摘要:
This paper discusses the appropriate conditions for determining trace cooper by flameless atomic absorption spectrophotometry with the aerosol deposition technique. Seventeen samples of cord blood serum from the new born infants in the Hefei district of China were analyzed by this method. The range of trace copper in the samples is between 0.40-0.77 ppm, mean value 0.55 ppm. The recovery is over 90%. The relative standard deviation is 2.8%, determined by measurement of six times on a certain serum sample.

摘要:
A simple method for the enantioselective determination of 2-halopropanoic acids and 2-halobutanoic acids with two bacterial 2-halo acid dehalogenases has been developed.L-2-Halo acid dehalogenase acts specifically onL-2-haloalkanoic acids, andDL-2-halo acid dehalogenase acts on both enantiomers of the acids. The dehalogenation was followed by determination of halogen ions released. Linear relationship was established between the absorbance at 460 nm, and the amounts ofL-2-haloalkanoic acids (0.025-0.5 μmol) or the racemates (0.05-1.0 μmol). TheD-isomers were estimated by subtracting the amounts ofL-isomers from those ofDL-2-haloalkanoic acids.

摘要:
The 2,3-diphosphoglycerate content of blood was determined by a bacterial NADH-linked bioluminescence measurement technique. The reaction conditions were chosen so that either an end-point or kinetic measurement was possible and easy to perform. Blood samples from twenty clinical patients were measured for their 2,3-diphosphoglycerate content using both luminescent and conventional spectrophotometric techniques. Correlation between the two methods was good and recovery for the bioluminescent method when adding a known quantity of 2,3-diphosphoglycerate was 95%. Luminescence analysis makes it possible to measure 2,3-diphosphoglycerate in just a few drops of blood taken from an adult's finger or from a baby.

A sensitive method for the rapid determination of activities of soluble or immobilized enzymes, based on the electrochemical detection of hydrogen peroxide is described. Kinetic studies (Vmax and KMdeterminations) can be performed for all H2O2generating enzymes (i.e. most of the oxidases) using an amperometric probe with a platinum anode at a fixed potential.

摘要:
An electrochemical method for a rapid assay of total cholesterol in serum using free cholesterol oxidase (EC 1.1.3.6) and cholesterol esterase (EC 3.1.1.13) is described. Hydrogen peroxide generated by cholesterol oxidase is amperometrically monitored at + 650 mV vs Ag/AgCl and two types of responses can be recorded : a steady-state response and a dynamic one. The dynamic response is obtained within one minute. Interfering species are detected prior measurement and a linear calibration curve can be obtained for total cholesterol in the sample in the range 0.4-8 mM useful for clinical analysis.

摘要:
Liquid-membrane electrodes sensitive to scopolamine+and N-butyl scopolammonium+are described for the direct or indirect assay of scopolamine hydrobromide and N-butyl scopolammoniumbromide, respectively, in pharmaceutical preparations. All electrodes exhibit a linear response to respective organic cation over the range of lo−1to lo−4M. No interference from the common imgredient substances was observed.

A method involving ion-exchange chromatography and titration was developed. A diluted sample was run through a cation-exchange resin column. After the eluate was boiled, it was titrated with 0.02NNaOH to find the sodium citrate content (Titration I). An equal volume of the diluted sample was eluted through an anion-exchange resin column and the eluate was titrated with 0.02NHCl to assay for total sodium bicarbonate and sodium citrate (Titration II). Subtracting the result of Titration I from that of Titration II, gave the sodium bicarbonate content. The cation-exchange chromatography step was validated previously. The anion-exchange chromatography step was validated by assaying pure sodium bicarbonate and sodium citrate separately. The overall percent recoveries (+ SD) were 98.9+0.2% for sodium citrate and 98.1+0.1% for sodium bicarbonate (n = 3). Average percent recoveries (+SD) from synthetic mixtures of the two salts were (n = 4): 99.3+0.5% for sodium citrate and 98.0+1.2% for sodium bicarbonate. The method was applied to commercially available formulations.
