At various times within a 24 hour period 26 while albino rats aged from 3 to 15 months received an intraperitoneal injection of colchicine (1 mg/kg body weight) and were sacrificed 31/2 hours later. Serial sections were prepared for the palate, tongue, gingiva and ear and were stained with hematoxylin and eosin. The mitotic activity was assessed for the epithelium of these tissues using three different mitotic index systems (mitoses/1 mm2surface area; mitoses/1000 cells; mitoses/1 mm basement membrane). For the same tissues in another 10 rats the number of nucleated epithelial cells and the length of the basement membrane were related to a reference unit of the epithelial surface. The coefficients of variation for these observations were calculated. The application of the traditional indices (mitoses per 1000 cells or 1 mm of the basement membrane) introduced an appreciable variability into the measurements and produced misleading results when the mitotic activity of various epithelia were compared. The index system expressing the number of mitoses relative to a reference unit (1 mm2) of the outer surface of the epithelium was found to be the most reliable method for assessing mitotic activity in stratified squamous epithelium.

Manifestations of inflammatory periodontal disease in a. colony‐maintained marmoset were studied. The disease was characterized clinically by an acute inflammatory response including redness, edema, exudation and spontaneous hemorrhage. Histopathologically, the cellular response observed was comprised of neutrophils and mononuclear cells, some of which could be definitively identified as macrophages. The lymphocyte‐plasma cell infiltrate which comprises a major feature of the disease in man was not a prominent feature in the marmoset. Indeed, only insignificant numbers of these cells were found even with electron microscopy. Thus, the host tissue response in the marmoset appears to differ significantly from that seen in man, and reservations must be exercised in the use of the marmoset as a model of the human form of the disease. The observation that the important clinical and histopathologic manifestations of chronic periodontal disease can occur in this animal without the presence of an extensive lymphocyte‐plasma cell infiltrate suggest that, contrary to current concepts, the activity of these cells at the local site may not be essential to the induction and progress of periodontal di

In the present investigation anin vivomethod is described for studying the chemotactic activity elaborated by factors in human dental plaque. Plaque was sampled from the buccal and lingual surfaces of teeth from dental students, who had refrained from mechanical tooth cleansing during one 4 and one 8 day period. The plaque samples were suspended in 0.15 M NaCl, homogenized, and centrifuged at 12,100 × g for 30 minutes at 4°C. The plaque supernatant was then separated from the cellular pellet and sterile filtered through a 0.45 μ Millipore filter. The chemotactic activity elaborated by the filtrates was examined in (i) Boyden'sin vitrochamber (ii) a wound chamber model described by Lundgren and Lindhe (1970). The results showed (i) that the wound chamber method is well suited for studying leukocyte chemotactic activity elaborated by dental plaque (ii) that factors in dental plaque stimulate the emigration mainly of neutrophils (iii) that there is no increase in the amount of chemotactic factors/mg plaque with increasing age of the plaque. After four weeks of repeated irrigations of intact labial gingival sulci of rabbit mandibular incisors with horseradish peroxidase (HRPO), antiperoxidase antibody was demonstrated ultra‐structurally in the cistemae of the endoplasmic reticulum of some plasma cells in the gingival lamina propria. Antibody was found also in all cell types of the plasmacytic series in the regional lymph nodes. It was present in the basement membrane and the adjacent connective tissue and in the extraneous cell coat material of crevicular epithelial cells. Continued topical irrigations eventually caused an allergic type inflammatory response which, if allowed to resolve, would reappear fifteen minutes after irrigating the gingival sulci with HRP

Two groups of adult female Hollzman rats, injected subcutaneously 5 times a week with low and high doses of norethynodrel plus mestranol for 91 days failed to gain weight at the same rate as the controls. In measuring both the mandibular and maxillary bone loss there was it trend showing the greatest amount of bone loss in the controls, moderate loss in the low dosage and the least loss in the high dosage. The differences of Total mandibular bone loss between the controls and rats treated with high dose of Enovid‐E® were statistically significant. However when the bone loss was measured as a percentage of total mandibular size the differences were not significant. A 2% silver nitrate staining method for delineating the cemento‐enamel junction was developed. Under the present experimental condition; the injection of Enovid‐E® compared to the controls did not demonstrate adverse effects on the gingiva or on alveol

