A new colorimetric procedure for blood alcohol determination is described. Blood proteins are precipitated, and the protein-free supernatant incubated with nicotinamide-adenine dinucleotide (NAD), alcohol dehydrogenase (ADH), nitro blue tetrazolium (nitro BT) and phenazine methosulphate (PMS). Ethanol in the sample reduces the NAD, and the reduced NAD so formed reduces nitro BT to a coloured formazan, PMS serving as an intermediate electron carrier. The reaction is allowed to proceed to completion and colour formation quantitatively relates to sample alcohol concentration. An ethanol standard of known concentration is included with each batch of determinations and sample alcohol concentration calculated from this. The method permits the examination of large numbers of samples with rapidity and precision and without the need for specialized apparatus. Comparison with an ultra-violet enzymatic procedure for blood alcohol determination, gave excellent agreement.

A quantitative method has been developed for the determination of the tricyclic antidepressant drug nortriptyline in plasma, utilizing a combined technique of gas chromatography - mass spectrometry called mass fragmentography. A metabolite, desmethylnortriptyline, may be analyzed in the same run. The compounds are first converted to their heptafluorobutyranide derivatives. A complete chromatogram takes less than five minutes. Quantitive determinations under the present conditions are possible down to a few nanograms of nortriptyline per milliliter. There is a good correlation between the new method and that previously used which was based uponin vitro311-acetylation labelling.

Three procedure for 17-ketosteroids are compared: The method of Callow et al., the method of Drekter et al. and the method of Corker et al. Good correlation has been found between results obtained by the methods of Callow and Corker. Analyses of single analytical steps showed that Drekter's method gives in some cases too high results because non-specific chromogens are included when readings are taken only at 520 nm. Of these three procedures that of Corker et al. is proposed as the method of choice.

A radio-frequency probe and a synchronized electronic ion-beam chapper have been used to determine ion-intensity variation with spark duration. Large peaks in multiply charged ion intensities are detected in the early stages of the radio-frequency spark, while later in the spark period the singly charged species predominate. The effects of these variations on the precision of analysis have been determined. A gate was selected in the spark pulse, and a factor of two increase in precision was obtained.

Error propagation in linear titration methods are considered in terms of the precision in measurement of the slopes and intercepts of the titration curve.
