摘要:
A new bromometric assay procedure for penicillins was developed, using an organic brominating agent. 1,3-Dibromo, 5,5-dimethylhydantion (DBH). The experimental conditions, the molar ratio as well as the identification of the reaction products were thoroughly studied. The end point could be detected visually or potentiometrically. Spectrophotometric titration could also be performed. The suggested procedure could be successfully applied to pharmaceutical preparations containing penicillins. The percentage recoveries for different penicillins were satisfactory.

摘要:
A liquid chromatographic method for the determination of antiepileptic drugs in serum is described. The drugs were adsorbed on activated charcoal from which they were recovered by extraction with methylene chloride. The organic extracts were evaporated to dryness and the residues were dissolved in small volumes of the mobile phase. The extracts were clean. Only trace amounts of endogenic compounds were detected. The chromatographic separation was performed with a radially compressed column (C18) that was used daily for several months.

摘要:
A simple, specific, rapid and sensitive method for the analysis of mecillinam in plasma and urine using high pressure liquid chromatography is described. The assay is performed by direct injection of a plasma protein free supernatant or a dilution of urine. A μBondapak phenyl column with an eluting solvent of 16% CH3CN-0.2% H3PO4was used, with UV detection of the effluent at 220 nm. Desacetyl-cephalothin was used as the internal standard and quantitation was based on peak height ratio of mecillinam to that of the internal standard. The lowest concentration detectable without extraction was 0.25 μg/ml for plasma and 8.9 μg/ml for urine. No interference from plasma and urine was noted.

摘要:
Spectrophotometric analysis presents very often the disadvantage of interferences by substances which can absorb near the substance of interest. The use of derivative spectra allows to overcome this difficulty. In this paper we describe a method for determination of bilirubin in amniotic fluid based on use of derivative spectrophotometry. This method presents some advantages of accuracy and quickness in comparison with the ones normally used.

摘要:
Conductimetric detection of bile acids in reversed phase high-performance liquid chromatography is described. The solvent system of the mixture of water and organic solvent containing small amount of basic salts such as ammonium carbonate is found useable by removing the cation with the cation exchange column inserted between the ODS column and the conductance detector. Thus, a few ng of tauro-and glyco-conjugated bile acids can be detected without tedious derivatization and hydrolysis.

摘要:
A spectrophotometric method for determination of Vitamin B12(Cyanocobalamin) as cobalt in pharmaceutical preparations is described. The sample is first decomposed by concentrated sulfuric and nitric acids and the dry residue is solubilized in distilled water. The cobalt present is then spectrophotometrically determined at Λ= 317 nm as Co-HMPA-SCN complex. The complex is stable for a long time in the range of pH 3–10. The presence of many other metal cations does not interfere.

摘要:
Using a CL-amylose column it is possible to purify α-amylase from other protein contaminants (e.g. albumin, haemoglobin, etc.) and from interfering exoamylases, by its specific retention on CL-amylose. α-Amylase is eluted off the column in the second step by a mixture containing 0.1 M NaCl in acetic acid 0.01 M and CaCl20.015 M. The recoveries of glucoamylase and α-amylase were 74% and 65% respectively. The retention capacity of the stationary phase is 0.5 mg of α-amylase/ml bed volume.

Simple one tube method for the determination of steroid sulfatase activity in an enzyme preparation or commercially available preparation is described. Principle of the reaction is as follows; dehydroepi-androsterone sulfate (DHEA-S) as substrate is hydrolyzed by the catalysis of sulfatase and DHEA released is determined by the enzymic method using 3β-hydroxysteroid oxidase.

摘要:
The constituents of oil droplets in clove (bud ofEugenia caryophyllata) cell and crystals of berberine alkaloids in bark cell inPhellodendron amurensewere transfered into a microcapillary glass tube attached to a micromanipulator apparatus and analysed by HPLC. The method makes it possible to identify components in cells as long as they are in an oily or crystalline state. This new ultramicro-technique is entitled “Histochemical Chromatography”.
