Whole-body and myocardial Vo2were estimated in dogs during anesthesia with halothane at2to define the relative contributions of individual organs to the overall decrease in whole-body Vo2Wholebody Vo2decreased 27 per cent as halothane was increased from2resulting from decreased cardiac output and arterial pressure. Contributions of individual organs to the total decrease in whole-body Vo2expressed as percentages, were: myocardial, 47; skeletal muscle, 23; splanchnic, 9; renal, 5; cerebral, 2. Thus, 86 per cent of the total decrease in whole-body Vo2was accounted for, the remainder presumably occurring in tissues not studied (skin, bone, salivary glands, spinal cord, lung, and others).

Eight isometrically contracting rat left ventricular trabeculae carneae muscle preparations were exposed to concentrations of 25, 50, and 75 vol per cent of both N2O and N2Both gases depressed peak developed isometric tension and the maximum rates of tension development and relaxation in relation to the gas concentrations. In five other muscles, paired electrical stimulation reversed the negative inotropic effects of N2O and N2The relative potentiation of contractility achieved with paired stimulation was the same in the presence of O2N2O, and N2Since N2O did not produce any additional depressant effects over and above those observed with N2, and since the positive inotropic effects of paired stimulation were unchanged by this anesthetic, it was concluded that N2O does not possess any direct myocardial depressant or stimulant properties.

To investigate functional impairment subsequent to arterial hypocapnia, six healthy young men served as their own controls for two thiopental-N2O-d-tubocurarine anesthesias lasting mare than 125 minutes, one with and one without hypocapnia (Paco218 torr). Subjects were trained to stability on five recognized tests of cognitive function (simple and disjunctive visual reaction times, visual span of apprehension, forward letter span, and dichotic listening). Performance was evaluated daily for four days before anesthesia, 90 minutes after anesthesia, and daily thereafter until performance had reached the baseline values. No significant changes in performance were observed following either hypocapnic or normocapnic anesthesia. In part, this may have been related to the highly motivated and proficient population studied; therefore, these conclusions may not apply to hospital patients.

The uptake, distribution, and excretion of FX-80, a water-insoluble mixture of isomers of perfluorobutyl perfluorotetrahydrofuran, were studied in 17 dogs subjected to liquid breathing with oxygenated FX-80. The absence of excess fluoride ion in urine suggests that FX-80 is not metabolized, nor is it excreted in urine unchanged. Its distribution in the body depends on the lipid content of tissues. The uptake and desaturation curves are multiple exponential functions, the rate constants of which can be predicted from the ratios of lipid contents of blood and tissues and the fractions of cardiac output supplying the tissue compartments. During liquid breathing the concentration of FX-80 in arterial blood averages 0.43 mg/100 ml. Calculations indicate that at saturation a 13.8-kg dog would absorb 1.25 g of FX-80. Of this amount, 71 per cent would be deposited in adipose tissues and bone marrow, 25 per cent in muscles and skin, and 4 per cent in parenchymatous tissues. Desaturation is delayed because deposits of liquid FX-80 are sequestered in the lungs following liquid breathing.

Functional residual capacity (FRC) and dynamic lung compliance [C4ya(2)] were measured in ten normal sitting subjects while conscious and again during thiopental-meperidine anesthesia with succinylcholine paralysis and mechanical ventilation. At similar tidal volumes and respiratory frequencies, no significant changes in FRC and Cdya(1)were observed. Mean FRC's were 3.26 ± 0.26 (SE) liters and 3.29 ± 0.24 (SE) liters and mean values for C(1)were 182 ml/cm IL-O (SE ± 25) and 153 ml/cm H-O (SE ± 34) in conscious and anesthetized paralyzed subjects, respectively. The results suggest that the reductions in FRC and C(1)previously observed in supine anesthetized subjects are not the result of a direct pharmacologie effect of thiopental-me-peridine anesthesia on the lung.

Intracranial pressure (ICP) and cardiovascular changes were studied intraoperatively in eight neurosurgical patients undergoing craniotomy. Four patients with preoperative signs of increased ICP had acute elevations in ICP (34–83 torr). These occurred during intubation with thiopental and succinylcholine, initial exposure to halothane, application of a head clamp, and intracranial insertion of a saw guide. Some abrupt increases in ICP occurred without significant alterations in systemic arterial and central venous pressures or evidence of hypoxia or hypercarbia. In response to similar stimuli, patients presumed to have normal ICP's preoperatively had no marked changes in ICP. During administration of halothane, cerebral perfusion pressures associated with acute episodes of intracranial hypertension were less than or equal to 60 torr in the preoperatively-high-ICP group, while they remained above 80 torr in normal-ICP patients.

Information about the effects of neuroleptanalgesia (NLA) on the electrical function of the auditory cortex was obtained by recording gross electrical and unit activity in the primary auditory cortex of the cat after administration of droperidol and fentanyl (Thalamonal). In the EEG no change could be detected visually after an intramuscular injection of Thalamonal, nor did spectral analyses reveal any significant alteration. Pentobarbital, on the other hand, caused a highly significant slowing of the EEG. Single-cell activity was clearly depressed by pentobarbital, but during NLA most of the functional neurons of the auditory cortex were firing spontaneously and responded readily to various types of sound stimuli. From these findings the authors conclude that NLA at the level of surgical anesthesia does not significantly affect the electrical function of the auditory cortex and the auditory pathway in the cat.
