摘要:
A rapid second-derivative spectrophotometric assay procedure has been described for the determination of some benzenoid drugs in pharmaceutical dosage forms. Spectral interference from formulation excipients in simple uv spectrophotometric methods has been eliminated by the application of the proposed method. The different assay parameters, linearity and precision of the method have been assessed. The assay results obtained for eight batches of the pharmaceutical formulations of these drugs are in accord with the declared amounts. The high specificity, ease and simplicity offered by second-derivative spectrophotometry permit unit dose assay of these formulations.

摘要:
GLC and colorimetric methods for Capsaicin and its dosage forms are investigated. They offer an improvement in ease, speed and accuracy. The first GLC method is based on the formation of methyl derivative of capsaicin. The colorimetric procedure is based on the formation of O-nitroso-derivative between the drug and sodium nitrite.

摘要:
The complexation of ferron(7-iodo-8-hydroxy quinoline-5-sulphonic acid) with some metal ions of biological interest, such as Ag(I), Cu(II), Zr(IV), U(VI), Au(III), Ni(II), Zn(II), Mn(II), Fe(II), Fe(III), and Cr(III), has been studied potentiometrically using the Calvin-Bjerrum titration technique as employed by Irving and Rossotti. The protonation constant of the ligand and the stability constants of the respective metal complexes have been determined at constant temperature (25°C) and ionic strength (0.1M KNO3). Complexes of ferron with some metal ions have also been prepared and analyzed by ir. spectral data.

A sensitive spectrophotometric method, which directly measures the formation of pyruvic oxime from pyruvate and NH2OH in the UV, is described for the detection of pyruvic oxime hydrolase activity in biochemical systems. The method was used in an attempt to detect pyruvic oxime hydrolase activity in cell-free extracts from anAlcaligenessp. which can grow on pyruvic oxime. Although substantial pyruvic oxime + nitrite oxidative activity was detected in cells and cell-free extract of this bacterium, catalysis of pyruvic oxime formation was not observed within the error of the method (∼20% of the uncatalyzed rate constant of 1.9x10−3s−1at pH 7, 22°C). The rate of pyruvic oxime oxidation by cells and cell-free extract was at least 105−106times greater than the rate of its hydrolysis, thus implying that oxidation of pyruvic oxime need not require prior hydrolysis. The method would appear to be applicable to hydrolases directed toward a variety of oximes.

摘要:
The sera of seventy-one women and twenty-six men of various ages were examined by a two dimensional (crossed) immunoelectrophoresis technique which was modified to detect a protein (the Down's Syndrome protein) originally found in the sera of mothers of trisomy 21 children. The sensitivity of the method is reported to be 0.1 to 1.0 mg/L. Modifications included use of a Tris-Tricine buffer, high voltage in the second dimension, and relatively low antibody concentration to allow easy detection of the Down's Syndrome (DS) protein.

摘要:
A derivatization procedure was developed for the fluorometric determination of purified xanthomegnin. A highly fluorescent product is formed by reaction of xanthomegnin with ammonia and hydrogen peroxide under defined conditions. The derivative is a light brown compound that can easily be dissolved in water or polar organic solvents. Its excitation and emission wavelengths in methanol are 340 and 445 nm, respectively. As little as 0.1 ng of xanthomegnin can be detected using high pressure liquid chromatography in a reverse phase system with fluorescence detection.

摘要:
Fibronect in was shown to bind to solid-phase C1q and to inhibit the binding of aggregated lgG in an C1q-solid-phase enzyme immunoassay in the absence of human serum. In the presence of human serum the determination of aggregated lgG and circulating immune complexes is not influenced by fibronectin in this assay. Therefore, we suggest that fibronectin does not affect the determination of immune complexes in serum specimens by C1q binding assays.

摘要:
The use of cyanide generating substances for medical, home and business applications has necessitated the need for measuring sublethal levels of this substance because of the potential of inadvertent cyanide poisoning. The method we propose provides a means for more accurate recovery at an increased rate of reaction there by making its sensitivity more conducive to the measurement of minute quantities of cyanide.

A flow injection manifold containing a dialyzer and reactors with immobilized galactose oxidase and peroxidase was used for the determination of galactose in urine, lactose in milk and dihydroxyacetone in a biotechnological reaction medium. The hydrogen peroxide which is formed by the galactose oxidase reaction was detected by amperometric reduction of a mediator. The latter had been produced from hydrogen peroxide in a peroxidase catalyzed reaction. The hydrogen peroxide detection step was studied with several mediators and hexacyanoferrate (II) was selected. An ion exchange HPLC procedure was used to purify the galactose oxidase, in particular from catalase, and the kinetics and the selectivity of a reactor containing the immobilized enzyme was investigated. Columns for removal of certain interferents such as ascorbic acid were used in the determination of galactose in urine. The response to galactose standards was linear from the detection limit of 2 μM to 60 mM. The throughput was 45 samples per hour and the relative standard deviation 0.4%.

摘要:
A low-resolution method for simultaneous, rapid determination of radiolabeled glyburide and its metabolites in human plasma is described. Plasma samples were extracted with ethyl acetate. Extracts were redissolved in 300 μl of mobile phase, and injected into a 3 cm guard column, which was incorporated as a loop in a six-port switching valve. C14-glyburide was collected as a single 4-min fraction at a flow rate of 4 ml per min.
