AbstractPhospholipids extracted from a microorganism — the basidiomycetePolystictus versicolor— form liquid‐crystalline structures in water. After fixation by osmium tetroxide at known water contents and temperatures between +4 and +38 °C, ultrathin sections of embedded material were examined electron microscopically. As a result, part of the phase diagram was established and corresponding phases were characterized by their structural parameters at different conditions. Phases show micellar, lamellar, and hexagonal structures, respectively. Parameters of the lamellar phase were compared with the width of different types of membranes present in the intact organism. The results are not contradictorily to a far‐reaching bimolecular arrangement of lipids in membranes (unit membrane concept) and furthermore support the role of lipid association behaviour for cross‐sectional membran

AbstractIn cells of the stable and unstable spheroplast type L‐form ofProteus mirabilisD 52 small ellipsoidal particles (SE‐particles) could be found after investigation of ultrathin sections. The SE‐particles have diameters of 220–260 and 280–320 Å. They are composed of an electron lucent centre, an internal electron deanse layer, an electron lucent line and an outer electron dense layer. There is a porus of about 25 Å in the internal electron dense layer. The SE‐particles may be arranged in chains up to 10 particles. It seems that the particle formation starts near the cytoplasmic membrane at one or more sites in a cell.Comparing electron microscopic observations with data from literature it seems certain that SE‐particles are bacteriophage‐like structures. AbstractTechniques are described which allow the direct observation of water bacteria in their unchanged environment. Bacteria attaching to and growing on glass slides were observed with a partially submerged light microscope. Similarly, undisturbed living bacteria were studied in previously submerged PERFIL'EV glass capillaries, a necessity in the case of the fragileGallionellaorMetallogenium spp.Many new bacterial forms and surface structures were discovered when formvar‐coated electron microscope grids were used as the substrate for attached growth („Aufwuchs”︁).Several water bacteria can be obtained by covering slides with layers of agar media and coating these with thin, transparent plastic foil to encourage specific attachment. The foil was used like a cover slip on water agar‐coated slides. After microscopic observation the foil was peeled off and the colonies observed previously could then be isolated. Growth of pure cultures was successfully attempted in disposable Nalgene membrane filter units or in membrane‐covered chambers which had been submerged in the aquatic environment to facilitate nutrient and waste exchange with the outside. Natural bacterial populations could be transferred with only little disturbance from stratified sediments to the laboratory. This was achieved by keeping aquaria in the water for an extended period of time to allow for re‐establishment of the normal layering. During transportation, the aquaria were covered with plastic plates. In the laboratory some of the environmental factors such as the proper temperature and light were restored. With this method layered populations of even rare water bacteria could be kept alive for several months.Finally, an agar diffusion gradient plate is described. AbstractWährend des Wachstums auf verschiedenen Stickstoffquellen (Harnstoff und NH4Cl) ist der Ureasegehalt vonRhodotorula mucilaginosagering, die spezifische Aktivität beträgt etwa 0,2 μmol NH3/min/mg Protein. Nach Verbrauch der Stickstoffquellen steigt der Gehalt auf etwa den sechsfachen Wert an. Da das geringe Grundniveau während des Wachstums von der Stickstoffquelle und ‐konzentration unabhängig ist, wird es auf eine konstitutive Enzymsynthese zurückgeführt. Der starke Anstieg nach Verbrauch der Stickstoffquelle wird durch eine Derepression der Ureasesynthese verursacht. Das wurde durch den Zusatz von NH4Cl und Harnstoff zu ureasereichen Zellen bewiesen, der zu einer Repression der Ureasesynthese führt. Bei Applikation von NH4Cl geht während des Wachstums der Ureasegehalt auf das Grundniveau zurück, bei Harnstoff tritt ein wiederholter Abfall und Anstieg der spezifischen Aktivität auf. Diese Oszillation wird durch das zeitweilige Auftreten von Ammoniak aus der Harnstoffspaltung bedingt, das eine alternierende Repression und Derepression auslöst. Ammoniak fungiert dabei als hypothetischer Co‐Repressor. Oszillationen treten auch bei Abwesenheit einer exogenen Stickstoffquelle auf, sie werden wahrscheinlich durch endogene Schwankungen des Stickstoffmetabolismus bedingt.Der Anstieg der spezifischen Aktivität ist auf eine Neusynthese von Urease zurückzuführen, wie die Hemmbarkeit durch

AbstractIn der Zeit seit dem Erscheinen des ersten Teiles dieses Sammelreferates (KRAEPELIN 1967) hat der Atmungsdefekt, soweit er sich auf die sogenannte plasmatische „petite‐Mutation”︁ bezieht, noch keine definitive Klärung gefunden. Obgleich es zu diesem Thema bereits mehrere zusammenfassende Darstellungen gibt, in denen die zur Zeit wesentlich erscheinenden Fakten diskutiert wurden, mag die in manchem abweichende Betrachtungsweise dieses Referat dennoch rechtfertigen. Insbesondere mag es dazu beitragen, bekannte aber scheinbar unkorrelierte Phänomene einmal im Zusammenhang zu sehen.
