Seventy‐six weanling gnotobiotic rats of the Sprague‐Dawley strain were monoinfected in groups withActlnomyces naeslundii, Actinomyces viscosus or Streptococcus mutans.The animals were killed at intervals up to 84 days and the maxillae and mandibles examined histologically. Periodontal disease, as evidenced by the presence of plaque, migration of the epithelial attachment and regression of the alveolar bone was sometimes evident after 28 days and in all cases extreme by 84 days. No organisms were seen in the gingiva and inflammatory changes were slight. Periodontal disease was most marked between the first and second maxillary molars, the mandibular gingiva, apart from the presence of plaque, being normal over the experimental period. Bone loss was gauged by labelling the bone with3H proline, and osteoclasts were detected by the acid phosphatase technique.In another experiment, 44 conventional weanling cream hamsters were superinfected withA. naeslundii. Periodontal disease developed more slowly than in the gnotobiotic rats, usually taking about 160 days to be severe.In both experiments the pathological changes were similar. The gnotobiotic rats developed periodontal disease over the same time sequence with all three organisms. Bone loss was not accompanied by the presence of osteoclasts and seemed, in this model, to be more due to a gradual cessation of bone formation than to active resorption. Sterile extracts of whole human microbial plaque released substantial latent collagenolytic activity in gingival collagenase preparations, homogenates of polymorphonuclear leukocytes, and tissue macrophages. The plaque activation factor was thermolabile, nondialyzable, and had essentially no inherent collagenolytic activity.

Electron microscopic examination of the walls of periodontal pockets revealed that connective tissue mast cells were distributed throughout the tissue. These cells had a variety of shapes, and were often in close association with remaining collagen fibers, as well as with smaller fibrils and other collagen fragments. Totally distrupted, necrotic mast cells were also observed which had morphologically intact granules in continuity with the extracellular space.

The buccal and lingual gingival tissues in the vervet monkey were compared. Blocks of fresh tissue, consisting of mandible and associated soft tissues, were prepared for histological examination. Coronal and transverse sections, 7 and 25 μm thick, were cut through the full thickness of the mandible and stained with haematoxylin and eosin, picromallory and a variety of elastic stains. Photomicrographs of step serial sections were enlarged to standard magnification for histometric analysis. The buccal epithelium is thicker and shows a greater degree of keratinization than the lingual epithelium. The rete pegs are more prominent and penetrate deeper into the lamina propria on the buccal aspect. There appears to be two sets of elastic fibres in the submucosa on the buccal aspect and only one on the lingual. The buccal elastic fibres consist of a sub‐epithelial plexus of fine fibres and a longitudinal set of thicker fibres. The lingual fibres are situated sub‐epithelially only. This investigation was designed to test the co‐destructive factor hypothesis that a traumatic lesion subjacent to marginal periodontitis alters the progression of the periodontitis and produces angular bony defects. Marginal periodontitis was produced around 28 mandibular teeth in 7 squirrel monkeys by tying a silk ligature at the gingival margin to facilitate plaque accumulation. Six months after inducing periodontitis a severe traumatic lesion was produced subjacent to the marginal periodontitis in 15 of these teeth by thermal injury via the root canal. These specimens (Group I) were analyzed from 3 days to 6 months after injury subjacent to the periodontitis. The duration of periodontitis in the remaining 13 teeth (Group II) was the same as Group I allowing a comparison between the groups. It was concluded that the presence of a severe traumatic lesion and its sequelae subjacent to a marginal periodontitis did affect the progression of the periodontitis when evaluated by apical migration of the junctional epithelium and loss of alveolar bone. Angular bony defects occurred only when periodontitis was induced around isolated tooth surfaces and were related to the presence or absence of the traumatic lesion. In continuation of a series of investigations designed to evaluate co‐destructive factors in periodontal disease, a single severe mechanical injury was produced subjacent to marginal periodontitis by wedging a toothpick between second and third bicuspids in squirrel monkeys. Marginal periodontitis alone was induced on the contralateral side for the same period of time for comparison of the progression of periodontitis with and without the subjacent traumatic lesion. This study was designed to evaluate the effect of four commercially available dressings (Coe, PPC‐Eugcnol, PPC‐Non Eugenol, and Ward's Wondr Pak) on a monolayer of cells, employing the cell culture assay plate assembly. Monolayers of L929 fibroblasts were grown out on the cell culture assay plates and stained with a vital stain, .01 % Neutral Red, 8 mm discs of prepared periodontal dressings, as well as sterile filter paper and high density polyethylene discs, which served as controls, were placed on the monolayer. Observations were made at 1, 2, 6, 8 and 24 hour intervals, and measurements of the toxic zones were recorded. Results indicate that all periodontal dressings tested in this system demonstrated definite cytotoxicity. Both macroscopically and microscopically, the first signs of cell damage and/or death were observed adjacent to Ward's Wondr Pak andPPC‐Non Eugenol dressings. The periodontal condition of 162 splinted approximal abutment tooth surfaces was compared with that of 162 contralateral tooth surfaces which were not fastened together. The Plaque Index (Silness&Löe 1964), the Gingival Index (Löe&Silness 1963), and the Margin Index (Silness 1970a) were recorded. Pocket depths were measured as explained by Glavind and Löe (1967). In bridge recipients who had received periodontal treatment and instruction to improve oral hygiene and who had no subgingival retainer margins in the embrasure areas between the splinted teeth, the periodontal condition was no better or worse than that of the contralateral area with which it was compared. The daily post‐operative use of topical chlorhexidine application has been compared to daily toothbrushing. Periodontal defects were experimentally created in the bifurcations of lower premolars in dogs. The defects were treated by surgical debridment. Chlorhexidine application and toothbrushing were performed post‐operatively for six weeks on contra‐lateral pairs of teeth. The amounts of connective tissue and bone regeneration were evaluated from microscopic sections of the bifurcation areas.
