ContentsThe surge of LH and the process of ovulation were determined in nine gilts by means of repeated laparoscopy after synchronization of estrus and stimulation of ovulation using 50 μg Gn‐RH (Gonavet “Berlin‐Chemie”). The mean duration between Gn‐RH application and the beginning and the completion of ovulation was 35.5 ± 2.7 h and 39.7 ± 1.9 h, respectively. A period of 31.1 ± 2.2 h and 36.4 ± 2.5 h was between the peak of LH and the beginning and the completion of ovulations. The mean duration of ovulation, including the first up to more than 80% of ovulated follicles, was 5.9 ±1.7 h. About 20% of all follicles/gilt ovulated in a period of 2.9 ± 1.1 h. The duration of the ovulation from 20% up to more than 80% of the follicular population was 3.0 ± 0.7 h.Inhalt: Beziehungen zwischen derdurch exogenes Gn‐RH induzierten LH‐Ausschüttung und der Ovulationsdauer bei JungsauenDie LH‐Ausschüttung und der Ovulationsverlauf mittels wiederholter Laparoskopie wurden bei 9 Jungsauen nach Brunstsynchronisation und Stimulation der Ovulation mit 50 μg Gn‐RH (Gonavet “Berlin‐Chemie”) bestimmt. Die Zeitdauer zwischen der Gn‐RH Applikation und dem Beginn bzw. Abschluβ der Ovulation betrug 35,5 2,7 h bzw. 39,7 1.9 h. Zwischen dem LH‐Peak und dem Beginn bzw. Abschluβ der Ovulation wurde ein Intervall von 31,1 2,2 h und 36,4 2,5 h ermittelt.Die durchschnittliche Ovulationsdauer, gemessen von der Ovulation des ersten bis zu mehr als 80% der Follikel, betrug 5,9 1,7 h. ContentsThis study describes qualitative and quantitative maturation specific changes in polypeptide synthesis of porcine oocytes. Changes were detected by means of high resolution two dimensional polyacrylamide gel electrophoresis. Various new polypeptides appeared in the protein patterns of in vivo and in vitro matured oocytes while some of the polypeptides synthesized in immature oocytes disappeared in the protein patterns of in vivo matured oocytes. In uivo matured oocytes showed a markedly different protein pattern in comparison to in vitro matured oocytes. The polypeptide synthesis of porcine oocytes during in vitro maturation was not influenced by the addition of estradiol‐17β and progesterone together or subsequently to the culture medium. It is suggested that the failure in vitro matured oocytes to acquire a normal fertilizability and developmental capacity might be caused by the inability to induce adequate changes in the synthesis of specific polypeptides during culture. Supplementation of estradiol‐17β and progesterone to the culture medium alone seems not to be sufficent for this induction.Inhalt: Polypeptidsynthese bri in vivo und in vitro gereiften SchweineoozytenIn der vorliegenden Arbeit werden qualitative und quantitative Veränderungen in der Proteinsynthese von in vitro und in vivo gereiften Oocyten des Schweines beschrieben. Mit Hilfe der zweidimensionalen Gelelektrophorese konnte ermittelt werden, daβ während der in vivo und in vitro Reifung zahlreiche neue Proteine von der Eizelle synthetisiert werden. Im Vergleich zu ungereiften Oocyten wurde ferner beobachtet, daβ die Synthese von einigen Proteinen im Verlaufe der in vivo Reifung sistiert. Zwischen den Proteinsynthesemustern von in vitro und in vivo gereiften Eizellen zeigten sich deutliche Lrnterschiede, die auch durch den Zusatz von Östradiol‐17β und Progesteron zum Reifungsmedium nicht beeinfluβt werden konnten. Das abweichende Proteinprofil der in vitro gereiften Eizellen deutet auf eine unvollständige zytoplasmatische Reifung dieser Oocyten kin, die vermutlich für deren fehlende oder nur mangelhaft ausgebildete Befruchtungs‐ und frühembryonale Entzoicklungsfähigkei

