The adsorption of CO and CO2on Ru/RVC and Ru/Pt electrodes has been studied by a potentiodynamic technique. It was shown that there are some differences between CO adsorption products on these two kinds of electrodes.CO2adsorption on Ru electrode was not observed.

An enzyme membrane electrode usable for the assay of oxalate in foodstuffs is described. A commercially available preactivated polyamide membrane was used for the immobilization of oxalate oxidase. The bioactive disk thus obtained was associated with an amperometric transducer. The resulting self-contained enzyme electrode wich allows oxalate determination in various materials with minimal pretreatment exhibits a linear calibration ranging from 10–7 M and 10–4 M in the cell. The response-time was comprised between 20 seconds and 1 minute, depending on the oxalate content in the sample. The electrode-response was very stable for at least 4 months, a period during which more than 150 assays were performed.

Three methods for determination of dissolved oxygen in samples of natural water are compared; patentiometric stripping analysis, PSA compares well with oxygen selective electrodes. Although potentiometric stripping analysis and oxygen selective electrode methods are found to be simple, rapid and of higher reproducibility than the usual Dinkier procedure, the use of oxygen selective electrodes has many disadvantages.

The formation and extraction of copper (II) in the form of benzyl bismuthiol and naphthyl bismuthiol chelates has been studied. The reagents' solvation in butanone is investigated and optimum conditions for extraction are established. Both chelates have been studied in solution and in solid form. After studying the influence of 14 possible interferences, an atomic absorption spectrometric determination of copper using both reagents was carried out in tap water samples.

Differential scanning calorimetry (DSC) is employed to evaluate the relative proportions and strength of acid-sites in gammaalumina and amorphous silica-alumina through presorption of tri-ethylamine (TEA) followed by its thermal desorption in the DSC equipment. From the data obtained, total acid-sites in silica-alumina is found to be 2.3 times as large as those present in gamma-alumina, and 5–5% of total acidity in alumina is of Bronsted type whereas in silica-alumina this type comprises 28.9%' The Bronsted acidity strength in alumina and silica-alumina is identical although greatly different in quantity. Lewis acidity in silica-alumina is significantly stronger than that in alumina.

Four staining methods for aspartate aminotransferase (AAT) detection after electrophoresis have been reported. Comparison of the minimum AAT activity detected by these methods showed the following sequence: Sakakibara et al.> Rej and Herder > Decker and Rau > Banks et al. Addition of ADP and diaphorase to the stain of Banks increased its sensitivity 10 fold. The presence of lactate dehydrogenase (LDH > activity in the samples gave electrophoretical artifacts when the methods of Banks et al. and Rej & HoSrder were used. These artifacts were due to the nothing dehydrogenase (NoDH) activity of LDH as demonstrated by chromatography on 5′ AMP-Sepharose 4B (LDH affinity support).

A small scale sandwich enzyme immunoassay for macromolecular antigens using β-D-galactosidase fromEscherichia coliand horseradish peroxidase as labels is described. An antibody IgG-coated glass ball of 1.0 mm in diameter was incubated with antigen in 5 μl and subsequently with affinity-purified Fab'-enzyme conjugate in 5 μl. Using B-D-galactosidase, the detection limits of ferritin, IgE, a-fetoprotein and chorionic gonadotropin were 0.001 amol (600 molecules), 0.03 amol, 0.03 amol and 0.07 amol, respectively. Using peroxidase, the detection limits of ferritin, α-fetoprotein and chorionic gonadotropin were 0.005 amol, 0.1 amol and 0.2 amol, respectively. These detection limits were 3 to 30-fold less than those obtained by the previous sandwich enzyme imnunoassays using antibody IgG-coated polystyrene balls of 3.2 mn in diameter in 150 μl.

A gradient elution HPLC method for analysis of propranolol and three basic metabolites in bile is presented. Before or after hydrolysis with beta-glucuronidase, acetonitrile is added to the bile sample, the resulting mixture is centrifuged at high speed, a portion of the supernatant is diluted with buffer and an aliquot is injected onto the HPLC column. The method utilizes gradient elution, reverse phase chromatography with fluorescence detection, and is sufficiently sensitive to quantitate 1.25 mcg/ml of compound using 50 μl to 100 μl of bile sample.

An HPLC method for the analysis of nitrofurantoin (NTR) in human plasma has been developed and evaluated. This method is based on a selective extraction and reverse-phase chromatography which results in chromatograms with 2400 to 2500 theoretical plates for NTR and the internal standard (furazolidone). The method is highly reproducible and has been validated over the range of 5 to 4000 ng/ml.
