ABSTRACTThe similarity of 88 isolates of 27 species ofLeptographiumwas studied using enzyme electrophoresis. UGPMA cluster analysis of similarity matrices (Nei genetic identity, I) generated from data of 267 electrophoretic forms (electromorphs) of 15 enzymes showed a close correspondence between electrophoretic similarity and morphology. Isolates of a species had high similarity and clustered at I≥0.60, and in cases where different taxa clustered at I>0.60, an examination of the morphology and history of the isolates suggested conspecificity. Isozyme characterization was also useful for identifying the extent of morphologic variation within versus between species and for comparing isolates with degenerate morphology after prolonged culture.

ABSTRACTThe anamorph ofThuemenella cubisporais shown to be a species ofNodulisporium, andT. cubisporais referred to the Xylariales, Xylariaceae. The remaining species ofThuemenellaare reevaluated and considered congeneric withSarawakus lycogaloides. Sarawakusis recognized for members of the Hypocreales, Hypocreaceae, havingHypocrea-like stremata and eight, unicellular, warted or tuberculate, green, pale yellow or rarely colorless ascospores. The following are placed inSarawakus: Thuemenella brittanica, T. fragilis, T. hexaspora, T. izawae, T. sordida, Hypocrea subtrachycarpa, andH. trachycarpa.The new speciesSarawakus roseusis described and illustrated.

ABSTRACTThe distribution of the actin cytoskeleton was examined in hyphal tips of the plant pathogenic fungusSclerotium rolfsii.Actin was visualized with light microscopy using immunofluorescence techniques and with ultrastructural immunocytochemistry. Computer-aided, three-dimensional reconstruction analysis of ultrathin serial sections and immunoblot analysis techniques were also employed in this study. Immunofluorescence data indicated that the majority of actin was localized in brightly fluorescent plaques that were principally localized in the first 10 to 12μm of the hyphal tip. Focusing through the hyphal cell revealed that most plaques were positioned in the peripheral cytoplasmic regions. Actin microfilaments were not resolved at the light microscopic level; however, coarse actin fibers oriented parallel to the long hyphal axis were observed in subapical regions and in association with septa. A diffuse, amorphous cytoplasmic staining was present in the apical regions of the hypha except for that area occupied by the Spitzenkörper. Ultrastructural immunocytochemistry and three-dimensional reconstruction analysis suggested that the fibrillar coating of filasomes contained actin and that filasomes represented the ultrastructural equivalent of actin plaques. Immunoblot analysis of total protein extracts separated by one-dimensional sodium dodecyl sulfate Polyacrylamide gel electrophoresis identified two actin related proteins, one with a relative molecular mass of 43,000 and the other migrating with a slightly higher molecular mass of 47,500.

ABSTRACTSpore concentration, tube culture age, nutrients, high temperature, and pH were studied to determine their effect on germination ofRhizomucor mieheisporangiospores. Germination was evaluated by incubating the spores for 6 hours at 45 C on a defined glucose-basal agar (aqueous solutions of nutrients were used to evaluate their effect on spore germination). The six spore concentrations tested did not affect the percent spore germination. A sharp drop in the percent spore germination occurred when spores were obtained from cultures incubated at 40 C for more than 10 days. Spores did not germinate in distilled water nor in solutions of KNO3or vitamins. Spores did not germinate after being incubated in distilled water for 5 minutes at 65 C, for 2 hours at 60 C, or for 12 hours at 55 C, but not all of the spores were killed by these treatments. Percent germination was not affected when the pH of the buffered, liquid glucose-basal medium was between 4.5 and 7.

ABSTRACTWhen culture blocks were incubated on V-8 agar plates containing 300 ppm streptomycin sulfate, mutants appeared as fast-growing sectors within 30 days. Streptomycin-resistant mutants were isolated from the sectors of both A1and A2mating types ofP. parasitica, P. palmivora, P. capsiciandP. cinnamomi.Streptomycin also caused mating type change inP. parasitica.Streptomycin-dependent mutants were also isolated from the fast-growing sectors of A2isolates ofP. parasiticaand A1and A2isolates ofP. palmivora, P. capsiciandP. cinnamomi.Presence of CaCl2in the medium induced a greater need for streptomycin for the dependent isolates ofP. parasitica.Fast-growing sectors also appeared when culture blocks were incubated on medium containing 100 ppm chloramphenicol. Chloramphenicol-resistant mutants were isolated from sectors of A1and A2mating type ofP. parasitica, P. palmivora, P. capsiciandP. cinnamomi.By incubating streptomycin-resistant mutants ofP. parasiticaon medium containing chloramphenicol and vice versa, mutants resistant to both antibiotics were obtained.

ABSTRACTThe development of the heteroecious, macrocyclic rustPuccinia zoysiaeon zoysiagrasses (Zoysiaspp.) andPaederia scandenswas studied using bright-field and scanning electron microscopy. This fungus grows intercellularly through the mesophyll of the zoysiagrass leaf, invading parenchyma cells, and forming haustoria. However, its growth is restricted by the vascular bundles and the bulliform cells, which the fungus does not penetrate. As a consequence, longitudinal lesions are formed, predominantly on the adaxial leaf surface. Soon after the onset of infection, paraphyses and echinulate urediniospores are produced, and are rapidly exposed due to rupture of the epidermis. Teliospores form later, first within the uredinia, and subsequently in telia which appear on both leaf surfaces. These spores are two-celled, persistent, and pedicellate. They germinated after two months of suspension in water at 4 C, followed by exposure to a saturated atmosphere for 24 hours. Germination begins with the emergence of a basidium (promycelium) from either or both cells of the teliospore. This is followed by the formation of septa and sterigmata from which basidiospores arise. These spores infect leaves ofP. scandens, producing inconspicuous, intercellular spermogonia. Aecia form intercellularly and eventually erupt through the lower leaf surface. The aecial peridium has an unusual double-layered wall, the inner wall of which consists of non-viable aeciospores. Aeciospores are verrucose and are produced in basipetal succession.
