ABSTRACTThe morphology of both conidia and setae ofC. fragariae, C. gloeosporioides, andC. acutatumisolates from strawberry were compared and found to be distinct for each species. Strawberry leaf agar was an excellent medium for the production of morphologically uniform conidia typical of a species. Conidial shape was more useful than size in species determination.C. fragariaeis not synonymous withC. gloeosporioidesas proposed by von Arx. Isolates ofC. gloeosporioidesthat did not form the teleomorph were reliably identified by conidial and setal morphology alone. Radial growth ofC. acutatumfrom 21–30 C was significantly slower than that ofC. fragariaeandC. gloeosporioides.At 33 C only isolates ofC. gloeosporioidesthat did not produce the teleomorph grew well. Isolates ofC. acutatumthat formed a red pigment in culture are discussed.

ABSTRACTDispersal via basidiospores has predictable consequences for fungal population structure. Spatial distribution of 60 heterokaryons in a natural population ofPleurotus ostreatuswas examined through study of their somatic incompatibility interactions and analysis of mating compatibility factors. In order to estimate the minimum genetic relationship detectable by somatic incompatibility testing, pairing studies were performed using sib-composed dikaryons and their parental dikaryons. Most pairings (93–100%) between sib-composed dikaryons ofP. ostreatusresulted in somatic incompatibility responses. Patterns of somatic incompatibility interactions among 60 naturally occurring isolates from a 6-hectare area in the Duke Forest indicate that most isolates represent discrete individuals, with as many as 15 individuals occupying a single log. A total of 53 somatically distinct individuals were identified from the population, distributed among 21 logs. Somatically compatible strains from different logs were not observed. The distribution of mating alleles was analyzed for a sample of 10 dikaryons by analysis of mating interactions between dedikaryotized progeny. The observed patterns of distribution for mating alleles supports the view that somatically compatible isolates in this instance were clones. Isolates arising from different logs were all shown to possess unique mating factors. Although four of seven isolates from a single log each carried unique mating factors, the presence of common mating factors in two remaining pairs of strains also suggests nonrandom mating factor distribution at the level of a single substrate. The results of this study implicate basidiospore dispersal as a primary mechanism for dikaryon establishment. However, further genetic studies are still necessary to determine the extent of different mating patterns between individuals in the population.

ABSTRACTPodochytrium dentatumoccurred during spring and fall on chitin bait placed in a Maine lake. Zoospore ultrastructure is of the chytridialean type and is a variant of theChytridiumsubtype. The zoosporangium forms from an apical expansion of the encysted zoospore. At maturity the dentate apical part of the zoosporangium, with orange contents, is separated from the basal part of the zoosporangium by a wall. Zoospores discharge in a mass surrounded by papillar material. Rhizoids branch from a knob-like rhizoidal base and are either fine or coarse.

ABSTRACTMating and infection structures ofUstilago maydisproduced in culture and on maize plants were examined with light and electron microscopy. Fusion tubes formed between low numbers of compatible haploid sporidia incubated in water for 8–16 hours. Both nuclei from paired sporidia migrated into a rapidly elongating hypha that did not differentiate further in the absence of host cells. Sporidial fusion and hyphal emergence also occurred rapidly and consistently in leaf whorls of inoculated maize seedlings, and hyphae had entered maize leaves after 12 hours. Only young leaves in the inner part of the leaf whorl were infected. Hyphae did not grow through stomatal openings, but, instead, entered epidermal cells by penetrating the cuticle and then either growing directly through the underlying cell wall or first growing between epidermal cells before penetrating an anticlinal wall of one of the cells. Invaded epidermal cells exhibited no necrosis or other dramatic response to entering hyphae.

ABSTRACTClamp connections were observed in basidiospore dilutions from each of four North American biological species ofArmillaria(A. gallica, A. sinapina, A. ostoyae, andA. calvescens) following spore germination and hyphal anastomosis. The occurrence of clamp connections in intrabasidioma pairings of single-germinated basidiospores and of excised margin-hyphae ofArmillaria ostoyaeandA. gallicawas that expected of a bifactorial incompatibility system. All intraspecific pairings between basidiospores or excised margin-hyphae from actively growing haploid cultures from two different basidiomata ofA. ostoyaeand ofA. gallicaproduced clamp connections. The development of the crustose colony morphology was almost fully coincident with the formation of clamp connections in compatible matings with eitherA. ostoyaeorA. gallica.Results indicate that clamp connections occur predictably and provide an additional precise and useful criterion for recognizing biological species ofArmillaria.For unexplained reasons, haploid isolates paired more than 3 mm apart failed to produce clamp connections.
