摘要:
A high-performance liquid chromatographic procedure for the determination of pirenzepine dihydrochloride as a bulk material and in its tablet dosage form (GastrozepinR) is presented. Normal phase liquid chromatography has been performed on a Micro-pack Si-10 column using ammonium hydroxide (28–30% NH3) in methanol (0.75: 99.25% v/v) as mobile phase at a flow rate of 2 ml/min. Clobazam has been used as internal standard with retention times of 1.9 and 2.8 minutes for clobazam and pirenzepine dihydrochloride, respectively at 254 nm. Analytical calibration yields a linear relationship between 5 and 25 μg/ml, with correlation coefficient of 0.999. Tablets each labelled to contain 25 mg pirenzepine dihydrochloride give mean percentage found of 99.98 ± 0.4. A plot of logarithm of concentration against time for a solution in 6 N hydrochloric acid gives a straight line with a slope of - 0.197 day−1. The proposed method is, therefore, a stability indicating method.

Hexa-anminecobalt (III) tricarbonato cobaltate is used for direct determination of 1 to 100 mg L-ascorbic acid in synthetic standards and different pharmaceutical preparations. The proposed method depends upon visual titration of L-ascorbic acid in the presence of ferroin as indicator.

摘要:
Native low-temperature phosphorescence of mebendazole and flubendazole in ethanol is used for the determination of these imidazoles in anthelmintic preparations with wavelength maxima and detection limits of λEXC= 322 nm, λEM= 454 nm; 10 ng ml−1and λEXC= 325 nm, = λEM= 455 nm; 5 ng ml−1, respectively, with linear response up to 8 μg ml−1and 9 μg ml−1, respectively. The structural basis of these phenomena is discussed for both compounds and for related imidazoles and benzimidazoles. Apart from good sensitivity and excellent specificity offered by the technique, the use of cryogenic equipment (liquid nitrogen, special cuvettes, expensive dewar cells) implies some disadvantages for routine analyses.

摘要:
The use of derivative and difference-derivative spectrophotometry for the determination of certain drugs in oily injection is presented. The method is illustrated by the determination of oestradiol valerate, oestradiol dipropionate, oestradiol benzoate, testosterone propionate and progesterone in their oily injections. This method is simple, rapid and accurate. The results obtained are reasonably reproducible with a coefficient of variation less than 2%.

Benzimidazoles and benzoyl benzimidazoles show interesting native luminescence properties that can be used as additional methods for qualitative and quantitative work. A comparison is made between fluorescence emission in solution, fluorescence on filter paper, low temperature phosphorescence (77°K) and room temperature phosphorescence.

摘要:
A rapid quantitative gas-liquid chromatographic procedure for the determination of 2-diphenylmethoxy-N, N-dimethylethanamine (diphenhydramine, dimedrol) hydrochloride, in pure form and when it is dispensed in eye drops and lotion, is described. The antihistaminic drug requires no conversion to the free base, and the assayed pharmaceutical formulations need no sample cleanup, preliminary drug separation or derivatization. The method is adopted for a concentration range varied between1.0 - 10 mg.ml−1of the drug in ethanolic solutions. The accuracy obtained was 98.6 ± 0.31%, 93.3 ± 0.82%, and 87.9 ± 1.1% for the authentic diphenhydramine hydrochloride, its eye drops and its lotion. The 50% - addition of the drug to the labelled claim in the galenical forms, eye drops and lotion, gives average mean recoveries of 100.8 ± 0.74% and 97.9 ± 0.45% respectively.

摘要:
A simple and reliable procedure for the simultaneous determination of dopamine (DA), serotonin (5-HT) and their acidic metabolites 3,4-dihydroxyphenylacetic acid (DOPAC) and 5-hydroxy indoleacetic acid (5-HIAA) using high-performance liquid chromatography with electrochemical detection has been developed. Minimal sample preparation is required before injection into the liquid chromatograph. The present method can be applied to the analysis of several rat brain regions. The procedure offers good possibilities for routine analysis of biogenic amines and their acidic metabolites in the picogram range.

A simple, accurate, and specific -1H-NMR spectroscopic method is presented for the assay of succinylcholine chloride injections. After freeze-drying the sample solution, a mixture of the residue with acetamide, serving as the internal standard, is dissolved in deuterium oxide, and the spectrum of the solution is recorded. The quantity of drug in the dosage form is calculated from the integral values for the resonance signals at ca. 2.01 ppm (acetamide) and ca. 3.27 ppm (succinylcholine chloride). The mean ± SD recovery value from synthetic formulations was 99.93 ± 0.60% (n = 10), with a corresponding CV of 0.60%. Assay values for a group of 10 commercial samples ranged from 86.1 to 100.7 (mean = 98.01)% of declared. Injection additives such as methyl paraben and benzyl alcohol did not interfere with the assays. The proposed method will also permit the simultaneous monitoring of the hydrolytic degradation of succinylcholine to its ester components.

The determination of phosphates and polyphosphates has been effected using an hybrid electrode, which consists of a glucose oxydase enzymic membrane and a Solanum tuberosum tissue slice. This membrane, rich in acid phosphatase, catalyses the glucose-6-phosphate hydrolysis. This reaction is quantitatively inhibited by phosphate. Several factors involved in the electrode response, like substrate concentration, pH, ionic strength and type of buffer are discussed in detail. At a 4.10−4M glucose-6-phosphate concentration, the linear ranges of phosphates and polyphosphates are, respectively, 6.10−5M to 1.6.10−3M and 3.10−5M to 1.10−3M. The urinary phosphate contents determinated by this biosensor are in good agrement with those obtained by usual spectrophotometric techniques.

摘要:
A rapid and sensitive method for the quantitation of microgram antibodies immobilized on polystyrene is proposed. A procedure of protein staining on polystyrene with coomassie, brilliant blue G 250 follows Beer's law after release of the coloured complex. The lower limits of detection are determined.
