摘要:
We describe a method in which microwave irradiation is used to reduce substantially the incubation time for immunoperoxidase staining of antigens in cryostat sections of pso-riatic skin. An incubation time of 5–9 min irradiation at 80 W generated similar or better staining intensity for all antibodies used compared to the standard methods using 30–60 min incubation at room temperature. Although we found that microwave irradiation could be used with all antibodies tested, independent of whether they recognized extracellular, membrane or cytoplasmic antigens in skin, the conditions needed to be optimized for each antibody.

Neutral buffered formalin (NBF) has been the standard fixative in histopathology for many decades: however, new technologies and increasing time constraints have made this common fixative less widely applicable. The attributes of universal and ideal fixatives are reviewed and compared with those of NBF. On the strength of a growing body of literature and experience in the histopathology field, zinc formalin solutions are recommended as a functional replacement for NBF. Zinc formalin permits better morphological preservation and preserves immunoreactivity even after prolonged fixation times. Coordinated zinc ions are postulated to maintain macromolecules in their original three-dimensional conformation as formaldehyde forms addition products with them. Significant crosslinking and gross distortion of tertiary structure are thereby avoided.

Using divergent Giemsa staining protocols designed for other plant species, G-banding was visualized in the chromosomes ofAllium cepaL.,A. fistulosumL., and their interspecific (A.fistulosum×A.cepa) hybrid. This is the first demonstration of G-banding ofAlliumchromosomes. Differences and similarities with other Giemsa banding patterns in Allium are discussed.

Leaf tissue of tall fescueFestuca arundinaceaSchreb., hard fescueFestuca ovinaL., red fescueFestuca rubraL. and perennial ryegrassLolium perenneL. was stained with rose Bengal or aniline blue to detect the presence of the fungal endophyteAcremonium sp., Specimens were cleared using methyl salicylate, an optical clearing agent, and viewed using bright field microscopy. Tissue was presenred as dried tissue or stored in 70% aqueous ethyl alcohol before staining and clearing. Tissue was observed at 2, 4 and 12 weeks following clearing to check for stain retention. Staining with rose Bengal was inferior to aniline blue when followed by the clearing agent methyl salicylate. Fungal mycelia stained lighter with rose Bengal and were more difficult to detect than mycelia stained with aniline blue. The results illustrate the usefulness of combining staining and methyl salicylate clearing for detecting fungal endophytes.

Rhodamine-phalloidin was used to label F-actin in unfixed cells of 13 species of filamentous and blade-forming red algae from the three families Ceramiaceae, Acrochaetiaceae and Bangiaceae. Labelling was achieved only after treatment with eitherβ-glucuronidase or a combination of cellulase and an extract of snail gut enzyme. Different species required different enzyme treatments and incubation times for successful labelling. All species examined showed extensive arrays of F-actin which generally are confined to the peripheral cytoplasm and are oriented longitudinally. Transverse arrays are present beside the crosswalls ofGriffithsia pacifica. andAudouinellaspecies show actin concentrations at the tips of apical cells and in developing branch initials.

摘要:
A method is described for selective fluorescent staining of polysaccharide bodies, such as those found in Alzheimer's disease, Lafora's disease and adult polyglucosan disease. Formalin fixed, paraffin embedded sections of human autopsied brain tissue were stained with the fluorescent compound dansyl hydrazine. It specifically stained polysaccharide bodies in tissue sections with great sensitivity and little background. The dansyl hydrazine staining technique also reveals a high degree of structural detail in the stained tissues.

A procedure for stabilizing fluorescent markers used to study the microvascular geometry and morphometry of muscle tissue is described. The procedure involves fluorescent labeling of plasma. fixation of muscle tissue in 10% buffered formalin. and quick freezing. This procedure prevents extravasation of the fluorescent dyes out of the capillaries as frequently seen in other muscle microvascular techniques, thereby greatly increasing the time that capillaries are visible. We found that formalin may actually increase the rate of fluorochrome bleaching by photo-oxidation, but the increased rate of bleaching is more than offset by the greater concentration of dye trapped in the capillaries. Further, formalin fixation results in little distortion of the muscle fibers themselves, making this approach ideal for morphometric studies.

摘要:
A new method is presented for staining various types of senile plaques isolated from the brains of patients with Alzheimer type dementia and related diseases in paraffin embedded sections using a modified Bodian's method with methenamine. This methenamine-Bodian method made it possible to observe diffuse plaques and other amyloid deposits which are barely detected by Bodian's original method. The staining of senile plaques by the method presented here was comparable to that of immunostaining with anti-β-protein. The new method also stained neurofibrillary tangles. Therefore, the methenamine-Bodian method could be widely used for the detection of senile changes in paraffin embedded sections from autopsied human brains.