ContentsIn the ovary of mammals, inflammatory‐like reactions are absent in spite of pronounced regressive processes occurring during the sexual cycle. Thus, we presumed local changes of chemotactic activity. 28‐day‐old golden hamsters were killed 6, 12, 24, 36, 60, and 84 h after induction of superovulation by human chorionic gonadotrophin (hCG). Extracts obtained from unstimulated and stimulated ovaries as well as from control organs were examined whether they acted on chemotaxis and adherence of circulating blood granulocytes. The modified Boyden chamber technique was used and 3 μm2‐ or 0.4 μm2‐pore size filters, respectively, were mounted into the chambers. Ovarian extracts were chemotactic on their own. Chemotactic activity caused by endotoxin‐activated serum was reduced in the presence of 6–60 h‐extracts. Reduction became apparent at a statistically significant level by use of the 36 h‐extract. Preliminary results on adherence demonstrated more adherent blood granulocytes using the 36 h‐extract compared to other extracts. Studied by scanning electron microscopy (SEM), adherent granulocytes were moderately or strikingly spread under the influence of the 36 h‐extract. As concluded for the golden hamster, ovarian extracts appear to exert influence on chemotactic activity and adherence of blood granulocytes. The influence varies with the functional status of the ovary.Inhalt: Ovarielle Extrakte beeinflussen die Chemotaxis von Blutgranulozyten. Eine Untersuchung bei superovulierten GoldhamsternIm Säugerovar fehlen entzündungsähnliche Reaktionen trotz ausgeprägter regressiver Prozesse, die während des Sexualzyklus zu beobachten sind. Deswegen vermuteten wir lokale Veränderungen der chemotaktischen Aktivität. Bei 28 Tage alten Goldhamstern wurden Superovulationen durch menschliches Choriongonadotropin (hCG) ausgelöst. Die Tiere wurden 6, 12, 24, 36, 60 bzw. 84 h später getötet. Extrakte, hergestellt aus unstimulierten und hCG‐stimulierten Ovarien sowie aus Kontrollorganen, wurden überprüft, ob sie die Chemotaxis und Adhärenz zirkulierender Blutgranulozyten beeinfluβten. Die Versuche wurden mit Hilfe der modifizierten Boyden Kammer durchgeführt, in die Filter mit 3 μm2bzw. 0.4μm2groβen Poren eingesetzt waren. Ovarielle Extrakte besaβen chemotaktische Aktivität. Die chemotaktische Aktivität, ausgelöst durch Endotoxin‐aktiviertes Serum, wurde durch 6–60 h‐Extrakte verringert. Stark verringert war die chemotaktische Aktivität in Gegenwmt des 36 h‐Extraktes. Wie vorläufige Ergebnisse zur Adhärenz zeigten, fanden sich mehr adhärente Zellen beim Gebrauch des 36 h‐Extraktes, verglichen mit den übrigen Extrakten. Im Rasterelektronenmikroskop waren adhärente Granulozyten mäβig oder stark ausgebreitet, wenn der 36 h‐Extrakt untersucht wurde. Somit scheinen ouarielle Extrakte die chemotaktische Aktiuität und Adhärenr uon Blutgranuloryten des Goldhamsters zu beeinf

InhaltDie Mikroinjektion von Spermien kann im Zusammenhang mit der Spermientrennung zur Erzielung der Befruchtung rnit kleinsten Spermamengen von Nutzen sein. In einem ersten Experiment wurden Befruchtungs‐ und Furchungsraten nach Spermainjektion (SPI) mit einer Kontrolle nach in vitro‐Befruchtung (IVF) nach 24 h in vitro Kultivierung ermittelt. Dabei erwiésen sich die Ergebnisse der IVF‐Gruppe (n = 40; 57,5 und 42,3 %) denen der SPI‐Gruppe überlegen (n = 84; 41, 7 und 28,6 %). In einem weiteren Experiment wurden mit insgesamt 345 Oocyten in 6 Behandlungsgruppen der Einfluβ der Spontanaktivierung sowie derjenige eines speziellen Zuckermediums zur Vergröβerung des perivitellinen Saftraumes (PVS) auf die Befruchtungsund Furchungsraten untersucht. Hier konnte nach SPI eine der IVF gleichwertige Befruchtungs‐ und Furchungsrate (n = 52; 61,5 und 51,9%) erzielt werden. Trotr meiner hohen Spontanaktivierungsrate bei der entsprechenden Placebo‐Kontrolle (n = 66; 28,8 und 19,7%) wird aufgrund der ausreichend hohen und signifikanten Differenz davon ausgegangen, daβ ein hoher Anteil der zytologisch als befruchtet ermittelten Oocyten und Furchungsstadien normal befruchtet war. Der Zusatz des Disaccarids Trehalose zum Manipulationsmedium mit dem Ziel der Erweiterung des PVS und damit Erleichterung der Injektionsprozedur führte zu einer markanten und signifikanten Erhöhung der Spontanaktivierungsrate bis auf 50%. Andererseits waren die Befruchtungs‐ und Furchungsraten, insbesondere in der IVF‐Gruppe stark reduziert.Contents: Microinjection of motile bovine spermatozoa into the perivitelline space of oocytesMicroinjection of spermatozoa may be a suitable tool for fertilization of bovine oocytes