摘要:
Platelet aggregation induced by ADP in canine platelet-rich plasma was inhibited by volatile anesthetics. The partial pressures at which platelet aggregation was inhibited 50 per cent correlated closely with the pressures used clinically: methoxyflurane, 3.5 torr; halothane, 7.5 torr; diethyl ether, 36 torr; cyclopropane, 120 torr. Nitrous oxide at 640 torr inhibited platelet aggregation 34 per cent.

摘要:
Mammalian hepatoma liver cells were grown as a suspension tissue culture to allow investigation of the effects of nonvolatile anesthetics on cell replication. Logarithmic growth of a rat hepatoma tissue culture was reversibly inhibited by thiopental, methohexital, amobarbital, phenobarbital, lidocaine, and prilocaine in a dose-dependent fashion. Barbituric acid also reversibly inhibited logarithmic growth, but only at drug concentrations greater than those necessary for inhibition by the anesthetic drugs.

The circulatory response to hypercapnia was studied in nine volunteers. The inhalation of carbon dioxide by conscious subjects led to increases in cardiac output, heart rate, mean arterial pressure, and myocardial function (measured indirectly by ballistocardiography). Central venous pressure and total peripheral resistance decreased. During anesthesia at both 5 and 9 per cent alveolar fluroxene concentrations, responses of cardiac output to CO2were attenuated by 40 per cent, primarily because of depression of the heart rate and the myocardial contractile response to hypercapnia. The effects of CO2on other cardiovascular variables were similarly attenuated. The data suggest that fluroxene attenuates the sympathetic response to hypercapnia. The depression of the response of cardiac output to hypercapnia was less with fluroxene than with halothane or deep cyclopropane anesthesia.

Increasing partial pressure of oxygen by hyperbaric oxygenation improves the efficiency of membrane oxygenators sufficient to provide almost total respiratory support with membranes of practical sizes. The oxygenating capacities of the Teflon-Peirce, Landé-Edwards and G.E.-Peirce membrane oxygenators were tested using surface areas of 1 to 3 m2, flow rates of 500 to 2,000 ml/ min, and ambient pressures from 1 to 3 ATA (atmospheres absolute) in the hyperbaric chamber. For every 3 m2-membrane tested at 2 ATA or more the amount of oxygen in the blood leaving the oxygenator approached its maximum oxygencarrying capacity. The Teflon-Peirce membrane decreased in efficiency with increasing flow rates and is unsuitable for clinical use. The Landé-Edwards (3 m2) and the G.E.-Peirce (2 m2) at flow rates of 2,000 ml at 2 ATA were capable of transferring oxygen at the rates of 200 and 240 ml/min, respectively. It was not necessary to pressurize above 2 ATA to achieve efficient use of the membrane oxygenators tested.

The kinetics of radiolabeled Ieucine, thymidine, uridine, and acetate uptake in the presence of 25 to 100 mg/100 ml halothane were studied with spinner cultures of HTC cells (Morris rat hepatoma 7288C) maintained in continuous logarithmic growth. At a time when exposure to halothane produces only slight reductions in DNA and protein synthesis and has no effect on RNA synthesis, a marked increase in lipid synthesis was detected. No preferential accumulation of activity was found in any of the neutral and phospholipid fractions examined, and the increases in the acid-soluble and acid-insoluble fractions were proportionate to the increase in total uptake of labeled acetate. Hence, it appears that the primary effect of halothane is the acceleration of transfer of acetate into hepatoma cells. Trifluoroacetate and trifluoroethanol, two proposed metabolites of halothane, had no effect on incorporation of the above-mentioned macromolecular precursors.
