AbstractNo intracellular enzyme possessing milk‐clotting activity was found in the mycelium ofAspergillus niger.There was a time‐lag in the clotting stage on using small amounts of the culture filtrate in the reaction mixture. Dilution of milk caused a decrease in the milk‐clotting activity and this was attributed to several factors. Phosphate buffer was found to accelerate the enzymatic coagulation of milk when the reaction mixture had a pH less than 6.3. The addition of CaCl2to buffered reaction mixtures caused decrease in pH and time of milk‐clotting. The extent of any decrease depended on the amount of CaCl2added, the type and pH of

AbstractDie durch Permethylierung des Polysaccharidteils der O‐Antigene vonShigella flexneri2 (Serotyp 1 b) und 1290/63 (Variante Y), Totalhydrolyse, Reduktion und Peracetylierung gewonnenen Alditolacetate wurden gaschromatographisch getrennt und massenspektrometrisch zugeordnet. AbstractThe effect of the exudates ofAzotobacter chroococcumin nitrogen‐free liquid medium and in sterile soil, on its ability to fix nitrogen was studied. Cell‐free culture filtrates and water extract of sterile soil inoculated withAzotobacterwere fractionated into ether soluble and insoluble aliquots, concentrated and chromatographically separated into ten consecutive zones. These were eluted and tested for their effect on nitrogen fixation byAzotobacter.UV‐fluorescence, and chemical tests for indoles, reducing substances, amino acids, and hydroxyl groups were made. Detection of gibberellins and water soluble vitamins was attempted. A biological test for activity of indolic compounds was carried out.The results showed thatAzotobacterproduces biologically active indolic compounds in culture and soil, acting as a significant stimulation for the process of nitrogen fixation. Not all the zones of the chromatogram which were stimulatory for nitrogen fixation were the same for coleoptile growth and vice versa. Amino acids in some cases were associated with non‐significant stimulation of nitrogen fixation. AbstractBei Phosphatmangelzellen vonCandida utiliswurden durch Thermoinaktivierung und Aciditätscharakteristik zwei strukturgebundene Pyrophosphat‐Phosphohydrolasen gekennzeichnet, die bei pH 2 bis pH 7 exogenes Pyrophosphat in Orthophosphat überführen.Das Aciditätsoptimum des Enzyms I liegt bei pH 2,5, das des Enzyms II bei pH 6; bei gemeinsamer Wirkung der Enzyme I und II liegt das Optimum bei pH 2,5.Das Enzym II ist bereits nach 10 min Vorbehandlung bei Temperaturen über 70 °C weitgehend, das Enzym I jedoch erst bei entsprechender Behandlung über 90 °C inaktiviert. AbstractBacillus subtilisforms a xylanase complex which does not depend on induction by xylane or xylose in the medium.The enzyme production is influenced by the kind of the carbon source, probably due to catabolic repression. The regulation of enzyme formation is complex in nature, because the C/N/P‐Relation in the medium, cell mass, and growth phase influence the amount of enzyme in the culture fluid. Maximal enzyme production was achieved at high but balanced amounts of C/N/P, although growth was strongly reprimed under these conditions. AbstractInRhodopseudomonas spheroidesandRhodospirillum rubrumlack of thymidine kinase activity has been shown to be responsible for the failure of labelling DNA by thymidine or by thymine plus a deoxyribosyl donor, and for the obvious impossibility of selecting thymine auxotrophes by the aminopterin or trimethoprim method.Fluorodeoxyuridine, afterin vivophosphorylation known as inhibitor of the formation of thymidine monophosphate, becomes inhibitory to light‐induced pigment syntheses inRhodopseudomonas spheroides.This inhibition is overcome by uracil but not by thymidine, indicating that the inhibition by fluorodeoxyuridine is brought about via splitting this compound and incorporation of the fluorouracil moiety into some RNA fraction

AbstractDNA of the temperate bacteriophage SP02 is active in transfection experiments. Competent recipient bacteria can also be lysogenized by such DNA extracted from mature phages. When phage DNA was isolated from the lysogenic strainBacillus subtilis168M (SP02), the lysogenization frequency was higher than in similar experiments with DNA from mature phages.

AbstractDie temperatursensitive Mutantets33‐6 vonBacillus subtilisSB 19 wächst bei 46 °C als Filament und bei 30 °C als normales Kurzstäbchen. Die wichtigsten Makromolekül synthesen der Mutantets33‐6 und des Wildstammes SB 19 verlaufen ähnlich.In Rohextrakten aus Kurzstäbchen ist die Aminoacyl‐tRNS‐Synthetasen (AAS)‐Aktivität hoch, ein Nachweis dieser Enzyme in Rohextrakten aus Filamenten ist nicht möglich. Durch Temperaturwechselexperimente (Umsetzen wachstumsaktiver Filamente von 46 °C auf30 °C) wird Zellteilung induziert und die AAS‐Aktivität nimmt zu. Der WildstammB. subtilisSB 19 zeigt bei 30 °C und 46 °C AAS‐Aktivität. Eine Hemmung der AAS‐Aktivität durch Temperaturbehandlungin vitro(46 °C) gelingt bei 30 °C‐Rohextrakten der Mutantets33‐6 nicht, wenn das Enzym durch ATP und Aminosäuren geschützt wird. Durch Temperaturvorbehandlung der 30 °C‐Rohextrakte der Mutantets33‐6 steigt die AAS‐Aktivitätum 170%. Dieser Effekt kann bei Rohextrakten vonEscherichia co
