摘要:
Fluorescamine has been shown to be a promising derivatizing reagent for the room temperature phosphorescence analysis of primary amino acids. The fluorescamine concentration used for derivatization in solution was 600 μg/ml and for direct derivatization, 1160 μg/ml. For derivatized primary amino acids the limits of detection were found to be between 0.8 ng and 2.9 ng with linear dynamic ranges between 2 and 3 orders of magnitude.

摘要:
The first use of perdeuterated,15N-Tempamine spin label to selectively monitor cell surface sialic acid of human erythrocyte membranes is reported. The molecular reasons for significantly increased sensitivity and significantly decreased scatter in the data over that of protonated,14N-Tempamine are discussed. These advantages are illustrated by monitoring the effects of wheat-germ agglutinin on the motion of terminal sialic acid of membrane glycoconjugates.

Intracellular components, which are picked up with the help of micromanipulator unit of histochemical chromatography are subjected to mass fragmentography, by which a biosynthetic study of alkaloidal components was carried out and it was found that cis-N-methyl-tet-rahydroberberinium salt is incorporated in the formation of allocryptopine inMacleaya cordataroots so far as cytochemical point of view is concerned.

摘要:
This paper describes a rapid enzymic procedure, based on calorimetry, for specific determination of oxalic acid. Oxalate is oxidized by immobilized oxalate oxidase to hydrogen peroxide and carbon dioxide. The heat generated by this reaction is measured in a calorimetric device, the enzyme thermistor. Oxalate concentrations as low as 0.02 mM can be determined. Also described is purification and immobilization of the enzyme, as well as the effect of some of its inhibitors. The urinary oxalate content of 16 persons was determined using the enzyme thermistor. For samples containing a very low concentration of oxalate (less than 0.2 mM) an extraction step with tributylphosphate can be introduced to purify and concentrate the oxalate. The oxalate content in different kinds of food was also determined.

摘要:
A method is described for the rapid and simple preparation of acetoacetic acid in its salt form. The preparation is based on the saponification of ethyl acetoacetate or on the alkaline hydrol sis of acetoace-tamide. The most successful results, in terms of purity and yield, are achieved using ethyl acetoacetate and 5N NaOH as the saponification medium and incubating for 12 hr at 4 °C with no pH adjustment.

摘要:
A biosensor using permeabilized yeasts (Hansenula anomala) is described for the determination of L-lactate. The same electrode can be used either potentiometrically or amperometrically. The linear ranges are respectively of 4.10−5M to 2.10−3M for potentiometry and 8.10−7M to 3.10−3M for amperometry with a reproducibility and a precision of ± 2 to 3%. The parameters involved in the optimization of electrode response, such as pH, temperature, cofactor concentration and ionic strength of the buffers, are discussed in details for both types of measurements. The amperometric technique is suitable for lactate determinations in biological media. In this case, differential measurements are used in order to eliminate interferences of biological redox reactions.

A method for effective separation of phospholipids and neutral lipids of E. Coli in one dimensional thin layer chromatography on ready-made plates has been proposed. The chromatographing was made successively in three different solvent systems. About ten lipid components were shown to be separated on the same plate.

摘要:
A selective spectrophotometric method for the determination of oxyphenbutazone is described. The method is based on nitrosation reaction and simultaneous formation of copper(II) or cobalt (II) chelate of the nitrosoderivative. The formed chelates are extractable with organic solvents giving yellow solutions whose absorbance are proportional to the concentration of oxyphenbutazone. In addition, the chelate extracts, upon treatment with diethyldithiocarbomate develop an additional indirect method of high selectivity for determining oxyphenbutazone. The developed methods are highly accurate and comparable with an official method.

摘要:
A rapid and simple method for the determination of cephalexin, cephalothin sodium and cephradin without prior separation from their alkali-induced degradation products is presented. By measuring the values of the first and second derivative spectra at certain wavelengths, the concentration of the intact drug can be calculated directly without interference of degradation products. The method was proved using synthetic mixtures of the intact drugs with their degradation products, and its suitability to monitor the stability of the drugs was demonstrated.
