摘要:
An improved method for the conjugation of Fab' to B-D-galactosidase fromEscherichia coliwas developed. The enzyme with thiol groups was treated with excess of N, N'-o-phenylene-dimaleimide to introduce maleimide groups, and the maleimide-enzyme was reacted with thiol groups in the hinge of Fab' to form Fab'-B-D-galactosidase conjugate. Gel filtration of Fab' was required only once, and the recovery of Fab' in the conjugate was 71–81%, while in the previous method, gel filtration was required twice and the recovery was 31–38 %. Although the enzyme activity was decreased slightly (8–12%) by the dimaleimide treatment, the conjugate was as useful for sandwich enzyme immunoassay as that obtained by the previous method. 1gG also could be conjugated to B-D-galactosidase more efficiently in the present method than in the previous one.

摘要:
A novel biosensor, biophotodiode, is proposed for the determination of substances of biochemical and clinical importance. The biophotodiode is a unique combination of a photodiode and matrix-bound peroxidase. Since the enzyme catalyzes the luminescent reaction of the luminol-H2O2system, this assembly is effective on the determination of hydrogen peroxide. The photocurrent of the sensor is correlated with the hydrogen peroxide concentration ranging from 1mM to 10 mM. A biophotodiode for glucose is also constructed by assembling a glucose oxidase-peroxidase bienzyme membrane and a photodiode.

摘要:
Li FT-NMR was used for the detection of different lithium species in human serum. Two serum samples from mentally ill patients undergoing lithium treatment were studied. Both samples gave spectra with a major peak corresponding to aqueous lithium ion and a much smaller peak at −0.3 ppm relative to the lithium ion peak. Two serum samples from normal persons were spiked with lithium but the resulting spectra showed only the lithium ion peak.

摘要:
The level of human chorionic gonadotropin (hCG)-like substance in normal serum was measured using 50 μl of serum specimen by a highly specific and sensitive sandwich enzyme immunoassay for hCG which did not significantly cross-react with hLH. The level was calculated using a standard hCG and expressed in mIU/ml as that of hCG. In normal male (20–59 years of age) and normal, non-pregnant female (20–42 years of age) adults, the level was below 0.6 mIU/ml with mean levels of 0.16 mIU/ml (n=209) and 0.14 mIU/ml (n=36), respectively. In older subjects, however, the level tended to rise. In normal male adults, aged 60–91 years, the level was mostly below 0.8 mIU/ml (mean=0.56 mIU/ml, n=99), but levels of 15 specimens out of 99 were 1.1–5.5 mIU/ml. In normal, non-pregnant female adults, aged 45–88 years, the level was widely distributed between 0.1 and 11.7 mIU/ml (mean=1.6 mIU/ml, n=141). In these older female adults, levels of hCG-like substance were not well correlated to either levels of hLH and hFSH, while levels of hLH and hFSH were well correlated to each other.

摘要:
The initial rate of hydrolysis of N-acetylmethionine by hog kidney aminoacylase (EC 3.5.1.14) was determined by repetitive flow injection analysis. The reaction mixture was slowly pumped through an injection valve from which portions were injected into a reagent stream consisting of o-phthaldialdehyde and mercaptoethanol in a borate buffer. This reagent converted the product methionine into a spectrophotometrically-detected isoindole. In a typical assay 15 data points were obtained within the first 22 min of reaction. Initial rates obtained by the method were equal to those obtained by ninhydrin assay.

A new procedure for the determination of lecithin and sphingomyelin in amniotic fluid is described. rapid column chromatography in the sample preparation steps with computer-assisted infrared spectroscopy in the final determination. Lipids are extracted from amniotic fluid with a chloroform/methanol solvent. The organic layer is separated and evaporated to dryness.
