AbstractGrowth of the blue‐green algaAnacystis nidulanstreated with methyl methane sulphonate (MMS) at pH 5 or 6 was better than that at pH 7 or 8, both in regard to the duration of the lag‐phase and the growth rate. Toxicity of MMS was directly proportional to the temperature at which the cells had been treated. Growth of cells incubated at 5000 lux during MMS treatment was better than those that had been treated in the dark or at 15000 lux.Ten successive treatments with MMS were ineffective in significantly shortening the lagphase of treated cells.The frequency of ultraviolet‐resistant mutants but not of streptomycin‐resistant mutants was substantially increased following single treatment with MMS of 40‐60 minute duration; treatments of longer duration resulted in a steep fall in the number of mutants suggesting thereby that treatments of up to 60 min induce genetic changes which are largely unaccompanied by physiologic

AbstractIfTorulopsis utilisis grown in mineral solutions containing 2.5 and 5% sugar the yields of yeast dry matter are only 80% and 26%, respectively, relative to the theoretical value if an economical coefficient of 50% is used. In the presence of 30% of waste liquor of the citric acid fermentation (CAF‐waste liquor) the yields attain 70‐85%. The effects of corn‐steep, group B vitamins or of biotin are much lower. Theoretical and higher yields of biomass are obtained by the combination of the CAF‐waste liquor with corn‐steep. The stimulation of yeast growth by wet or dry mycelium of the mouldAspergillus nigerwas similar to that of the CAF‐waste liquor, both when acting alone and in combination with other carriers of vitamins or growth factors. A good effect was observed with a mixture of the CAF‐waste liquor and the mould mycelium, both products of the citric acid fermentation. Mineral salt solutions containing 7% sugar may be fermented with theoretical yields by adding larger quantities of CAF‐waste liquor and mould mycelium. AbstractUnusual structures ‐ crystals, crystal‐associated microtubules, and bundles of microtubules ‐ have been observed in protoplasts, but not in normally grown cells ofSaccharomyces carlsbergensisandS. cerevisiae.The leaflet‐like crystals (diameter 0.3‐3 μm, thickness 0.01‐0.1 μm) occurred in the cyto‐ and karyoplasm. Crystals perpendicularly sectioned to the plane frequently showed highly regular arrays of electron‐opaque lines (periodicity 15 nm). Lamellar, rectangular, and hexagonal patterns could be seen on crystals sectioned parallelly to the plane.Smaller microtubules (outer diameter 15 nm) packed to compact bundles or fixed on crystals occurred only in the karyoplasm and they did not show any direct relations to the thicker microtubulus (outer diameter 20 nm) of the spindle apparatus.It is suggested that the crystals could be aggregations of tubulin

AbstractUsing 4 different methods, a total of 126Bacillusstrains representing 33 different species, was examined for their ability to degrade methoxylated and demethylated pectin. The highest number of pectinolytic strains (35) was found with a newly developed, tubed polypectate‐gel, which gave complete liquefaction upon pectinolysis. The pectinolytic activity appeared to be restricted to 8 recognized species belonging to 5 groups: TheB. maceranspolymyxagroup, theB. licheniformis‐pumilius‐subtilisgroup,B. pantothenticus, B. alvei, andB. lentus.However, not all strains of a single species revealed a positive pectinolytic activity. Thus, only a part of the strains of the second and fifth group were found to be pectinolytic. Pectinolysis withB. alvei(ILM 21, JO 42) andB. lentus(CCM 35, CCM 2214) was recorded not until a prolonged incubation in the tubed pectate‐gel was used.Pectinolysis of selectedB. licheniformisandB. pantothenticusstrains was stimulated markedly by ammonium‐ and phosphat‐ions at various pH levels. The simultaneous addition of calcium and phosphate to the medium decreased the intensity of pectinolysis clearly. AbstractIn replacementexperiments L‐tryptophan was rapidly metabolized byCandida guilliermondii, strain H17, cultivated before in a medium containingn‐alkanes as sole carbon sources. After growth in a medium containing glucose tryptophan utilization was suppressed. Thin layer chromatography showed tryptophol as main metabolite. Kynurenine, anthranilic acid, and other substances of the aromatic pathway of tryptophan metabolism could not be detected. Feeding experiments showed a significant degradation of anthranilic acid, tryptamine, indolylpyruvic acid, and indolylacetic acid. As a metabolite of tryptamine utilization tryptophol has been identified. Degradation of formyl kynurenine and formyl anthranilic acid was within limits of error. Transamination and decarboxylation are discussed to be the primary reactions in the degradation of L‐tryptophan under the conditions used in this
