Contents: At the Bundesanstalt Wels tests with frozen‐thawed boar semen in plastic straws were conducted. The influences of straw volume, method of freezing and thawing extender were investigated. The straw volumes of 1, 0 and 0, 5 ml showed significantly better thawing results in a preliminary trial than the 5, 0 ml Macrotub. When semen was frozen in a computerized freezer with automatic seeding, all tested straw. volumes gave significantly better results than the straws frozen in static N2‐vapor. A ready to use commercial extender was used for thawing with good results to simplify the handling of deep frozen semen for on farm insemination. In the main trial 60 ejaculates from 10 boars were frozen in 1, 0 ml straws in the computerized freezer. Three in vitro parameters for fresh semen (motility, osmotic resistance and keeping quality under standard conditions) and two parameters for thawed semen (motility and percentage of sperm with normal apical ridge) were recorded and correlated. The osmotic resistance test proved to be well suited as means of predicting the fitness of an ejaculate for deep freezing, but the other two fresh semen parameters showed poor correlation with the parameters for thawed semen. From the parameters for thawed semen a freezing score was derived as a measure of the freezability of single ejaculates and boars. A preliminary insemination trial gave satisfying farrowing rates.Inhalt: Zur Tiefgefrierung von Ebersamen: Untersuchungen zum Einfluß von verschiedenen “Straw”‐Volumina, Einfriewerfahren und AuftaubedingungenAnder Bundesanstalt Wels wurden Tiefgefrierversuche mit Eberspermain Kunststoffpailletten durchgeführt. Die jeweiligen Einflüsse von Paillettenvolumen, Gefrierverfahren und Auftauverdünner wurden untersucht. In Vorversuchen erwiesen sich die Paillettenvolumina 1, 00 und 0, 5 ml gegenüber den 5,0 ml Makrotüb hinsichtlich Kopfkappenintegrität und Auftaumotilität signifkant überlegen. Die Auftauergebnisse der im computergesteuerten Freezer mit automatischem Seeding eingefrorenen Proben waren für alle untersuchten Paillettengröβen signifikant bis hoch signifikant besser, als die Werte der im statischen Stickstoffdampf eingefrorenen Proben. Als Auftauverdünner wurde ein handelsüblicher Fertigverdünner gewählt um eine besonders für den Eigenbestandsbesamer wichtige einfache Handhabung der TG‐Besamung zu gewährleisten. Im Hauptversuch wurden 60 Ejakulate von 10 Ebern verwendet und in 1, 0 ml Pailletten im programmierbaren Freezer eingefroren. Drei Frischsamenparameter (Motilität, osmotische Resistenz und Haltbarkeit unter Laborbedingungen) und zwei Auftausamenparameter (Motilität und Kopfkappenintegrität) wurden erhoben und korreliert. Dabei enwies sichder osmotische Resistemtest als für die Beurteilung von Frischsperma hinsichtlich TG‐Eignung gut geeignet, während die beiden anderen Frischsamenparameter keine bzw. nur schwache Korrelation zu den Auftauergebnissen aufwiesen. Aus den Auftausamenparametern wurde eine “Einfrierbarkeitszahl” als Maβ für die Eignung des Spermas zur Tiefgefrierkonservierung der einzelnen Eber bzw. Ejakulate erstellt. InhaltEin Zusatz von Alkylsulfaten bzw. von TRITON X 45 in sehr geringer Konzentration zum Medium für die Gefrierkonservierung von Schafbocksperma führt zu verbesserten Auftauergebnissen. In einem Medium ohne Kryoprotektiva wird die Empfindlichkeit gegenüber Kälteschock bzw. osmotischen Schock verringert. Der vermutlich membranstabilisierende Effekt von Natriumhexadecylsulfat