TheN‐terminal peptide of salmon proopiocortin has been isolated and the primary structure including two disulfide bonds elucidated. The peptide consisted of 76 amino acid residues, which is 27 residues shorter than the bovine and human peptides. TheN‐terminal 44 residues of the teleost peptide exhibited significant sequence homology to those of the mammalian peptides. The synthesis and isolation of symmetrical anhydrides ofNα‐9‐fluorenylmethyloxycarbonyl (Fmoc) amino acids using water soluble carbodiimide is described. These compounds were used in a solid phase peptide synthesis of methionine‐enkephalin on a p‐benzyloxybenzyl ester polystyrene 1% divinylbenzene resin support. Homogeneous free pentapeptide was obtained in 42% overall yield. Beta‐endorphin has been isolated from equine pituitaries. Conformational energy calculations were carried out on the peptide enkephalins (ENK) and selected analogs to find those conformers of low energy. The analogs studied include [D‐Ala2]Enk‐NH2, [D‐Ala2]Enk, [D‐Met2, Pro5]Enk‐NH2, [D‐Ala2, D‐Phe5]Enk, [D‐Ala2, D‐Leu5]Enk, [D‐Ala2, (N‐Me)Phe4, Met5] Enk‐NH2and [D‐Ala2, (N‐Me)Met5]Enk‐NH2. The a‐helix content of human β‐endorphin (βh‐EP) has been determined by circular dichroism (CD) in solutions ranging from 0 to 95% methanol in water. In addition, the CD spectra of βh‐EP‐(1–30),‐(1–29),‐(1–28),‐(1–27),‐(1–26), ‐(1–21) and ‐(1–15) have been examined in 90% methanol, and their a‐helix contents estimated using parameters determined for this solvent.
