ABSTRACTEuryancale phallosporais described as a new species of the Zoopagales found in Japan. The conidium is phallus-shaped and composed of a bulbous basal portion tapering gradually towards the apex which is shaped like an arrowhead. Infection is initiated by the conidium, the apical portion of which lodges in the nematode's mouth. High voltage electron microscopy shows clearly that the mature conidium has a dome-shaped apex. The periphery of the apical dome has 14–15 electron-dense, bar-shaped appendages flaring like a skirt below the apex. Additional, almost median, appendages are present on the conidial wall. The apical dome is composed of an electron-dense hemispherical core covered with an extremely thick cell wall. The thick wall is coated with a layer of weakly staining material. A pouchy appendage connected to the outermost layer by a short and narrow stalk is clearly seen at the distal end of the conidium.

ABSTRACTSeptal pore ultrastructure of selected species of basidiomycetous yeasts was observed.Rhodosporidium dacryoidumandLeucosporidium felliiof the teliospore-forming yeasts have simple septal pores, whereas those ofMrakia frigidaandCystofilobasidium infirmominiatumof the Filobasidiaceae possess dolipores without parenthesomes. InErythrobasidium hasegawianumandKondoa malvinella, also of the Filobasidiaceae, the septa are simple, uniperforate, and associated with pore occlusions.Erythrobasidium hasegawianumhas the septal pore structure typical of members ofRhodosporidiumand related genera in the teliospore-forming yeasts, whereasK. malvinellahas septal pores similar to those ofAtractogloea stillata(Atractiellales). In addition to these features,E. hasegawianumis characterized by the multilayered cell wall structure and enteroblastic conidiogenesis typical of basidiomycetous yeasts.Sympodiomycopsis paphiopedili, a yeast-like anamorph with basidiomycetous nature, has a simple septal pore structure suggesting a possible affinity with the teliospore-forming yeasts. Comparisons between the ultrastructural, morphological and chemosystematic evidence are made, and the taxonomic decisions of recent years are discussed.

ABSTRACTGonapodya proliferawas isolated from a pond in Athens, Georgia, and its development in pure culture is reported for the first time. While the fungus grew well on a diversity of media, the morphology was closest to published descriptions when the fungus was grown on dogwood fruits with twigs held under reduced oxygen conditions. The distributions of chitin, cellulose, glycogen and N-acetyl-D-galactosamine residues in the cytoplasm, cell walls and pseudosepta of freeze substituted hyphae were studied with gold-complexed enzyme and lectin probes. The presence of cellulose in the cell walls of this member of the Chytridiomycetes was substantiated by compositional and methylation analyses of a cell wall extraction that was selective for cellulose. The observation thatHelix pomatiaagglutinin binds to glycogen in thin section is discussed, as is the first reported detection of cellulose in a member of the Chytridiomycetes.

ABSTRACTBasidiospores ofCronartium quercuumf. sp.fusiformewere germinated under different conditions of agar concentration, pH, washing times, and spore exudate concentrations. At agar concentrations of 0.05-3% the percentage of washed spores that germinated directly, i.e., produced germ tubes, was significantly different from the percentage of unwashed spores that germinated directly. Ninety-nine percent of the washed spores germinated directly at all agar concentrations, while only 4–11% of unwashed (directly cast) basidiospores germinated directly. On pH 3 agar, germination of directly cast basidiospores was only 2%, but 98% of germinating spores germinated directly (produced germ tubes). On pH 4–11 agar, spore germination varied from 70–90% and was predominantly indirect (forming secondary basidiospores). When spores were washed in water at pH 2.2 or pH 5.5 for 2 or 5 min, percent direct germination was 10 times greater than those spores not washed with water. More than 95% of spores germinated directly after 30-min washing with water at either pH 2.2 or 5.5. Both spore exudates washed from the spores and those produced during germination were collected and reapplied to the spores. These applications increased the percentage of direct germination in directly cast spores but did not alter germination type in washed spores. The exudates inhibited germ tube length in both directly cast and washed basidiospores.

ABSTRACTThe synnematous, fungal entomopathogenHirsutella longicollavar.longicollais distinguished from a varietyHirsutella longicollavar.cornutaon the basis of the synnematal morphology. Random Amplified Polymorphic DNA (RAPD) fingerprinting supports the view that these fungi are genetically distinct. Two different banding patterns, one corresponding to isolates of the species and the other to the variety, were generated with two primers and DNA from 19 isolates. Further analysis using nine isolates (four of the species and fi ve of the variety) with three additional primers confirmed the distinction between the species and its variety. This work illustrates the usefulness of the Random Amplified Polymorphic DNA technique in addressing taxonomic questions.
