ABSTRACTReliable methods for oospore production byPhytophthora syringaewere developed using low temperatures and media amended with vegetable oil or made from the leaves of stone fruit trees. All of the fourteen isolates tested individually produced oospores in media after 4 weeks at 5 and 9 C but most did not produce oospores at temperatures above 12 C. Similarly, oospores were observed only in almond leaves kept at 12 C and below. Oospore production was greatly increased when corn oil, linseed oil, or wheat germ oil was added to various base media and the number of oospores produced increased with the amount of wheat germ oil added (up to 48 ml/L medium). The addition ofβ-sitosterol resulted in a slight but significant increase in oospores. Abundant oospores were produced by an almond isolate ofP. syringaein almond leaf medium (75 g fresh leaves per liter medium). Oospores were also produced in media made from the leaves of other stone fruit trees, but not in apple leaf medium.

ABSTRACTTwo new species of xerophilic fungi are described:Geomyces pulvereusHocking&Pitt, andMonascus eremophilusHocking&Pitt. TheMonascusspecies is an obligate xerophile, which does not appear to have an anamorphic state. A further occurrence ofEurotium halophilicumis reported, and a full description of this rarely encountered species is given.

ABSTRACTThe influence of the type of carbohydrate and its concentration and of light on fruitbody development inClavicorona pyxidatawas studied. Fruiting was stimulated by transfer of inocula grown on cellophane squares on a nutrient-rich agar medium to a nutrient-limited agar medium. Fruiting began after 6–60 days, depending on the source and concentration of carbohydrate. Fructose and maltose at 20 and 40 g/L produced the highest fruitbody yields, and fruiting was initiated quickly. At these concentrations, glucose produced spindly fruitbodies in significantly lower yields, and fruiting was delayed. A comparative study was made of the effects of glucose and maltose on timing of fruiting and fruitbody yield and morphology over a wide range of concentrations. Fruiting was also investigated on the following carbon sources which produced abnormal fruiting and lower yields: mannitol, sorbitol, cellobiose, cellulose, sucrose, and arabinose. Light was required for fruiting.

ABSTRACTBasidiospore formation inLactarias lignyotellusis a complex phenomenon involving cytoplasmic and wall layer synthesis and interaction. Four stages in early basidiospore development prior to nuclear migration into the spore cytoplasm were delineated, as evidenced by changes in gross morphology, wall tegumentation, and cytoplasmic differentiation. The establishment of early spore asymmetry in heterotropic basidiospores ofL. lignyotelluswas correlated with the presence of a hilar appendix body. The hilar appendix body was poorly differentiated, but appeared to be a demarcated region of cytoplasm that influenced localized spore wall synthesis. The amyloid portion of the spore wall was initiated as an inner, electron-transparent zone. Specialization of the hilar appendix region for forcible discharge was not evident in early spore development inL. lignyotellus.Organelle composition of the cytoplasm throughout early basidiospore development inL. lignyotelluswas comparable to that reported previously for other fungi.
