摘要:
The DC polarographic behavior of the Cu(II)-inosine complex was investigated, in aqueous solutions of inosine in Britton Robinson buffer solutions at pH's from 1.0 to 4.0. At low concentrations of free inosine ion, only one polarographic wave was found corresponding to a two-electron reduction.

摘要:
A simple procedure for separation of ribose-5-phosphate, deoxyribose-1-phosphate and ribose-1-phosphate is based on high performance liquid chromatography using reversed phase 4 × 300 mm “μ Bondapak/NH2” column. The column is equilibrated with 0.13 M borate buffer (pH 7.5) followed by gradient elution of ribose-5-phosphate, deoxyribose-1-phosphate and ribose-1-phosphate using water, 0.05 M borate buffer containing 0.1 M MgCl2(pH 9.6) and 0.05 M sodium acetate-acetic acid buffer containing 0.1 M MgCl2(pH 5.0) as eluants respectively. Eluates of borate complex “μ Bondapak/NH2” column are brought to pH 9.6 by the addition of 1 N KOH and enzymatically hydrolysed with alkaline phosphatase (EC 3.1.3.1) to release the free pentoses. The free pentoses are mixed with a reagent solution prepared from aqueous solution of 2% cyanoacetamide and 0.6 M borate buffer (pH 9.6), and the mixture is boiled for 10 minutes and the absorbance of the product is measured at 276 nm using a spectrophotometer.

摘要:
The coupling of an enzyme immunoassay for factor VIII-related antigen with a commercial glucose oxidase based amperometric sensor permits the determination of 1.6 to 16 ng of factor XIII-related antigen in human plasma. Further pure amperometric sensors or amperometric enzyme sensors for determination of the main marker-enzymes of enzyme immunoassays are described.

摘要:
The selectivity of a previously reported bacterial arginine electrode has been greatly enhanced, with no loss of response to arginine. The response of this sensor to glutamine and asparagine was found to be due to a contaminant growing in the bacterial layer of the electrode. The addition of 1 × 10−3M sodium azide to the working buffer and cold storage of the electrode effectively controlled the growth of the contaminating bacteria and maintained the low initial response of the electrode to these amino acids in the concentration range of 4 × 10−5to 1.5 × 10−3M. At these optimum conditions, the response of the sensor to arginine was unaffected by the presence of a mixture of amino acids at physiological levels.

摘要:
A “simultaneous” enzyme-linked immunoassay for the measurement of anti-thyroglobulin autoantibody in human serum was studied, in which human thyroglobulin-coated silicone rubber rod, ß-D-galactosidase- or horseradish peroxidase-conjugated human thyroglobulin and serum sample were mixed at the same time. In order to obtain a suitable range of the measurable antibody, an appropriate amount of human thyroglobulin-enzyme conjugate was carefully selected. A successful simultaneous assay for antithyroglobulin was obtained using thyroglobulin-ß-D-galactosidase conjugate, in which the minimum amount of detectable antibody was approximately 10 ng/ml using 5 μl of serum. This sensitivity was 20-fold higher than that in two-step sandwich enzyme-linked immunoassay reported previously.

摘要:
An atomic absorption spectroscopic (A.A.S.) method for microdetermination of sulfathiazole, sulfadiazine and sulfamerazine has been developed. The procedure is based upon the measurement of the metal content of the sulfur metal chelate. The experimental conditions as well as the molar ratios are thoroughly studied. The percentage recoveries for the 3 compounds tested were found to be 99.5 ± 1.30, 99.7 ± 0.80 and 100.6 ± 0.78, respectively.

Electrochemical monitoring of 1, 4-dihydroxy-5, 8-bis-[2-(2-hydroxyethy1) aminoethy amino]-9, 10-anthracenedione was employed for the quantitation of this cancer chemotherapy drug following high performance liquid chromatography. Detection was performed by oxidation of the compound at a glassy carbon electrode at +0.80 V vs. SCE. A rapid and convenient sample treatment procedure was developed which is suitable for determination involving physiological samples. A detection limit of 2 ng injected was obtained for urine samples.
