SUMMARYThe construction and use of a thin culture chamber for time-lapse photomicrography of fungi at high magnifications are described. A photomicrographic sequence of blastospore development illustrates the potential of the chamber.

SUMMARYThe current postulate that organic acids added to a culture medium affect growth of fungi only by buffering the medium was tested by culturingClaviceps purpureaPRL 1980 in a medium containing glucose, ammonia, salts and 2, 3-14C-succinate and analyzing various fractions of the mycelium for the presence of radioactivity. In addition, radioactivity of respired CO2was measured. It was found that succinate was taken up before growth was evident and was assimilated and respired both before and after exogenous glucose was consumed. Radioactivity appeared in the hot water extract, ether-ethanol extract, hot TCA extract, protein extract, and in the crude cell wall preparation as well as in two sugars isolated from the cell wall. Thus, exogenous succinate participated in metabolism and did not influence the culture only in its capacity as a buffer.

SUMMARYSamples were taken from the top 2 inches, including the surface, of virgin soils from 24 localities in the Sonoran Desert. There were 107 genera and 229 species isolated in addition to some unidentifiedPhomaspp., mycelia with clamp connections, and large numbers of other sterile mycelia. No thermophiles were isolated.There does not seem to be a fungus flora that is characteristic of desert soils. The melanic Fungi Imperfecti occurred in large numbers, though not necessarily in greater numbers of species than non-melanic forms. Many of the isolated species are pan-world. The soil contained many species more commonly associated with dung, others known to be plant or animal pathogens and others as laboratory contaminants. Many of the species may not be true soil fungi and their presence in the soil simply fortuitous.

SUMMARYIntermediate-chain-length fatty acids (C8-C14) inhibit growth and respiration of fungi under conditions where the longer- and shorter-chain acids are noninhibitory. Under the conditions tested, lauric and capric acids (0.02 M,pH 5.5) inhibited endogenous O2uptake byCunninghamella echinulata.The inhibition was not permanent and the organism recovered when given sufficient time in contact with the substrate. Recovery from the inhibition was O2dependent, since cell preparations incubated under nitrogen failed to recover. Supplemental coenzyme-A stimulated the rate at which the organism recovered from the respiratory inhibition by capric and lauric acids. Furthermore, pantothenic acid reversed the inhibition by these acids, and hydroxylamine stimulated the rate of recovery from laurate inhibition. The rate of recovery was inversely related to the chain length of the acid at equimolar concentrations. The findings indicate that the mechanism by which these acids inhibit endogenous respiration is to be understood at the enzymic level. Moreover, the data presented show a definite involvement of coenzyme-A and its acyl derivatives in the mechanism by which the intermediate-chain-length acids inhibit endogenous respiration in this fungus.

SUMMARYLyophilized ampules of 447 strains of fungi, mostly Aspergilli and Penicillia, were checked for viability. Many of the ampules had been processed 23 years previously. All strains sporulated in cultures made from lyophil preparations as well as, or better than, in agar slant cultures that had been periodically transferred many times. It was necessary to open a second lyophil ampule for only 11 strains to obtain viable cultures.

SUMMARYStudy of the ultrastructure of dividing nuclei in the somatic phase of the life cycle of the myxomyceteClastoderma debaryanumhas revealed a mitotic apparatus that is acentric (without centrioles) and has a continuous nuclear envelope. Groups of spindle microtubules of the metaphase mitotic apparatus converged slightly and terminated on or near the inner membrane of the nuclear envelope to produce at the flattened“poles”microtubule configurations that were reminiscent of those seen when microtubules are associated with centrioles. It is suggested that the myxomycete mitotic apparatus may contain an evolutionary forerunner of the centriole of higher organisms.
