Abstract.The effect of different amounts and types of dietary fat on faecal steroid excretion in healthy volunteers was examined. In the first experiment with sixty volunteers four diets which differed in type and amount of dietary fat only were compared during a test‐period of 5 weeks. Faeces were collected during 7 days before and at the end of the test period and analysed individually. In the second experiment with thirty‐five volunteers, a low fat diet containing 22% of energy as total fat (4% as polyunsaturated fatty acids) was compared with a moderate fat diet (30% of energy as total fat; 11% as polyunsaturated fatty acids) during 13 weeks. Faeces were collected during the first 7 days of the test period and during 7 consecutive days near the end of the test period and pooled per dietary group.Excretion of faecal neutral steroids and bile acids was not significantly influenced by amounts of total fat ranging from 22% to 40% of daily energy or by differences in polyunsaturated to saturated fatty acids ratio from 0.2 to 1.7. The only exception was the excretion of bile acids in the second experiment which was 0.19 mmol/24 h (75 mg/24 h) higher on the low fat diet than on the moderate fat, high polyunsaturated fat diet (P<0.

Abstract.A study of plasma and cardiac concentrations of amiodarone during the course of long‐term oral therapy was made possible by the improvement of analytical high performance liquid chromatography of plasma and tissue extracts.The plasma level was found to increase linearly with the daily dose of the drug, above a threshold value ofc.1.9 mg kg‐1day‐1. Similarly, the cardiac content increased linearly with the daily dose, with no threshold, showing that the drug is taken up and accumulated in the cardiac tissue, with no obvious difference between atrial and ventricular samples (P>0.05). Both plasma and heart showed no saturation at high drug intake, a justification for increasing the oral intake in severe cases. Abstract.Serum hydroxyproline‐containing peptides were separated into three fractions, i.e. large protein, polypeptide and smaller peptides, by gel filtration on Sephadex G‐200. The large protein was presumed to be the Clq subunit of the first component of complement by an immunological analysis. The polypeptide had an electrophoretic mobility corresponding to β‐globulin and its molecular weight was estimated between 35 000 and 45 000 by gel filtration analysis. The polypeptide might well be defined as hyproprotein. The molecular weight of smaller peptides was estimated between 1400 and 3000.As compared with the control value, hyproprotein levels were significantly increased in rheumatoid arthritis, systemic lupus erythematosus and chronic liver disease. Clq levels were slightly lowered in rheumatoid arthritis and systemic lupus erythematosus, and were slightly increased in chronic liver disease but were not significant. In all patient groups smaller peptides levels remained in the normal range. Abstract.Porcine mucosal heparin was chemically depolymerized. The depolymerization was stopped at different steps to obtain two low molecular weight (LMW) heparins with a molecular weight of 10 000 and 6000, respectively. The LMW heparins were testedin vitrofor anti‐clotting activities and for platelet serotonin release in different systems in comparison with normal heparins, dermatan and heparan sulphate.After addition of various amounts of heparin preparations to washed platelets, no significant release was observed for all tested heparins. On the contrary, different heparins showed an inhibition of serotonin‐release induced by collagen in platelet rich plasma, whereas the ADP‐induced release was increased. The effect on the platelet release appears related to the molecular weight. In fact, it is significant only for normal heparins whereas it is not for LMW heparins. A good relation was observed, also, between anti‐activated factor X activity/antiglobal clotting activity (Xa/APTT) ratio of different heparins and the effect on platelet

Abstract.Duodenopancreatectomy induces a severe glucagon deficiency and elevated plasma concentrations of alanine, aspartate, glycine, proline, serine, arginine, citrulline, ornithine, phenylalanine and tyrosine. Restoring high physiological plasma glucagon in six such patients by infusing 0.3 mg/24 h of exogenous glucagon reduced significantly (P<0.01 or 0.001) the mentioned amino acids (except phenylalanine) and further asparagine, glutamine, methionine and threonine. In six normal subjects the same infusion reduced significantly (P<0.05 to 0.001) plasma alanine, asparagine, glutamate, glutamine, glycine, proline, serine, threonine, arginine, ornithine, lysine and tyrosine. However, the effect was significantly (P<0.01 or 0.001) less marked for alanine, glutamine, glycine, methionine, serine, threonine and arginine. This particular glucagon sensitivity of duodenopancreatectomized patients suggests that glucagon deficiency is the cause of their hyperaminacidaemia. By co‐trust, lipoprotein concentrations were virtually unaffected by either glucagon deficiency or its replacement.In the light of the marked hypoaminacidaemia in glucagonoma patients these results attribute to glucagon a major role as a regulator of protein metabolis

Abstract.Among 156 patients admitted to hospital because of suspected solvent intoxication or dementia following long‐term occupational exposure to organic solvents, twenty‐three had elevated aminotransferases in serum. In thirteen of these no particular cause of this elevation could be found and the chronic exposure to aliphatic and aromatic organic solvents is therefore likely to have played a pathogenetic role and a liver biopsy was performed.Liver histology revealed significant steatosis in eleven patients and in six of these focal necroses were also observed. Moreover, six of the biopsies showed enlarged portal tracts with fibrosis. Necrosis was only observed in liver specimens from patients who recently had been exposed to solvents.A causal relation between occupational exposure to aliphatic and aromatic organic solvents such as white spirit, xylene, toluene and styrene, and toxic liver injury is suggested by this descriptive study but needs confirmat

Abstract.As a result of overdosage a 77‐year‐old patient with heart disease developed digitoxin intoxication, associated with arrhythmias, extracardiac symptoms of intoxication and severe thrombocytopenia. Treatment with digoxin‐specific antibody fragments relieved the signs and symptoms of intoxication within a few hours. The rise in platelet count from the pretreatment value of 26 000/mm3to 47 000 within 12 h and to over 60 000/mm3within 16 h of starting the antibody infusion may also be attributed to the treatment with antibodies.
