The effect of indomethacin on plasma renin activity (PRA) and sodium excretion was examined in normal conscious dogs before and during water diuresis. PRA did not show significant change with the administration of indomethacin (5 mg/kg): PRA was 5.00 ± 1.69 ng/ml/h during control periods and was 5.56 ± 1.93 ng/ml/h (p > 0.1) 80 min after the administration of indomethacin. Mean arterial blood pressure (MABP) was 100.5 ± 5.1 mm Hg during control periods and was 93.3 ± 6.6 mm Hg (p > 0.1) 80 min after the administration of indomethacin. Administration of indomethacin into these dogs undergoing water diuresis did not change the clearance of either inulin or p-aminohippurate. Sodium excretion (UNa 0.1). UNaV was 89.7 ± 22.9 µEq/min during control periods and was 73.2 ± 12.6 µEq/min 80 min after the administration of indomethacin. In conscious rats inhibition of kallikrein formation with aprotinin suppressed urinary prostaglandin (PG) E2 excretion. This was associated with a decrease in inner cortical perfusion in the absence of changes in GFR or hippuran clearance. During acute expansion of the extracellular fluid volume (ECFV) with isotonic saline PGE2 excretion significantly increased but was suppressed during stable ECFV expansion. With ECFV expansion GFR increased, while total and regional cortical perfusion remained unchanged. Aprotinin suppressed the early rise in PGE2 excretion and reduced GFR and hippuran clearance as a result of decreased outer cortical perfusion. It is suggested that integrity of the kinin-prostaglandin system represents an important prerequisite for the functional adaptation of the kidney to acute expansion of the ECFV with isotonic saline.

We evaluated the effect of intrarenal vasodilatation on renin release in the indomethacin (2 mg/kg) treated pentobarbital anesthetized dog. Pretreatment with indomethacin resulted in a significant decrease in renal plasma flow (RPF) without change in glomerular filtration rate (GFR). Intrarenal infusion of prostaglandin E2 (PGE2, 2 µg/min) and acetylcholine (ACh, 40 µg/min) completely restored RPF in these dogs. Following PGE2 administration a significant increase in absolute sodium excretion (UNaV), renal vein renin activity (RVRA), and renin secretory rate (RSR) were observed. Despite similar effects on RPF and UNaV, acetylcholine did not alter renin release as measured by plasma renin activity (PRA), RVRA or RSR. Effects of hypocalcemia on Ca2+ metabolism of microsomes from rat kidney cortex were studied. Microsomal activities of Ca2+-ATPase, Mg2+-ATPase and (Na++K+)-ATPase increased 30–33 days after Ca2+ restriction or low Ca2+· vitamin D depletion, but alkaline phosphatase activity decreased. Calcium repletion in diet and injection of lα-hydroxycholecalciferol or 1,25-dihydroxycholecalciferol were partially successful in returning Ca2+-ATPase and Mg2+-ATPase activity, but not (Na++K+)-ATPase and alkaline phosphatase activity, to normal. A study of the influence of different diets (rich and deficient in polyunsaturated fatty acids) and of indomethacin on the mean count of renomedullary interstitial cell (RIC) lipid granules was undertaken. A correlation between dietary linoleic acid (LA) content (the main precursor of endogenous prostaglandin biosynthesis) and the number of granules in RIC was demonstrated: in comparison to the salt-loaded rats fed standard pellet diet, the number of RIC lipid granules in salt-loaded rats fed LA in excess increased, while in salt-loaded rats on LA-deficient diet the RIC granularity tended to decrease. Indomethacin treatment caused an increase in RIC granularity in salt-loaded rats fed a standard pellet diet and in those fed LA in excess, but had no effect in salt-loaded rats fed LA-deficient diet.

Whole kidney and micropuncture techniques were employed to investigate renal function in early pregnant (day 6 of gestation) and virgin Munich-Wistar rats. No changes were seen in glomerular filtration rate (GFR) or in any of its determinants at day 6 of pregnancy. In separate studies, the measurement of plasma volume showed that whereas a marked expansion had occurred by day 12 of pregnancy only slight increases in plasma volume were apparent at day 6. These findings, together with evidence from a previous study which indicated that a significant increase had occurred in GFR by day 12 of pregnancy, suggest a relationship between GFR and plasma volume in the pregnant rat.

The early effect of triiodothyronine (T3) on renal handling of K+ and Na+ in hypo-thyroid and euthyroid rats was studied. Individual rats were kept in metabolic cages. Sodium and potassium intake as well as urinary Na+/K+ ratio and excretion were determined daily for the first 2 days after one injection of T3 was administered to hypothyroid and euthyroid rats. Pre- and post-injection values for the urinary Na+/K+ ratios in the hypothyroid rats were consistently higher than the euthyroid rat. However, in both hypothyroid and euthyroid rats, no change in K+ and Na+ intake, K+ and Na+ excretion, and urinary Na+/K+ ratio was observed at 24 or 48 h after T3 injection. The results of these studies suggest the absence of an early response of aldosterone secretion to T3.

The components of acidic glycosaminoglycans (AGAG) prepared separately from cortical and medullary tissues of human male kidneys were analyzed by enzymatic methods. Heparan sulfate was the main component, comprising approximately half of total AGAG, and was significantly higher in the cortex than in the medulla. In the cortex, the major AGAG was heparan sulfate (67% of total AGAG) followed by dermatan sulfate (17%). In the medulla, the main AGAG was also heparan sulfate (45%) followed by hyaluronic acid (15%) and chondroitin 4-sulfate (16%). A possible role of the different AGAG composition in the two regions is discussed.
