AbstractL‐Tryptophan did not exert any influence on peptide alkaloid formation in an ergotamine and in an ergosine‐accumulatingC. purpureastrain. A different picture was observed in a series of relatedC. purpureastrains. Tryptophan showed a slight stimulatory effect on the ergotoxine producer Pepty 695/S. A blocked mutant of it, designated as Pepty 695/ch which was able to accumulate secoclavines gave similar results. In a high‐yielding elymoclavine strain Pepty 695/e, the progeny of the former one, tryptophan up to a concentration of 25 mm stimulated remarkably clavine biosynthesis. Furthermore, tryptophan could overcome the block of synthesis by inorganic phosphate. Increased specific activities of chanoclavine cyclase but not DMAT synthetase were observed in cultures of strain Pepty 695/e supplemented with tryptophan. 5‐Methyltryptophan and bioisosteres of tryptophan were ineffective in alkaloid stimulation. AbstractGrowing cells of blue‐green alga,Anacystis nidulans, were treated with lysozyme in osmotically stabilized nutrient medium. Their development and regeneration ability was studied in liquid media. During 5–8 hrs about 95% of cells were converted to osmotically sensitive spheroplasts having a residual wall layer on their surface. The highest viability of spheroplast population was achieved using egg‐white lysozyme for spheroplast preparation and gradual dilution by nutrient medium instead of centrifugation for lysozyme removal. Most of such prepared spheroplasts survived for 3 days of cultivation in nutrient medium, and started to die gradually from 3 to 10 days. Spheroplasts were growing spherically, about 30% of them were growing for 8 days up to 40‐fold increase in volume. Multiplication of thylakoid cisternae in growing spheroplasts was proved by freezeetching. Electron microscopy further revealed newly synthesized amorphous walls of isodiametrical appearance by growing spheroplasts resembling the original cell walls of cells. In liquid media blue‐green alga spheroplasts ofAnacystis nidulanscan grow for a number of days, however, only regeneration of incomplete wall occur unable to promote reversion of spheroplasts

AbstractAcinetobacter calcoaceticus69‐V is not able to grow on carbohydrates or its derivatives and to use them as sole carbon sources. On the other hand, the partial oxidation of several monosaccharides (glucose, galactose, mannose, xylose, arabinose, ribose) by intact cells has been demonstrated by acid production and measurements of oxygen consumption.Glucose and fructose are taken up by intact cells. Different transport systems exist for the uptake of both hexoses. The “apparent”KM‐values for the membrane transport amount to 1.5 · 10−3and 1.7 · 10−4moles/l for glucose and fructose, respectively. Gluconolacton only inhibits the transport of glucose, whereas the transport of both hexoses is inhibited by uranylacetate. Other monosaccharides and metabolic inhibitors are without effect. An exchange of intracellular against extracellular glucose has not been found.Gluconate and CO2have been identified as extracellular products of glucose oxidation after an incubation period of more than 5 min, whereas gluconate and 6‐phosphogluconate have been detected as intracellular products. No oxidation products have been found using fructose as substrate.No activity of hexokinase, phosphofructokinase, pyruvate kinase, and 6‐phosphogluconate dehydrogenase have been detected in cell‐free extracts of sonicated cells. An aldose dehydrogenase coupled with an electron transport system occurs in the membrane fraction. The presence of this enzyme was demonstrated by the reduction of dichlorophenolindophenol as well as by oxygen consumption which is inhibited by cyanide.The aldose dehydrogenase exhibits a broad substrate specificity against different aldohexoses and aldopentoses differing by theirKM‐values. AbstractIn strongly acidic medium (70% HClO4) streptothricins form a fluorophore (λex= 312 nm; λem= 381 nm) with unknown structure. AbstractThe phosphorylation of dihydroxyacetone by means of the enzyme dihydroxyacetone kinase (DHAK) is a key reaction in the assimilation of methanol in yeasts.
