics andw quantification of the sites of destruction of111‐Indium‐oxine‐labeled autologous platelets were investigated in eight patients with idiopathic thrombocytopenic purpura. The mean platelet count was 17 ± 9 × 109/liter; platelets were separated by differential centrifugation and labeled with 5.6 ± 2.5 MBq111In. Whole body and organ111In‐platelet distribution was quantitated with a scintillation camera and a computer‐assisted imaging system acquisition matrix. Areas of interest were selected with the computer and organ111In‐radioactivity expressed as a percentage of whole body activity. Mean platelet survival was 49.5 ± 29.6 hr and the survival curves were exponential. Equilibrium percentage organ111In‐radioactivity was (normal values in parentheses): spleen 33.7 ± 8.8(31.1 ± 10.2); liver 16.1 ± 9.5(13.1 ± 1.3); thorax 22.8 ± 3.7(28.2 ± 5.6). Percentage organ111In‐activity at the time when labeled platelets had disappeared from the circulation was: spleen 44.5 ± 16.4 (40 ± 16); liver 16.0 ± 11.5 (32.4 ± 7.2); thorax 19.7 ± 6.0 (17.7 ± 10.3). Thorax activity corresponds to bone marrow radioactivity. Three patterns of platelet sequestration were evident. Three patients had mainly splenic sequestration, two mainly hepatic sequestration, and three diffuse reticuloendothelial system sequestration with a major component of platelets destroyed in the bone marrow. AbstractA quantitative bioassay for serum erythropoietin in anemic patients was established with erythroid colony‐forming technique using methyl cellulose. Fetal mouse liver contains many erythropoietin‐dependent erythroid colony‐forming cells (CFU‐E) with the concentration as well as sensitivity to erythropoietin being the highest at 13–14 days of gestation. Linearity in the erythropoietin response curve for the number of colonies from CFU‐E in fetal livers of 13–14 days gestation was obtained for erythropoietin concentrations in the culture medium of 3.9 to 125 mU/ml. When newborn liver cells were used instead, the linearity occurred at 31.2 to 250 mU/ml. Measurement of erythropoietin by the erythroid colony‐forming technique for sera from patients with various anemias was compared with that of the assay method using radioiron incorporation into heme in the suspension culture of fetal mouse liver cells. There was a good correlation between the values of both assay methods (r = 0.856, p<0.001). AbstractA 59‐year‐old woman, after complete recovery from an episode of drug‐induced agranulocytosis, was found to sustain a chronic absence of recognizable mature and immature eosinophils in blood and bone marrow during a follow‐up period of 8 years. Her bone marrow and peripheral blood cells cultured in vitro were able to produce normal numbers of eosinophil colonies. The present disorder was thus not caused by a lack of EO‐CFU‐C, nor by a defect in EO‐CSA, nor by an immunologically mediated mechanism acting on mature or immature eosinophils or on EO‐CSA producing cells.
