ABSTRACTThe spore wall ofFuligo septicais composed of an exterior electron-dense, spined layer and an interior more electron-transparent, fibrous layer. The fibrous material in the interior layer is often tightly packed. During germination, the exterior layer splits, but the inner layer does not. A remnant of this inner layer surrounds the newly released protoplast if the remnant is not left behind. The remnant appears to be derived from the fibrous wall material. However, swarm cells appear to be free of structural wall components. Cytochemical tests suggest peroxidase activity in the inner waH layer. Initial localization of peroxidase activity near the split in the outer wall may indicate peroxidase plays a role in the germination process.

ABSTRACTIxechinusHeim ex Horak andMucilopilusWolfe are evaluated in the light of new information revealed in type studies of taxa belonging to or suspected of belonging to the Ixechineae. It was found thatMucilopilusandIxechinusdiffer consistently in four primary and secondary characters. They are, therefore, viewed as good genera.Mucilopilus mexicanus(Guzmán) Wolfe andIxechinus viridis(Heinemann&Goossens) Wolfe are proposed as new combinations in light of the new data presented. Spore print color as a basis for delimiting genera of boletes is discussed.

ABSTRACTFruiting bodies ofSphaerobolus stellatuscollected in the field and grown in Petri dish culture were examined by light and electron microscopy at near maturity and at maturity (both pre-glebal discharge and post-discharge). Basidiospores, gemmae, and“fat cells”which comprise the ejected glebal mass were studied. The parenchyma cell layer and its underlying hyphal mat which are collectively involved in glebal ejection are nearly indistinguishable before and after ejection except for the marked diminution of a-glycogen rosettes in the parenchyma cell layer after discharge. The fat cells of the collenchyma layer making up the glebal surface broke down near fruiting body maturity and became amorphous masses of cellular debris with interspersed fragments of cellular membranes, most noticeably sets of parenthesomes. Following ejection of the glebal mass, previously germinated gemmae continued to grow and additional gemmae germinated. These constitute the germinal cell population in cultured material. Basidiospores did not germinate under normal laboratory conditions. The stickiness of the discharged glebal masses was clearly due to material from degraded collenchyma cells.

ABSTRACTPisolithus tinctoriusused several different carbon sources, as determined by biomass measurement, and generally metabolized monosaccharides more efficiently than disaccharides. No obvious difference was noted when utilization of amine and nitrate forms of nitrogen were compared. The optimum carbon/nitrogen ratio for conditions tested appears between 10/1 and 20/1. Modified Melin-Norkrans medium buffered with citric acid/sodium citrate was used to optimize sugar and nitrogen concentrations. A liquid inoculum in shake flask culture formed a biomass in six da that would otherwise require six wk by static flask or solid substrate methods. By optimizing growth rate in a medium suitable for deep tank fermentation, and with proper formulation, it is feasible to consider producing this ectomycorrhizal fungus for commercial use in forestry.

ThesePhyllostictaspecies differ in morphology, colony characteristics, and pathogenicity.Phyllosticta vacciniicauses a fruit rot and leaf spot of cranberry in Massachusetts and New Jersey.Phyllosticta elongatais associated with latent infections of cranberry leaves and fruit, berry speckle, and is widely distributed.

ABSTRACTExtracellular, phosphate-repressible ribonucleases (RNases) have been partially purified from the imperfect yeast,Cryptococcus laurentiiand a haploid, yeast-like isolate of the heterobasidiomycetous fungus,Tremella foliacea.The purified enzymes were similar in several respects, (i) Maximal hydrolytic activity occurred at acid pH (5-5.5). (ii) Divalent cations were not required for enzyme activity, (iii) Ethylenediaminetetraacetic acid, p-chloromer- curibenzoate, and iodoacetic acid were not inhibitory, (iv) Both were found to be phosphotransferase enzymes producing nucleoside-2′,3′-cyclic phosphate intermediate products with subsequent hydrolysis to the corresponding nucleoside-3′-phosphates. (v) Degradation of RNA was by an apparent exonucleolytic mechanism. The data support a possible phylogenetic relationship between the two species.

ABSTRACTThe occurrence of five species of Endogonaceae previously unreported from Florida is noted and seven new species (six inGlomus) are described.Glomus aggregatumis the only new species from Florida which forms sporocarps in and around roots and is similar in appearance to the previously described taxonG.fasciculatum. Glomus intraradicesforms yellow-brown spores almost exclusively in the root.Glomus claroideumhas hyaline to yellow spores which are smaller but similar in appearance to the previously described taxonG. darum. Glomus leptotichumhas relatively large white spores with a thin wall bearing a faint reticulum. Spores ofGlomus fecundisporumare gray-white, have two walls and are frequently borne in clusters.Glomus tortuosumhas a sinuous hyphal mantle covering the spore.Gigaspora albidahas generally smaller spores with slightly thicker walls but is similar in spore shape and color to the previously described taxonG. gigantea.A synoptic key to the 34 species of Endogonaceae described in Florida is included.

ABSTRACTThirty-four commercial and 162 research-maintained lines ofAgaricus brunnescensdeposited in The Pennsylvania State University Mushroom Culture Collection were examined electrophoretically for isozyme variation. Based on the variability at five structural gene loci (Gpt, Adh, Mpi, Pep-LLL-1, and Pep-LLL-2) they were partitioned into 27 genotypic classes. The allelic variability observed in mycelial extracts of these lines potentially allows the recognition of over 20,000 unique genotypic classes. The fact that only five genotypic classes were actually observed among commercial lines examined is evidence that the common mushroom is a near monoculture. Data from single spore-derived cultures ofA. brunnescensrevealed that most of these cultures retained heterozygosity at those loci heterozygous in the parental line. A limited number of recombinants were noted, i.e., heterozygous for some loci and homozygous for others. A small fraction (<5%) were homozygous at multiple loci suggesting that they may be homokaryotic. These observations support the contention that low levels of meiotic recombination occur and the chromosomal segregation is nonrandom inA. brunnescens.
