AbstractSpontaneous mutants ofSaccharomycopsis lipolyticawere selected and partially characterized. Several antibiotics and antimetabolites were used for selection of spontaneous resistant mutants fromSaccharomycopsis lipolytica.The frequencies of such mutants were mainly arranged between 1 × 10−7and 5 × 10−6mutants per cell. But one class of glucosamine resistant mutants (GAMRA) occurred more frequently. Among the resistant mutants different types of dominant and recessive resistant mutants could be observed.UV light was used for inactivation of cells and induction of mutants fromS. lipolytica.Comparing four haploid strains only small differences were detected in sensitivity to UV light. UV light at a dosage of 135 J/m2was applied to increase the mutant frequencies in three haploid strains. Besides auxotrophic, temperature sensitive and colony morphology mutants, some new mutant types like small colony forming mutants, red‐brown coloured mutants, allylalcohol, glucosamine, 2‐deoxyglucose or nystatin resistant mutants, hitherto not described forS. lipolytica, were isolated and partially char

AbstractComperative studies on the activities of isocitrate lyase (ICL) and malate synthase (MS) were carried out withSaccharomycopsis lipolyticaincubating the yeast on media with different carbon sources. When cells were incubated in minimal medium with glucose, the activities of both enzymes were very low. In contrast, in minimal medium with acetate enhanced enzyme activities could be demonstrated. It is probably that the synthesis of ICL is repressed in presence of glucose. Furthermore the activity of ICL was inhibited by tricarboxylic acid cycle intermediates like succinic acid and oxalacetic acid. It was concluded that the syntheses of enzymes are derepressed. When cells ofSm. lipolyticawere incubated in minimal medium with acetate, a high enzyme activity is evident. Synthesis of ICL on acetate was inhibited by cycloheximid and actinomycin D. The results were discussed comparing them with data obtained from other organisms.

AbstractDuring the turimycin fermentation hydrolytic enzymes are excreted responsible for orthophosphate release from phosphate‐containing dissolved and undissolved complex medium constituents. Following a phosphate‐limited growth period the phosphate release leads to a second growth period (diauxic growth). Depending on the rate of phosphate release the length of the lag phase of diauxic growth changes in different fermentations.The resulting second growth period is correlated with a transient delay in the formation of turimycin, of phosphatases and of nucleases.The amylolytic activities are formed already within the first hours after the beginning of fermentations. Phosphatases, nucleases and protease are excreted parallel to turimycin formation after extracellular phosphate limitation in presence of ammonia and glucose. AbstractCommonly the TCA cycle fulfils an anabolic and a catabolic function in case of aerobic chemo‐organoheterotrophic nutrition. In methylotrophic growth the TCA cycle is dispensable as a bioenergetic pathway. This is reflected by properties of citrate synthase in facultative methylotrophic bacteria. Two citrate synthases, a ‘chemoorganoheterotrophic’ one, which is inhibited by NADH (or ATP inAcetobacterMB 58), and a ‘methylotrophic’ one, which is not or less affected by energy indicators, were found inPseudomonas oleovorans, PseudomonasMS,PseudomonasMA, andAcetobacterMB 58. The concentration of these citrate synthases depends on the manner of nutrition. Bacteria with ICL‐negative‐variant of the serine pathway and with ribulosebisphophate pathway seem to possess only a ‘chemoorganoheterotrophic’ citrate synthase. AbstractThe lysogenic state of the methylotrophic strainAcetobacterMB 58/1 is completely demonstrated by curing and lysogenization experiments. During these investigations we found that some phenotypic characteristics are modified by the presence or loss of the prophage MO 1. It could be shown that changes of the serological behaviour, the adsorption of the phages and the sensitivity against oxytetracycline are caused by lysogenic conversion. The phenotypic alterations of the bacterial cells induced by the phage genome are the result of modifications of the lipopolysaccharide structures on the cell surface. In the case of oxytetracycline resistance, interactions between the modified lipopolysaccharide structures and specific transport proteins of the cell membranes must be assumed.

AbstractPhosphate limitation induces the turimycin biosynthesis as well as the cAMP phosphodiesterase and phosphatase. The results are discussed in connection with the observation of general high activities of dephosphorylating enzymes and low concentrations of phosphorylated intermediates under conditions of phosphate limitation and secondary product biosynthesis, respectively.
