The fatty acid composition of the total lipids from twoConidiobolusspecies was studied by gas—liquid chromatography. The major fatty acids ofC. lampraugeswere palmitic acid (C16:0), oleic acid (C18:1), linolenic acid (C18:3), and arachidonic acid (C20:4). ForC. eurymitus, myristic acid (C14:0), C16:0, and linoleic acid (C18:2) were the most abundant acids. The fatty acid composition ofC. eurymituswas quite different from that of theConidiobolusspecies as mentioned in other reports. The lipid composition of the total lipids ofC. lampraugesandC. eurymituswas also studied by thinchrography on quartz rods. The resistance of spores ofBacillus subtilisNCTC 10073 to glutaraldehyde, sodium hypochlorite and povidone‐iodine was compared. Revival of treated spores was examined by use of defined germination media and conditions, protein denaturing agents, ultrasonics and heat. A microcomputer‐based system for the storage and retrieval of information on strains in a culture collection is described. The system was designed around commercially available software packages written for microcomputers. Two additional programs were written using the BASIC language to allow a catalogue of the culture collection to be printed in a specific format. The details of each strain in the collection were stored on a floppy disc. Details of new strains were added to this database and information relating to existing cultures was modified or, where necessary, deleted from the collection. The database can be searched to supply details of a particular culture or to identify those cultures which possess certain attributes. The records for the whole collection were sorted alphabetically by species name, and numerically by accession number, and a word‐processing package was used to print a catalogue of the culture collect

An effective selective medium for the enumeration ofAspergillus flavusandAspergillus parasiticushas been developed by modification of Bothast and Fennell'sAspergillusDifferential Medium. Results can be obtained with the new medium,Aspergillus flavusandparasiticusAgar (AFPA), after 42 h incubation at 30°C. The medium is thus suitable for use in quality control as a guide to the presence ofA. flavusand, potentially, of aflatoxins. AFPA has been extensively tested on peanuts and soils. Results were reproducible and comparable with those on standard fungal enumeration media incubated for much longer periods. A very low percentage of false positives or negatives was found

The capacity of six basal media to support the growth of thermophilic campylobacters was tested. The most successful was Nutrient Broth No. 2 (Oxoid) solidified with New Zealand agar but it gave at best only a 9% recovery rate. Various blood products, iron compounds, detoxifying agents, reducing agents, growth stimulants and an antimetabolite were added to the selected basal medium and counts of inoculated organisms were compared with counts on basal medium containing 5% lysed horse blood. Of 22 supplements tried only blood, Fildes' peptic digest of blood, haematin, iron salts, charcoal, sodium metabisulphite and sodium pyruvate greatly improved the basal medium. The ability of these supplements used singly and in combinations to facilitate aertolerance of campylobacters was investigated. Two aspects of aerotolerance were tested; (a) the ability of the supplements to sustain the viability of campylobacters seeded onto culture plates left on the bench for up to 6 h before microaerobic incubation; and (b) the ability of the supplements to facilitate the growth of campylobacters at increasing oxygen tension (6, 10 and 17% oxygen). A combination of 0.4% charcoal, 0.025% ferrous sulphate and 0.025% sodium pyruvate was found to be as effective as blood in both tests.

Contrary to expectations slight undercooking (68.5°C instead of 70°C for 90 min) dramatically increased the shelf‐life of chub‐packed luncheon meat stored at 25°C. The pH of undercooked chubs fell rapidly to below 5.0 as a result of the growth of enterococci. The accumulated acid prevented the growth ofBacillusspores and gave the luncheon meat a not unpleasant tangy flavour. Degradative changes associated with the spoilage of commercially cooked chub‐packed luncheon meat did not occur, even after 42 d storage. The susceptibilities to six antibiotics have been determined ofClostridium perfringensserotypes isolated from beef, pork and lamb carcases and from minced beef. Generally similar results were obtained to those of previous studies of mainly clinical isolates. Fourteen of the 229 strains tested were resistant to tetracycline; three of these, from the same beef carcase, were also resistant to erythromycin.
