SUMMARYFive new species and one new variety ofCeratocystis, isolated from sapwood of forest trees, were grown described from culture medium.Ceratocystis francke-grosmanniaewas from larval galleries of the insectHylecoetus dermestoidesin oak from Germany.Ceratocystis olivaceapiniwas fromDendroctonussp.-infested ponderosa pines in Arizona and New Mexico.Ceratocystis distortawas isolated occasionally from spruce and fir infested with ambrosia beetles, widespread in North America,Ceratocystis gossypinais a very common white mycelioid species in insect galleries in pines in North America. One isolate from insect-infested pine in southern New Mexico is described as a new varietyCeratocystis gossypinavar.robustaIt is similar in growth and culture appearance but has larger perithecia.Ceratocystis sparsawas isolated once from white spruce in Alaska. It also has white mycelium and grows very slowly with perithecia developing slowly or not at all in some transfers.

SUMMARYA laboratory procedure to obtain mushrooms by casing (covering) grain spawn (aseptic) with a layer of non-sterile soil is described. Fruit of the white, intermediate, and brown mushroom varieties were consistently and conveniently produced by use of the method. Mushrooms were obtained also by casing commercially available grain spawn. Fruit production from such spawn, however, was less reliable than from the specially-prepared spawn. In addition to being a new and alternate means to evaluate spawn, the method is useful for study of the physiology, genetics and diseases of the cultivated mushroom.

SUMMARYSingle-spore strains from six isolates ofDictyuchus monosporusexhibited the following types of behavior when mated to their respective parental strains and to tester strains of opposite mating reaction to that of the parental strains: (1) heterothallism, in which one of the strains reacted as male and produced antheridia which became applied to oogonia produced by the female-reacting strain; (2) heterothallism, preceded by homothallism in one of the strains in the region of contact between the paired strains, the homothallic strain reacting as male and the opposing strain as female; (3) homo-heterothallism, in which both strains were homothallic in areas adjacent to one another and both were involved in reciprocal cross reactions. With only one exception, there was no sexual reaction between the male isolates and the single-spore strains derived from them. The reactions of the single-spore strains derived from female isolates and mated to their respective parental strains were varied and differed according to the origin of the strains. Variations in sexual behavior also occurred among single-spore strains from some sporangia.

SUMMARYTemperature andpH significantly affect the growth and composition ofPythium irregulareandP. vexans.The apparent optimum temperature for mycelial production by either fungus was lower in liquid medium than on agar, andP. irregulareproduced greater amounts of mycelium thanP. vexansunder all conditions tested. The percentage of soluble sugar content in both fungi decreased markedly with increased incubation temperatures, whereas protein content increased.Pythium vexanscontained more sugar and less protein thanP. irregulareon a percentage basis. Lipid and polysaccharide content varied, but there appeared to be a balancing effect of these components with sugar metabolism. Within the ranges studied, temperature exhibited a more pronounced effect than pH on the growth and composition of these fungi.

SUMMARYThe buoyant density of deoxyribonucleic acid (DNA) from 32 species of poroid Agaricales, three in the Hymenochaetaceae and 29 in the Polyporaceae, was determined by CsCl density gradient equilibrium centrifugation. Expressed in moles per cent the guanine plus cytosine content (GC content) for the Hymenochaetaceae ranged from 50.0 to 51.0, with an average of 50.4 and for the Polyporaceae from 51.0 to 59.5 with an average of 56.1. The results suggest that in the Homobasidiomycetes the GC content of DNA may have taxonomic significance at the species, generic, family and higher levels but the sample is too small to provide a basis for firm conclusions.

SUMMARYA new species in theAspergillus clavatusgroup,A. longivesicaHuang&Raper, is described. Two isolates of this fungus were obtained from Nigerian soil. It is characterized by blue-green heads, a single series of sterigmata, and conidia with prominent connectives. There are two types of conidiophores: short and long. The short conidiophores bear globose vesicles with uniform elliptical conidia. The long conidiophores are wide and thick-walled, producing elongate, fusoid-clavate vesicles, fusoid-clavate heads in which the conidial chains do not adhere to form divergent columns, and conidia which vary in size and shape. The long conidiophore is positively phototropic and elongates only in the presence of light.

SUMMARYA cytological and microspectrophotometric study ofApodachlya brachynema(Hildebrand) Pringsheim was made to determine the site of meiosis in a member of the Leptomitales. Cytological observations revealed two successive nuclear divisions in the developing gametangia. Oögonial nuclei begin division prior to the nuclei in the attendant antheridium. The nuclei in any one gametangium enter into prophase of the first division synchronously and this synchrony is maintained into the following metaphase at which time the divisional figures become asynchronous. The synchronous prophase nuclei in the developing gametangia are 1.8-2.0μin diameter whereas the gamete nuclei resulting from the two divisions are 0.5-0.7μin diameter. Microspectrophotometric analysis reveals that the synchronous prophase and metaphase nuclei in the developing gametangia are 4C and a subsequent reduction division results in the formation of 1C gamete nuclei. Likewise, analysis of the hyphal nuclei clearly indicates a 2C DNA content. The evidence is completely consistent with, and indicative of, gametic meiosis inApodachlya brachynema.

SUMMARYThe basidiocarp ofPolyporus adustusis described as monomitic but its development involves three hyphal systems: generative hyphae, hyphal strands which form the framework and lead the growth of the advancing margin and the dissepiments and felted hyphae in which the hyphal strands are enmeshed.Primordia and basidiocarps show very rapid growth (24–48 hr to turity) and considerable plasticity of form, particularly the basidiocarps. These characteristics are associated with relatively little modification of the hyphae in the context, dissepiments and hymenium, and with marked sensitivity to alterations in the microenvironment.
