We have succeeded in completely bleaching a humic acid, extracted from peats and carefully purified, by the action of the peroxoacids, followed by a chlorhydric or enzymic hydrolysis. The degradation products characterized by chromatographic analysis on paper, and by GLC after silylation, contain 3 reducing sugars (glucose, rhamnose, arabinose), a disaccharide and 8 amino acids among which are lysine, serine and tyrosine. The reducing sugar content of the mixture (expressed in glucose) corresponds to a yield of 65% relative to the initial product.

Combining a simple chromatographic system on celite microcolumns with partially specific antisera as binding reagents, simultaneous radioimmunoassay of progesterone, 17-hydroxyprogesterone, and estradiol-17B could be performed on the same aliquot of plasma. These steroids have been measured in plasma samples obtained from subjects of both sexes under various conditions.

Lanthanide and actinide impurities in a total sample of 1 to 10 μg of transuranic matrix can be determined by employing the spark-source mass spectrograph and solution technique. A special source glove box permits safe manipulation of transuranium elements with α-emitting radioactivity up to 1012dpm. The minimum detection limit is about 1012atoms per electrode system and an exposure of 10−8coulomb. Relative sensitivity values for the lanthanides and actinides were established with respect to erbium, the internal standard.

A probe-type sample changer has been added to a spark-source mass spectrometer enabling the rapid determination of ppb levels of mercury by isotope dilution. The technique has also been used for determining Cd, Hg, and Zr in samples of air, water, and urine.

Oxygen (5–100%) in gas mixtures is determined by measuring the absorbance of its charge-transfer complex with dimethylaniline at 347 nm. The method is essentially specific, and merely requires bubbling the gas through the reagent, stabilized with EDTA. Oxygen is simply removed from the reagent by purging with nitrogen or carbon dioxide.

The complexation between Pb(II) and tartrate has been used to experimentally ascertain the effect of precision in the ΔE1/2measurement and ligand concentration range upon the data analysis of the polarographic method of studying metal complex equilibria. The need to obtain data at the lowest ligand concentration possible and with ΔE1/2≤ 0.4–0.6 mV was clearly shown. If this precision is unachievable, data from some other technique, based upon a different property, may be required. These experimental results are in agreement with the simulation study of Klatt and Rouseff (Anal. Chem.,42, 1234 (1970)).

The potentials of desorption of various alkyl alcohols have been studied as a function of concentration of alcohol and of the nature and concentration of various electrolytes. Specific adsorption of the electrolyte ions is reflected in shifts of the desorption peaks of the alcohols. At anodic potentials, the order of specific adsorption appears to be chloride > sulfate >perchlorate > fluoride, and at cathodic potentials (vs. S. C. E.), chloride > perchlorate > sulfate > fluoride.

Hydrobromic acid in glacial acetic acid hydrolyses the N-nitroso derivatives of secondary amines and amides to produce nitrite almost quantitatively. Other compounds containing a N-O bond such as N-nitramines and N-nitramides do not react similarly under the conditions employed.
