Antibody against fusarochromanone (TDP‐1) was obtained from rabbits after immunization with TDP‐1 conjugated to bovine serum albumin (BSA). An indirect enzyme‐linked immunosorbent assay (ELISA) using TDP‐1‐ovalbumin conjugate as the antigen coated on to the microtiter plate was used for monitoring the antibody liter. For toxin detection, a direct competitive ELISA in which TDP‐1 was conjugated to horseradish peroxidase (HRP) was used. Competitive direct ELISA revealed that the antibody had about 5.6 and 4.5 times greater binding efficiency for monoacetyl fusarochromanone (TDP‐2) and diacetylated TDP‐1 than TDP‐1. The concentration causing 50% inhibition of binding of TDP‐1‐HRP to the antibody by TDP‐1, TDP‐2 and diacetyl‐TDP‐1 were 2.8, 0.5 and 0.62 ng/ml, respectively. For the analysis of fusarochromanones in wheat and barley, the toxins were first extracted from the commodities with 100% methanol. A small aliquot of the extract was dried, acetylated, diluted in buffer and then analyzed directly by ELISA. The overall recovery for fusarochromanone in the wheat and barley samples spiked with TDP‐1 in the concentration range of 20 to 500 ppb were found to be 97% and 103.4% with cv of 15% and 11.2% for barley and wheat, respectively.

摘要:
The application and validation of a highly sensitive and specific enzyme immunoassay for the analysis of atrazine residues in natural water samples, plant extracts and soil extracts is described. The EIA is performed on microtitre plates. The range of detection of atrazine analysis lies between approximately 1 ng/l and 10 μg/l. Several synthetic and environmental water samples, plant extracts and soil extracts were analysed with the EIA. A close correspondence was found between the results of the EIA and GC or HPLC measurements, respectively.

Four haptens analogous to phenylurea herbicides were synthesized. Immunogens and coating proteins prepared by coupling these haptens to human serum albumin and ovalbumin, respectively. Antisera raised in rabbits immunized with three of the immunogens bound strongly to microtitration plates coated with homologous hapten, such that competitive assays were developed using heterologous coating proteins. The assay systems were characterized with respect to specificity to 13 related compounds and one selected for application to the determination of monolinuron, diuron, and linuron in foods. Satisfactory recrovery data were obtained by direct analysis of methanol extracts of foods fortified with 40–160 ng/g monolinuron or 0.25–2.0 μg/g diuron. Solvent partitioning and silicic acid column chromatography were only partially successful in removing interferences in the determination of linuron.

Two triazine derivatives, ametryn sulphoxide (2‐ethylamino‐4‐isopropylamino‐6‐methyl‐sulphoxide‐1,3,5‐triazine) and dichloroatrazine (2,6‐dichloro‐4‐isopropylamino‐1,3,5‐triazine) were conjugated to bovine serum albumin (BSA) and used for the immunization of BALB/c mice. Hybridomas were produced by cell fusion of immune spleen and mouse myeloma cells (PAI‐B3AG8.I). After screening with a competitive enzyme‐linked immunosorbent assay (ELISA), four anti‐triazine monoclonal antibodies from permanent hybridoma cell lines were selected for further characterization. Cross‐reaction studies with the antibodies developed against the ametryn sulphoxide conjugate showed strong affinities to terbutryn and prometryn. ELISAs with antibodies from clone AS‐K1F4 reached a detection limit of approximately 0.1 μg/l for terbutryn, and with AS‐K1A11 antibodies a limit of 0.3 μg/l for prometryn was attained. Cell lines from mice immunized with the dichloroatrazine‐conjugate produced antibodies with the highest affinities to aziprotryn. The detection limit of the corresponding ELISA was 1 μg/l (clone DC‐C3K5). The scope of the monoclonal antibodies for use in pesticide residue analysis is discussed with respect to polyclonal antibodies.

摘要:
A brief immunological resume of each of the marine toxins is reviewed in this report. These toxins are brevetoxin (PbTx), palytoxin (PTX), okadaic acid (OA), paralytic shellfish toxins (PSP) and ciguatoxin (CTX). The immunological and biotechnical procedures developed for each of these toxins are reviewed. The cross‐reactivity of the polyether‐type toxins (PbTx, OA and CTX) are discussed. The capabilities of immunological technology for assessment of toxins in seafood products appears promising and will probably be widely used in the seafood industries.