The capillary arrangement under the crevicular and attachment epithelium was observed in relation to the degree of inflammatory cell infiltration in clinically healthy marginal periodontium in dogs. Both factors were investigated in adjacent tissue segments. The vascular bed was studied on corrosive specimens and the infiltration On histologic specimens stained by haematoxylin and eosin. There was to some extent a relation between the pattern of crevicular vessels and the degree of inflammatory cell infiltrate. Variability of the vascular bed was found, which was not related to the degree of infiltration. The anatomy of the vascular bed did not depend on the particular tooth, surface, arch or animal studied.

Fifty‐six, female, BNL strain mice, 26 weeks of age, received a modified “gingiveclomy” excising, the gingival papilla mesial to the maxillary left first molar. The right maxillae served as the uninjured control site. Additional non‐injured mice were utilized as further controls. At varying time intervals, groups containing 4 mice each were sacrificed over a 3 months experimental period. One hour prior to sacrifice, each animal received a subcutaneous injection of 1 uCi of H3TDR/gm of body weight. At sacrifice, both left and right maxillae were dissectedd out and prepared for histologic and autoradiographic study. Following injury, the highest proliferative activity was encountered at the epithelial wound edge. This activity became prominent about 8 to 15 days after injury, while the adjacent crestal epithelium demonstrated a markedly increased activity as early as 16 hours after wounding. The reformed junctional and crevicular epithelium, and adjacent fibroblasts, still exhibited elevated activity at one month post‐injury and returned to control range values between one and three month following wounding. The remaining fibroblasts, osteoblasts and cementoblasts around the wound periphery showed markedly elevated activity as early as one to two days post‐injury which returned to control levels between 2 to 4 weeks aft

One hundred 11–15 year old clinic patients were examined, and the presence or absence of unequivocal gingivitis and periodontal pocketing was recorded separately for each permanent tooth, From the full‐mouth data, each subject was also scored using combinations of teeth recommended for partial recording with the PMA, GI, and PDI index systems. Spearman's Rank Correlation Coefficient (rs) was selected, after inspection of the frequency distributions, to express the associations between the partial recording and full‐mouth scores. From the values of rs, the relative efficiency of each partial recording combination was computed, in terms of the number of subjects required to provide a statistical test of similar power to that provided by a full‐mouth examination of 100 subjects. The GI combination proved to be the most efficient. Values for rsof 0.899 for gingivitis and 0.846 for periodontitis were obtained between this combination and the full‐mouth scores. It was estimated that 124 and 139 subjects, respectively, would be required for gingivitis and periodontitis assessments, using the GI selection, compared with 100 for a full‐mouth examination. Three examiners who had achieved inter‐examiner agreement using the PMA and Gingival Index parted for four months. After this period of time without contact only two examiners were still comparable when using each index. A training programme for use with each index was drawn up and used by each examiner to train a selected pupil. Two examiners were able to train their pupils so that there was inter‐examiner agreement between pupil and instructor. The third pair were un‐able to achieve consistent agreement with either index. The relative effectiveness on plaque removal and gingival health of three brushes containing 6: 10 nylon filaments of varying filament diameter and tuft density was evaluated in an un‐supervised double‐blind, cross‐over trial conducted on twenty subjects. The two brushes with filaments of 0.28 mm diameter were greatly superior to the brush containing filaments of 0.14 mm diameter. Although the brush of fine diameter filaments contained twice and thrice as many filaments as the others this did not compensate for the narrower filament. Differences between the two brushes with the broader filaments were not significant and increasing the number of tuffs by 40 per cent did not confer any benefits.
