摘要:
Differential pulse polarography was utilized in voltammetric immunoassay to determine human albumin and albumin antiserum. Albumin was labelled with lead, which exhibited a reduction peak with E1/2at −680 mV vs SCE. The current is proportional to the concentration of lead labelled albumin, and hence the albumin at a fixed lead concentration, and it decreases in proportion to the amount of albumin antiserum added. Albumin concentrations of 2.0 × 10−6–10 × 10−6M and 10–50 μl of goat albumin antiserum (titer 1:16 = maximum dilution causing precipitation with equal volume of 1 mg/ml human albumin) were measured in the presence of 2 × 10−6Mlead.

摘要:
A new immunoassay separation technique was developed based on the use of antibody-polymer conjugates with reversible solubility characteristics. Antibody was coupled to sodium alginate with water-soluble carbodiimide. These conjugates could be used in place of unmodified antibody in various assay formats and removed from solution when desired by converting the polymer to an insoluble form through pH adjustment or addition of metal ions. An enzyme immunoassay for theophylline was used to demonstrate the usefulness of the new approach for both manual and partially automated procedures.

Effect of temperature was examined on the sensitivity of sandwich enzyme immunoassay for human chorionic gonadotropin (hCC) with anti-hCG Fab'-horseradish peroxidase conjugates prepared by using three different reagents (N-succinimidyl 4-(N-maleimidomethyl) cyclohexane-l-carboxylate, glutaraldehyde and metaperiodate). The non-specific bindings of the conjugates to anti-hCC IgG-coated polystyrene balls were much lower at 20°C than at 37°C, and the specific bindings were slightly higher at 20°C than at 37°C. The lowest non-specific binding and the highest specific binding were obtained by incubation at 20°C with the maleimide conjugate. As a result, the sensitivity could be more easily improved by incubation at 20°C than at 37°C and the highest sensitivity was obtained with the maleimide conjugate. Similar results were also obtained for other macromolecular antigens such as human ferritin and α-fetoprotein.

The aerobic oxidation of reduced pyridine phosphonucleotides catalyzed by horseradish peroxidase (E.C.1.11.1.7) (HRP) in combination with Mn2+-ions, phenolics and redox dyes was used to perform NADH and NADPH determinations with an amperometric HRP enzyme electrode. Optimal operational conditions have been elaborated for such a sensor and a linear relationship between derivative current (dI/dt) and NADH-concentration was obtained between 2.10−5and at least 2.10−4mol/l. The coefficient of variation was equal to or smaller than 4 %. The time for one measurement was less than 10 minutes including equilibration of the electrode.

摘要:
A procedure is described which permits the determination of maprotiline in biological fluids for concentrations in the range of 10 to 1000 ng/ml. It relies on the use of N-desmethylclomipramine as internal standard, on derivatization of the secondary amines with heptafluorobutyric anhydride, and the use of gas chromatography with electron-capture detection. The method is linear, specific, accurate and precise. It is suitable for routine analyses and is particularly applicable for monitoring the levels of maprotiline in the blood of patients treated for depressive illness.

摘要:
An ion selective electrode based on a liquid membrane, constituted by a solution of tributylcetylphosphonium benzoate in nitrobenzene, is employed to determine cholic acids in pharmaceutical products by calibration curve and standard addition methods. The results are compared with those obtained with enzymatic and calorimetric methods. The proposed technique is simple, rapid and sufficently accurate.