hatte keinen Einfluβ auf das Befruchtungsergebnis nach Insemination gefrierkonservierten Schafbockspermas.Ein Zusatz von N‐Acetyl‐cystein bei der Gefrierkonservierung von Schafbocksperma verbessert Motilität und Resistenz nach dem Auftauen signifikant. Andere Vitalitätsparameter werden nicht beeinfluβt. Im Besamungsversuch konnte der positive Effekt dieses Zusatzes bestätigt werden, ohne daβ damit praxisrelevante Ergebnisse erreicht wurden.Contents: Cryopreservation of ram spermatozoa in the presence of surface active substancesThe addition of alkylsulphates or TRITON X 45 in very low concentrations to diluents for the cryopreservation of ram semen leads to an improvement of after‐thawing results. In a diluent without cryoprotectives the sensitivity against cold shock as well as osmotic shock is diminished. The membrane stabilising effect of sodium‐hexadecylsulphate has no influence on the fertility rate after insemination of cryopreserved ram semen.The addition of N‐acetyl‐cysteine in cry opreservation of ram spermatozoa increases the motility and lifetime in a resistance test after thawing significantly. Contents:The in vitro developmental potential of blastomeres from early porcine embryos war studied. Two‐, 4‐, and 8‐cell embryos were separated into component blastomeres and replaced into zonae pellucidae in the following configurations (designated by number of blastomeresl original cell stage): half embryos (1/2, 2/4, and 4/8); quarter embryos (1/4 and 2/8); and eighth embryos (1/8). Culture was done in 50 μl droplets under paraffin oil in either Whitten's medium supplemented with 4 mg/ml bovine serum albumin (BSA) and 10% fetal calf serum (FCS) or in the same medium supplemented with porcine oviductal cells. Cultures were observed on a daily baris when the morphology was recorded and at the end of culture (144—168 h after the expected time of ovulation), the embryos were stained with Hoechst 33342 and cell counts made. Generally, no significant differences in developmental rate were found between the two culture system and the data was pooled. Blastocyst formation rate in half embryos was 23%, 28%, and 44% for 1/2, 2/4, and 4/8 embryos, respectively. The respective blastocysts contained an average of 24.3 ± 4.4 and 28.8 ± 2.0 versa 41.2 ± 6.4 cells (p<0.05 and p<0.05). Blastocyst formation rate in quarter embryos was 15% and 35% (p<0.01) for 1/4 and 2/8 embryos, respectively and they possessed an average of 17.0 ± 1.8 and 22.8 ± 1.4 cells (p<0.02), respectively. Blastocyst formation rate in 1/8 embryos was 9% and the blastocysts averaged 13.0 ± 1.3 cells. Two‐,4‐, and 8‐cell control embryos formed blastocysts at rates of 22%, 85%, and 64%, respectively, and had average cell counts of 45.8 ± 10.8, 69.1 ± 4.0, and 75.4 ± 7.3, respectively. The timing of the initiation of blastocoel formation was similar for reaggregated and control embryos that started culture at the same stage. This along with the cell counts of reaggregated embryos at cavitation, which were proportional to the reduction in cell mars (7.2 ± 0.6, 12.6 ± 1.9, and 25.6 ± 2.8 for quarter, half and control embryos, respectively) supports the assumption that in pigs blastocoel formation occurs independent of the number of cells present.Inhalt: Entwicklung von frühen in vitro kultivierten Schweineblastomeren nach Abtrennung und ReaggregationEs wurde die Entwicklungspotenz von Blastomoren aus frühen Schweineembryonen studiert. Zwei‐, Vier‐ und Achtzell‐Embryonen wurden in ihre