AbstractAn increase in the amount of membrane‐bound DNA was found inB. subtiliscells with UV‐induced DNA repair synthesis as compared to untreated cells. It was shown that DNA repair synthesis occurred in DNA membrane complexes (DMC) formed during UV‐irradiation. UV‐induced formation of DMC was observed in cells of wild type strains which were capable of repairing damaged DNA but not in a mutant defective in DNA‐polymerase I. It was demonstrated that DNA‐polymerase I is located on the membrane ofB. subtiliscells. This suggested a participation of DNA‐polymerase I in binding of the chromosome to the membrane in UV‐irradiated cells. UV‐induced DMC did not dissociate when the cells were treated with inhibitors of DNA‐gyrase. It, therefore, was qualitatively different from the DMC found during replication. AbstractOxalic acid (as oxalate) was detected in four tubers commonly used for food in Nigeria –Dioscorea rotundata(White yam),Solanum tuberosum(Irish potato),Ipomoea batatas(Sweet potato), andManihot esculenta(cassava). Whereas healthyI. batatahad the highest oxalic acid content, healthyM. esculentacontained the lowest. When all tubers were artificially inoculated with four fungi —Penicillium oxalicumCURIE and THOM,Aspergillus nigerVAN TIEGH,A. flavusandA. tamariiKITA, there was an increase in oxalate content/g of tuber tissue. The greatest amount of oxalate was produced byP. oxalicuminD. rotundatatuber. Consistently higher amounts of oxalate were produced by the four fungi in infected sweet potato tuber than in any other tuber and consistently lower amounts of oxalate were produced by the four fungi in Irish potato tuber. AbstractProtoplast type L‐form cells ofStreptomyces hygroscopicusandS. griseuscontain different types of inclusion bodies. Cytoplasmic cores and paracrystalline structures are peculiar inclusions which could not be observed in normal parent bacteria. The cytoplasmic cores are 1–4 μm long and 0.05–0.25 μm broad straight and stiff non‐tubular structures consisting of homogeneous moderate electron opaque material. Paracrystalline inclusions have side‐lengths between 0.2 and 0.5 μm and show a characteristic pattern of 15–20 nm thick straight dark lines and electron lucent intervening spaces of 20–30 nm. The influence of rape‐oil as antifoam agent on growth and protease synthesis ofT. vulgariswas studied. The turbulence and the aeration conditions caused by the culture vessel have a fundamental importance for the so‐called oil effect. A higher aeration rate induces an enhanced lysis of the mycelium and consequently a reduced enzyme synthesis. The tentency to lysis of the mycelium at different selectants is variable. AbstractAcinetobacter calcoaceticus69‐V is capable of utilizingn‐alkanes as the sole carbon source. By conventional and high voltage electron microscopy the fine‐structure of these bacteria grown on hexadecane (with or without addition of DL‐carnitine), succinate or yeast extract was analysed.A. calcoaceticusgrown on hexadecane exhibited intracytoplasmic unit membrane aggregates in contrast to controls grown on the nonhydrocarbon substrates. Probably these aggregates are formed by the cytoplasmic membrane. Most of these membranes are surrounding and penetrating hexadecane inclusions. Ultrastructurally no essential difference caused by addition of carnitine could be seen. It is generally assumed that the first step of alkane degradation in several yeast strains and bacteria is catalyzed by a monooxygenase system containing cytochrome P‐450 as the terminal oxidase and a NADPH‐cytochrome P‐450 reductase as the electron transfer component. To prove this hypothesis both proteins were purified to homogeneity from the microsomal fraction of the alkane‐assimilating yeastL. elongisporusand reconstitutedin vitro.The cytochrome P‐450, having a specific content of 12–17 nmoles/mg protein turned out to be a typical member of this class of hemoproteins with respect to the molecular weight (53 500) and spectral properties.The reductase component with a molecular weight of 79000 contains FAD and FMN as prosthetic groups.The recombination of the hemoprotein and the flavoprotein results in an enzyme system which catalyzes the monoterminal hydroxylation of hexadecane.The activity of the reconstituted enzyme system is remarkably stimulated by nonionic detergents, suggesting the requirement of an additional lipid factor. Screening for methanotrophic bacteria led to the isolation of aMethylosinus trichosporiumstrain which was characterized by the ability to produce a bright red‐violet pigment, which was found to be nearly insoluble in water. This communication reports on the formation, isolation, and purification of the pigment and its separation into the components methylosin A and B.By comparison of physico‐chemical parameters we identified the pigment as a prodiginine‐type compound.
