摘要:
The determinazione of HCG is performed by the enzyme immunoassay technique using acetylcholinesterase enzyme as lable and a pH electrode to follow the enzyme activity. The antibody to HCG is immobilized in a membrane form with a polyethylene net; to the sample a certain amount of enzyme-labeled HCG is added, the antibody bound membrane is dipped for two hours; after it is taken out, washed and immersed in an acetylcholine Solution and a pH variation can be detected by placing a glass pH electrode with flat surface against the memberane for a few minutes. the pH variation is related tio the HCG concentration of the sample.

摘要:
Two different ATP extraction methods, one using perchloirc acid and the other boiling Tris bufer, for ATP determinations in myocardial samples werer investigated and compared. A bioluminescent assay was used to quantify the extracted ATP. In spite of some drawbacks, eprchloric acid extraction is zix times more efficient thatn boiling Tris buffer extraction for releasing ATP from myocardial tissue. The perchloric acid extraction was used to determine ATP depletion in the endocardium of isolated ischemic rat hearts.

摘要:
The fluorescence development from amino acids andj epptides using N-chlorodansylamide (NCDA) is described. NCDA gave fluorecence on reaction with proline which gives no fluorescence with o-phthaladehyde (OPTA) or with fluorescamine. NCDA showed excellent sensitivity in the assay of peptides which give only poor fluroescene on reaction with OPTA. The standard curves for amino acids and peptides were linear in the range from 10 nmol to 100 nmol/sample and passed through the origin.

摘要:
Glucose released from complement mediated immuno-specific damage of glucose loaded lipososmes sensitized with lipid antigens was detected by an amperometric glucose enzyme electrode in view of devising a simple method for electrochemical immunoassay. This method involves the indirect determinatio of antibody and complement levels by measurement of the relesed glucose. The results were correlated with those obtained by the prevous spectrophotometric method with excellent accuracy, indicating that the amperometric enzyme electrode can be another substitute for the previous spectrophotometric detectin of glucose released from immune lysis of liposme loaded with glucose.

摘要:
A mild and efficient two-spet procedure for conjugating rabbit Fab' and horseradish peroxidase using maleimide compounds was developed, and the conjugate obtained was evaluated as an analytical tool in enzyme immunoassay. The enzyme was treated with N-hydroxysuccinimide ester of N-(4-carboxyeyclogexylmethyl) maleimide to introduce maleimide groups. Then, the maleimideenzyme was allowed to react with thiol groups of Fab'. MOre than two thirds of Fab'and the enzyme used was converted to conjugates with a minfaml polymerization, and both activities of the enzyme and antibodies were well preserved in the conjugate. The conjugate Obtained by this method was more suitable for enzyme immunossay thatn polymerized conjugates prepared by the periodate method. The conjugation using N-hydroxysuccinimide ester of m-maleimidobenzoic acid provided similarly less polymerized preparations but was less efficient.

摘要:
A simple spectrophotometric method for the dectermination of D-glucosaminate with bacterial D-glaucosaminate dehydratase (EC 4.2.1.26) has been developed. The method is based on the enzymatic dehydration of D-gluco-saminatie and the spectrophotomeric determination of the product, 2-keto-3-deoxy-D-gluconate, as its semicarbazone. A linear relationship was established between the absorbance at 250 nm of semicarbasone and the amount of D-gluco-saminate (0.05 - 1.0 μmol). The method was applicable to the determination of D-Glucosaminate in bacterial crude extracts.

摘要:
The Homogeneous Coenzyme-immunoassay for quantitation of insulin in a solution has been developed. The method relies on the comptitive binding of free and coenzyme (NAD)-labeled insulin by insulin antibodies (guinea pig anti-porcine insulin). NAD-insulin conjugates were obtained by a covalent binding of NAD-N6-[N-(6-aminohexy1)acetamide] and insulin in the presence of water-soluble carbodiimide. The NAD-insulin conjugaties were cycled in the coenzyme regeneration system Which included horese liver alcoholdehydrogenase (AIH) and its coupled substrates-cyclohexanol and P-nitroson-N, N-dimethlaniline (NIMA)- and the cycling rate was used to measure the conjugate concentrations. the cycling was inhibited when the lignad (insulin molecule) was bound to the insulin antibodey. The unbound insulin-NAD conjugate could be measure in the presence of the protein-bound cojugate. The Sensitivity of the proposed method of insulin determination in limited by the value of the antigen-sntibody binding constant and permits and accurate measurement of concentrations up to about 10-7M.

摘要:
A single extraction procedure has been developed which allows simultaneous extraction of basic, neutral and weakly acidic drugs from the urine of greyhound racing dogs. The proccedure is rapid and uses small volumes of sample, allowing for the initial screening of up to 100 sample/day by on technician. over 60 drugs are detectavle with this procedure at concentrations as low as i ug/mL.

摘要:
A gas- iliquid-chromatographic method for the quanititive determination of authentic nadolol and nadoll in tablets (corgard p2) has been developed. Nadolol in tablets was extracted with absolute alconhol before injection into the gas chromatograph. The concentration range adopted varied between 1 mg to 8 mg/ml of nadolol in alcoholic solution and the accuracy obtained was 97.5±3.8.

摘要:
Primaquine reacts with diszo-p-nitroaniline in acid medium to give an orange yellow colour having and absorption maximum at 478 nm. On rendering the medium alkaline, a bathochromic ahift acompanied by hypochromic effect was revealed; the new maximum is located at 525 nm. The mean percentage recoveries for authentic samples amout to 100+, and 100.21+, 0.9 by the acid and alkaline procedures respectively, (p=0.05). Both methods could be applied to determine primaquine salts in pharmaceutical preparatins; the results obtained were in good agreement with those of the offictial method.