Blastomerenbestandteile zerlegt und nachfolgendem Schema in Zonae pellucidae zurückverbracht: Halbierte Embryonen (1/2, 2/4, 4/8) Viertelembrynnen (1/4 und 2/8) und Achtelembryonen (1/8). Die Kulturphase erfolgte in 50 μl Tropfen, die mit Parafinöl überschichtet waren. Es wurde Whitten's Medium, entweder mit 4 mg/ml BSA und 10% FCS verwendet oder das Medium mit porcinen Eileitenellen supplementiert. Die Kultur wurde täglich überpüft und die Morphologie beurteilt. Am Ende der Kulturzeit (144–168 Std nach dem berechneten Ovulationszeitpunkt) wurden die Embryonen mit dem Hoechstfarbstoff 33342 gefärbt und die Zellen gezählt. Signifikante Unterschiede in der Entwicklungsrate zwischen den beiden Medien wurden nicht gefunden und die Daten wurden zusammengefaβt. Die Blastozystenbildungsrate der Embryonenhälften betrug 23% und 44% für 1/2, 2/4 und 4/8 Embryonen. Diese Blastozysten enthielten im Mittel 24,3 ± 4,4 und 28 ± 2,0 gegenüber 41,2 ± 6,4 Zellen (p ≤ 0,05). Die Blastozystenbildungsrate aus Embryoversuchen betrug 1.5% und 35% (p ≤ 0,01) für 1/4 und 2/8 Embryonen. Im Mittel waren 17,0 ± 1,8 und 22,8 ± 1,4 Zellen (p ≤ 0,02) enhvickelt worden. Die Blastozystenbildungsrate in 1/8 Embryonen betrug 9% und durchschnittlich wurden 13,0 ± 1,3 Zellen in den gebildeten Blastozysten nachgewiesen. Zwei‐, Vier‐ und Achtzell‐Embryonen der Kontrollgruppe formten zu 22%, 85% und 64% Blastozysten und die mittlere Zellzahl betrug 45,8 ± 10,8, 69,1 ± 4,0 und 75,4 ± 7,3. Inhalt:Morphometrisch untersucht wurden die Nebennieren von 27 Kühen mit und ohne medikamentelle Geburtseinleitung bzw. nicht tragender Kontrolltiere (Gruppe I (n = 7): 10 mg Triamcirwlon −6,5 d ante partumund10 mg Flumethason −30 h ante partum; Gruppe II (n = 6): 1 ml NaCl (Placebo) −6,5 d ante partum und 10 mg Flumethason −30h ante partum; Gruppe III (n = 4): tragend, keine Vorbehandlung; Gruppe IV (n = 10): nicht tragende Kontrolltiere). Eine Geburtsinduktion mit Glukokortikoiden führt zu einer signifikanten Verkleinerung der Kerngröβen sowie einer Zunahme der Zellkernzahl pro definierter Flächeneinheit in der Zona fasciculata und einer signifikanten Verschmälerung von Zona fasciculate/Zona reticularis. Inhalt:Azyklische Hafingerstuten (n = 4), erhielten während des saisonalen Anöstrus 2 mal täglich HCG (750 IE) i.m. verabreicht. Die Medikationsdauer war wirkungsbezogen (10 bis 20 Tage). Die Stimulation wurde nach erfolgter Ovulation oder bei Ausbleiben der Follikelreijiung und Eifreisetzung abgebrochen. Sexualaktive Stuten (n = 3) blieben ohne Medikation und dienten als Kontrolle. Die Befunderhebung an den Ovarien erfolgte ultrasonographisch. Jede Stute wurde 2 mal täglich auf eine halbe Stunde zur Protokollierung des Sexualverhaltens mit einem Hengst aufeine gemeinsame Koppel gebracht. Bei drei von vier Stuten entwickelte sich während der Behandlung an einem Eierstock ein Follikel. Ein Tier reagierte erst nach einer Prostaglandinmedikation auf die HCG‐Stimulation mit einer Follikelanbildung und Ovulation. Die präovulatorische Gröβe der Follikel schwankte zwischen 3,2 und 5,2 cm (= 4,3 cm). Die Deckbereitschaft der Stuten war kein sicherer Hinweis auf die Präsenz eines ovulationsgerechten Follikels. Zwei HCG‐stimulierte Stuten hatten Perioden der Duldung, die nicht mit den Ovarbefunden korrelierten. In beiden Fällen wurde die Kopulation über mehrere Tage (5 bzw. 7) ohne nachweisbaren Follikel gestattet. Die Langzeitstimulation mit HCG war in allen Fällen erfolgreich und lieβ keine negativen Auswirkungen