Contents:From a group of thirteen 3‐year‐old Hanoveranian maiden stallions ejaculates were collected on 5 consecutive days and frozen each in 3 different packaging forms. Fresh, resuspended and frozedthawed semen was tested for motility and morphology with special regard to acrosomal integrity. Concerning packaging form no differences were found between 4 ml maxi straw, flattened 1.7 ml straw and 0.5 ml straw. Contents:Prolactin (PRL) was isolated from human amniotic fluid and than fractionated on Concanavalin A‐Sepharose (ConA‐S). Two fractions were tested for the3H‐thymidine incorporation in the DNA of bovine granulosa, oviductal and stromal cells as well as splenocytes. All cell types reacted to PRL I or PRL II in a dose dependent manner. In general the effect of PRL I was greater than PRL II, and low concentrations of both enhanced the cell proliferation more than higher doses. The incorporating effect of3H‐thymidine under the influence of PRL I was higher in stromal cells and splenocytes than in granulosa and oviductal cells. PRL II did not have such a stimulating effect, even the presence of 14 ng/ml resulted in a decrease of3H‐thymidine incorporation in granulosa cells. The reason of this different intensity of tested PRL variants may depend on the molecular heterogeneity. Contents:Repeatability and heritability coefficients were estimated for ejaculate volume, mass movement, individual motility, sperm concentration, first and second post‐freezing control of motility, number of doses frozen, total number of spermatozoa and total number of motile spermatozoa per ejaculate in the first ten ejaculates of 175 Swiss Holstein and 673 Simmental young bulls (200 pure Simmental, 473 Simmental x Red Holstein crosses). Data were analysed using the univariate individual animal model with effects of interval between consecutive collections, age of bull at collection, season of collection, breed group of Simmental bulls according to percentage of Red Holstein genes, bull (within breed group) and permanent environment. Crossbred bulls had a better performance in all traits. Repeatability coefficients ranged from 0.28 to 0.46. They indicate that the prediction of semen quality in young bulls of the Swiss Holstein and the Simmental breed is possible to some extent. Heritabilities in Swiss Holstein bulls were generally very low and did usually not exceed 0.10, but standard errors of these estimates were high. Contents:Cumulus‐oocyte complexes were matured for 24 hours in the presence or absence of bovine herpes virus‐1 (BHV‐1, 106TCID50/ml), fertilized in vitro and then co‐cultured on oviductal cells to the blastocyst stage. The percentages of oocytes which cleaved were 48% (n = 905) for oocytes matured in the presence of BHV‐1, and 51% (n = 1004) for the control oocytes. Comparing BHV‐1‐treated to control, the percentages of cleaved oocytes which developed to the blastocyst stage were 29% and 31%, respectively (p>0.05). Contents:An insemination trial using frozen semen is described. The freezing procedure was slightly modified from the Hannover method. The insemination dose consisted of 7 medium straws containing approx 1 109spermatozoa. A total of 28 mares of the Norwegian Trotter breed were inseminated during the 1991 season. During oestrus the mares were examined at 12 hour intervals, and the insemination was carried out after detection of ovulation. The pregnancy rate was 43% after the first insemination, increased to 68% after second and further to 75% after the third and last insemination. The foaling rate was 61.5%.

Contents:Investigations were performed to determine I) Cyclic changes in uterine immune response of buffalo‐cows that could be responsible for increased susceptibility of the diestrous uterus to infection, 2) The effect of some genital disorders on the immune response. The number and phagocytic capacity of uterine leucocytes as well as the immunoglobulin contents revealed significant cyclic variations. These parameters were obviously decreased in the uteri of buffalo with bilateral smooth inactive ovaries. On the other hand, marked increases were recorded in both uterine leucocyte counts and immunoglobulin contents in the uteri of buffalo with cystic ovarian disease, endometritis, pyometra and cervical retention cysts. Phagocytic capacity showed obvious increase in case of cystic ovarian disease while decreased in other studied disorder

Contents:Prolactin (PRL) was purified from human amniotic fluid and further fractionated on ConA Sepharose 4 B. The activities of the retarded migrating glycosylated PRL‐ConA‐S were examined in splenocytes from mice using three different test systems in comparison to PRL‐standard (PRL‐ST). These assays included the conductometrical cytometry for measurement of cell count and volume, the3H‐thymidine incorporation and the IgG‐expression. The sensitivity of splenocytes ro the PRL‐forms was different in the various assays and dose‐dependent. A small PRL‐ConA‐S dose caused a significant stimulatory effect on the cell count, the3H‐thymidine proliferation and the cell volume, whereas the IgG‐expression was only nonsignificantly increased in higher concentration. Even in a moderate physiological concentration (0.15 mlU/ml) PRL‐St exterted an inhibitory effect on the cell proliferation which was directly demonstrated by cell counting.