with small amounts of spermatozoa, used in connection with fractionation of semen. In the first experiment fertilization and cleavage rates were investigated after 24 hrs of in vitro cultivation of spermatozoa‐injected (SPI) and in vitro fertilized (IVF) oocytes. Better results were obtained in the IVF‐group (n = 40; 57,5 and 42,3%) than in the SPI‐group (n = 84; 41, 7 and 28,6%). In the second experiment 6 treatments with 345 oocytes were performed and the influence of spontaneous activation of oocytes after the injection of medium into the PVS (placeboO was investigated. Also the influence of trehalose‐medium to enlarge the PVS of the oocytes prior to injection was tested. The fertilization‐ and cleavage rates increased up to 61,5 and 51,9 % after SPI and reached 50,7 and 42,6% after IVF. Unfortunately 28,8% of oocytes were activated spontaneously in the placebo‐group. Nevertheless the differences between the SPI‐ and placebo groups were significant and this concluded that a large rate of oocytes in the SPI‐goup was really fertilized. The addition of trehalose to the medium for the enlargement of the PVS was connected with an increase of the spontaneous activation rate up to 50%. ContentsTesticular morphology of Shorthorn beef bulls at the age of 14 months (controls, n = 3) was compared with that of animals prepuberally actively immunized against testosterone (anti‐T, n = 3) or estradiol‐17β (anti‐E, n = 3). The volume percentage of the Leydig cell compartment in anti‐T‐animals was significantly higher than in the 2 other groups. Nuclear and cellular volumes of a single Leydig cell of anti‐E‐bulls were significantly smaller than in the other groups. Absolute number of Leydig cells per testis in anti‐T‐bulls was significantly higher than in controls. There were no significant differences in the relative volume percentages of Leydig cell organelles between the 3 groups. However, since anti‐T‐bulls have the highest Leydig cell mass per testis (10.8 ± 2.2 g) as compared with controls (6.0 ± 1.1 g) and anti‐E‐bulls (5.6 ±0.3 g), the Leydig cell mitochondrial, nuclear and SER masses per testis, which reflect the steroidogenic capacity of the organ, are also higher in anti‐T‐animals than in controls or anti‐E‐bulls.Inahlt: Morphologische Untersuchungen an den Hoden von aktiv gegen Testosteron und Oestradiol‐17β immunisierten ShorthornbullenUntersucht wurden die Hoden von Shorthornbullen im Alter von 14 Monaten, welche prapuberal aktiv gegen Testosteron oder Oestradiol‐17β immunisiert worden waren. Das relative Volumen an Leydigzellen war bei anti‐T‐Tieren signifikant gröβer als bei Tieren aus den beiden anderen Gruppen. Kern‐ und Zellvolumina einzelner Leydigzellen waren bei anti‐E‐Bullen signifikant kleiner als bei Tieren aus den beiden anderen Gruppen. Die absolute Anrahl von Leydigzellen pro Hoden war bei anti‐T‐Tieren signifikant gröβer als bei Kontrolltieren. Die relativen Volumina ausgewählter Leydigzellorganellen(Kern, Mitochondrien, glattes eR)unterscheiden sich in den 3 Gruppen nicht signifikant. ContentsIn vitro and in a field trial the boar sperm compatibility of the antibiotic additives lincomycin and spectinomycin (= Lincospectin®) was investigated. The semen was extended with Kiev or Androhep media containing the antibiotic additives. Lincospectin® concentrations up to 4500 μg/ml of extended semen did not have any deleterious effect on sperm motility and acrosome integrity. Likewise, no negative influence on sperm quality could be seen using 450 μg Lincospectin (150 μg lincomycin and 300 μg spectinomycin) in combination with 500 IU penicillin and 500 IU streptomycine or with 250 μg gentamycin. The field inseminations based on 248 first services using Lincospectin in combination with penicillin and streptomycin revealed no negative effects on farrowing rate and litter size.Inhalt: Untersuchungen zur Spermien‐ und Fruchbarkeitsverträglichkeit von Lincospectin® Lösung als Antibiotikumzusatz zu flüssigverdünnten EbersamenDie Spermienverträglichkeit der Antibiotika Lincomycin und Spectinomycin (=Linco‐spectin) wurde in vitro und im Feldversuch untersucht.
