摘要:
The paper deals with new amperometric analytical systems based on biocatalysts. Discussed are: the use of immobilized multienzyme systems, the increase in sensitivity of devices by chemical amplification, the use of dehydrogenases, hydrolases and microorganisms, and the application of biochemical cells as analytical devices. The macrokinetic dependence of the action of systems is considered and the ways for their perspective development are proposed.

摘要:
Authors propose o direct determination of nitroprusside in human serum by alternative current polaro-graphy. After protein elimination by perchloric acid, the polarographic measurement is made at pH 7,4. Levels about 1 10−5M (2 mg/1) can easily be determined. The method is rapid and sensitive.

摘要:
A new potentiometric gas sensing concept is used in conjunction with a flow injection technique to rapidly determine ammonia-nitrogen in the 10−5to 10−2mol/L range. The system designed utilizes a novel tubular flow-through PVC-nonactin membrane electrode to detect ammonium ions formed in a recipient buffer stream as the sample flows through a gas dialysis unit. At least 30 samples/h can be determined with good precision (⩽ ± 7%) for samples ⩾ 10−5mol/L in ammonia-N. Preliminary data concerning the analytical application of this system for blood ammonia-N measurements is presented. A novel gas dialysis method for removing ammonia from blood samples is also described.

摘要:
An enzymatic determination of urinary total 17β-hydroxysteroids is described. The principle of the method is as follows: after hydrolysis with β-glucuronidase and solvolysis, the ethyl acetate layer, which is washed with alkaline solution and water, is transferred to two test tubes, then each of them is evaporated. To one test tube is added 3β,17β-hydroxysteroid dehydrogenase (EC 1.1.1.51, from P. testosteroni), NAD+, 2-ρ-iodophenyl-3-ρ-nitrophenyl-5-phenyltetrazolium chloride (INT), and diaphorase (EC 1.6.99.2, from C. kluyveri) for determination of total 3β,17β-hydroxysteroids. To another test tube is added 3β-hydroxysteroid oxidase (EC 1.1.3.6, from B. sterolicum) for determination of total 3β-hydroxysteroids. The value of 17β-hydroxysteroids is calculated by subtracting the value for 3β-hydroxysteroids from the value for the 3β,17β-hydroxysteroids.

摘要:
A specific and relatively sensitive method for the determination of human urinary free methyldona has been developed. The recovery of methyldopa is fast and simple employing urine acidification, organic extraction of unwanted acids, and dilution. The chromatography is reverse-phase high performance liquid chromatography with an ion-pairing agent utilizing electrochemical detection. The range of precision and linearity is approximately 2–500 μg/ml methyldopa under the conditions set forth in this paper.

摘要:
A method is described for determining total platinum in tissues of animals receiving cisplatin. The method is based on total combustion of the tissue, leaving a platinum residue devoid of organic material that can be dissolved inaqua regiaand analyzed by carbon-rod furnace atomic absorption spectrophotometry. Combustion eliminates matrix effects in final read-out, providing a means for precisely and reproducibly analyzing a variety of tissues. Platinum levels of 200 ng/0.5 g of sample can readily be analyzed with recoveries from all tissues approaching 100%. The validity of the method is demonstrated by giving rats intravenous doses of cisplatin and subsequently measuring tissue levels of platinum.