erkennen. Alle im Versuch stehenden Stuten konzipierten und brachten gesunde Fohlen zur Welt.Contents: Long‐term HCG treatment for ovarien stimulation in mares during seasonal anoestrousHaflinger mares (n = 4) were treated with hCG (750 IU) twice a day during seasonal anoestrous. The length of the medical attention was due to the effect (10 and 20 days). The stimulation stopped as soon as ovulation occured or follicular development and ovulation failed to appear. Mares with active ovaries (n = 3) remained untreated and were used as control animals. The ovaries were checked upon by ultrasonographic monitoring. Furthermore breading behaviour was recorded by teasing each mare with a stallion twice a day for half an hour on a pasture. During treatment three of four mares developed follicles on their ovaries. After a singular prostaglandin injection the fourth mare also responded to the medication by growing an ovarien follicle. The preovulatory diameter ranged from 3,2 to 5,2 cm (x = 4,3 cm). Behavioural oestrous was not reliably linked to periovulatory follicles. Two of the stimulated mares showed periods of behavioural oestrous without corresponding ovarien condition. Both of them were mounted by the stallion for some days (5 and 7 days) without the slightest incidence of follicles. Contents:Semen samples, collected from bulls pesistently infected with bovine viral diarrhea virus (BVDV) and containing BVDV (titer 105‐ 106TCID50/ml), were subjected to sperm separation procedures (washing, swim up, Percoll gradient, glass wool filtration, glass beads filtration) that are commonly used prior to in vitro fertilization (IVF) to determine if these procedures would yield spermatozoa free from BVDV. The final sperm pellets from frozen and fresh ejaculates were tested for the presence of BVDV by the immunoperoxidase technique; all tests were positive for BVDV in the range of 103‐ 104TCID50/ml). The study shows that when semen containing BVDV at the level of 105‐ 106TCID50/ml) is used for IVF, the virus is not completely removed by any of the simple physical methods commonly used to prepare sperm for IVF.Inhalt: Feldversuch das bovine Diarrhoe Virus (BVDV) aus Bullensperma durch Swim up oder andere Trennverfahren in Verbindung mit der In vitro Fertilisation zu entfernenSpermaproben, die von Bullen gewonnen wurden, welche dauerhaft mit dem Virus der bovinen Virus‐Diarrhoe (BVDV) infiziert waren und einen Titer zwischen 105‐ 106TCID50/ml enthielten, wurden verschiedenen Trennverfahren unterzogen (Waschen, swim up, Percoll gradient, Glaswollenfiltration, Glaskugelfiltration). Diese Verfahren werden üblicherweise für die Vorbereitung zur In vitro Befruchtung verwendet, und es sollte geprüft werden, ob diese Verfahren auch das Sperma von dem BVD‐Virus befreien können. Das endgültige Spermapellet von gefrorenen/aufgetauten und frischen Ejakulaten wurde in Gegenwart des BVD‐Virus und mit Hilfe der immunoperoxidase Technik getestet. Inhalt:Am 27.8.1992 wurde beim Besamungsverein Neustadt (BVN) in Neustadt a.d. Aisch ein geklontes Fleckviehkalb geboren. Die Identität der Abstammung konnte eindeutig gesichert werden. Der erfolgreiche Kerntransfer in einem Versuchsprogramm soll belegen, daβ sich nach der Fusion einer Blastomere aus einem Spenderembryo mit einer in vitro gereiften und enukleierten Empfängeroozyte ein Embryo entwickeln kann, der nach Transfer auf ein Empfängertier zur Geburt eines gesunden Kalbes führt.Contents: A cloned calf was born in Neustadt ad.
