Testicular development was studied in prepubertal boys with retractile testes. Testicular volume, the diameter of the seminiferous tubules and the number of spermatogonia in the tubules were decreased in cases of unilateral retractile testis, when compared with values for the contralateral normally descended testis. On the other hand, in patients with a unilateral retractile testis and contralateral inguinal testis, there was no difference in the developmental parameters between the two testes. These results suggest that the retractile testis has developmental failures characteristic of a cryptorchid testis and therefore requires orchiopexy.

The fertility profile of 307 Thai male volunteers whose wives were currently in early pregnancy was established by genital examination, semen analysis and serum hormone analysis. Ninety percent of the subjects had testicular volumes of 12–25 ml (mean 17.2 ml) which was found to relate to body weight, height and the ponderal index as well as to the sperm count. However, values for semen analysis were significantly below the recognised standard for Caucasian males. There was an inverse relationship between testicular volume and the serum concentration of FSH, LH and prolactin. It is suggested that a common protocol for male fertility assessment should be used to establish standard parameters for racially different male population

A monoclonal anti‐human sperm antibody, raised against an acrosomal antigen and indicated to recognize in boar sperm the serine protease, acrosin, stained in human spermatozoa a 50 Kd antigen and several others in the region 24–34 Kd by immunoblotting. The 50 Kd band and the region of 30–34 Kd showed proteolytic activity by zymographic enzyme detection. The fate of the antigen was studied in the acrosome reaction induced by the calcium ionophore A23187. In control incubations 69.5 ± 14.2% (mean ± SD) of the spermatozoa had intact acrosomal staining according to indirect immunofluorescence using this antibody whereas in acrosome‐reacted samples only 21.0 ± 2.0% of the sperm were stained. Another marker for the acrosome, peanut agglutinin‐lectin (PNA), was used to detect the acrosome with similar results. Acrosome reactions were verified by electron microscopy. The effects of temperature and incubation time were examined on the concentrations of the following seminal plasma constituents in ejaculates from seven healthy volunteers: prostatic acid phosphate (PAP), citric acid, testosterone, prostaglandin E (PGE), fructose, protein, putrescine and the polyamines spermidine and spermine. The results indicated that the majority of glandular products were stable up to 3 h after ejaculation when stored at room temperature, with the exception of the prostatic diamine putrescine, which appeared to undergo rapid metabolism. Prolonged periods of incubation resulted in a gradual reduction in the levels of fructose and a significant increase in the activity of PAP and the levels of spermidine. On the basis of these findings it appears that an evaluation of the functional integrity of the accesory sex glands can be carried out on semen up to 3 h after ejaculation.

The addition of epidermal growth factor (EGF) at nanomolar concentrations to cultures of Sertoli cells from 16‐day‐old rats induced more than a 2‐fold stimulation of lactate production after 12 h of incubation. The effects of EGF, insulin and FSH on lactate production were very similar and the concentrations that produced half maximal stimulation were 11, 22 and 25 ng/ml or, in molar terms, 1.8, 3.5 and 0.6 nM for EGF, insulin and FSH, respectively. Immunohistochemistry was used to localize alphafoetoprotein (AFP) in 1 μm plastic sections of embryonic testes in order to investigate the cellular basis of AFP production in murine teratocarcinomas. The effect of seminal plasma inhibin on ornithine decarboxylase activity (ODC) in the rat prostate was studied. A microcapillary method was developed to measure the viscosity of small volumes of undiluted epididymal fluid. Fluid from the cauda epididymis registered 82 ± 17 centipoise which was much more viscous than fluid from the caput region (8 ± 2 centipoise). Initiation of sperm motility was strongly suppressed in the viscosity range of 7 to 150 centipoise. A significant increase in the viscosity of fluid from the caput region was observed when immobilin in the fluid binds with a lectin from Jack fruit (Artocarpus heterophyllus).
