摘要:
An indirect atomic absorption spectrometric assay for epoxide hydrolase was reported. The enzymatic product 1,2-diol was oxidized by an excess of potassium periodate, and the unconsumed periodate was preferrentially precipitated by lead periodate. The precipitate was acid digested followed by atomic absorption determination of lead in the resulting solution. Trans-stilbene oxide (TSO) induced rabbit and tilapia epoxide hydrolase activities by 350% and 210% at the 490 mg TSO/kg treatment while Aroclor 1254(AR) only slightly increased their activities at the 200 mg AR/kg treatment. The 1,2-epoxy-3,3,3-trichloropropane (TCPO) at 0.1 mM completely inhibited tilapia (control) and rabbit (490 mg TSO/kg) epoxide hydrolase activities.

A radioimmunoassay (RIA) for measuring total urinary estriol (E3) in pregnancy is described. Incubation of 0.1 ml urine for 30 minutes at 47°C with Helix pomatia enzyme containing both β glucuronidase and sulfatase activity, hydrolyzed estriol conjugates. Incubation of samples with E3-6,73H and diluted antiserum raised against estriol-6-one carboxymethyloxime coupled to bovine serum albumin, was carried out at 4°C for one hour. Recovery averaged 90 ± 6% (SD) for E3-16-glucuronide, 88.3 ± 5.7% for E3-3 glucuronide, and 84.6 ± 5.9% for E3-3 sulfate. The assay sensitivity was 25 pg, with a within and between assay coefficient of variation of 6.5% and 9.3% respectively. Results were unaffected by the presence of glucose, chlorothiazide, or hydrochlorothiazide. A close correlation was obtained between results by RIA and a specific colorimetric method. One technician can complete 50 samples within five hours.

摘要:
A reliable mass fragmentographic determination method for acetazolamide levels in body fluids was developed to favor the pharmacokinetic study on the patients receiving multiple doses. The use of 2-(N-propionamido)-1,3,4-thiadiazole-5-sulfonamide as the internal standard permitted the internal standard to be added to the sample before the extraction, and enabled the single ion measurement at m/e 249. Single ion measurement at m/e 249 detected 25 pg of acetazolamide. Multiple ion detection at m/e 249, 264 and 278 favored more accurate identification of each compound on the chromatogram. The assay results of the samples from volunteers were consistent with the ones reported elswhere.

Nadolol, a beta-blocking agent, has been assayed in pure form and in pharmaceutical formulation by acidimetric titrations using visual, potentiometric and conductimetric methods for detecting the end point in aqueous as well as in non-aqueous media. The method of assay involves the determination of the amount of nadolol in a given weight of sample. Six samples were run for each method and the percentages of recovery of nadolol from its tablets were calculated. For aqueous titrations, the average percentages of recovery for the visual, potentiometric and conductimetric methods are 100.6 ± 1.0, 99.3 ± 1.9 and 100.1 ± 2.0 respectively. For the non-aqueous titrations, the average percentages of recovery are 98.6 ± 0.8, 99.5 ± 0.5 and 98.4 ± 1.3 for the visual, potentiometric and conductimetric methods respectively.

A rapid and sensitive method is described for the direct assay of oxymetazoline hydrochloride using reversed phased high pressure liquid chromatography. It is as accurate and faster than the U.S.P. or N.F. methods.

摘要:
A simple and rapid spectrophotometric method for determination of thiacetazone is described. Thiacetazone forms a stable greenish coloured complex with Cobalt (II) under approprjate experimental conditions. Maximum absorbance occurs between 660 and 690 nm. The values are reproducible at 680 nm and Beer's law is obeyed at this wavelength. Thiacetazone is determined in dosage form and in presence of other anti-tubercular drugs. It is also determined in presence of commonly used drugs such as chlorpheniramine maleate, riboflavin, pyridoxine hydrochloride and dicalcium phosphate.
