摘要:
A continuous flow bioluminescent method for NADH analysis has been developed using nylon 6 as a solid support. Bacterial luciferase and NAD:FMN oxidoreductase are covalently co-immobilized to a nylon coil (1 m × 1.0 mm ID). The reactor (nylon coil) is placed in front of a photomultiplier tube inside a luminometer linked to an air segmented continuous flow system. Response is linear from 1 to 2500 pmol/tube allowing determination of NADH at picomolar levels. Inter and intra assay precision of the method is satisfactory (5-10%). Range of sample volume injected is 5-100 μl. More than 20 samples per hour can be analyzed and no carryover was observed. The stability of the nylon immobilized enzymes is high (over 2 months) and over 500 samples can be analyzed with a few mg of enzymes.

摘要:
An amperometric technique has been developed for the determination of enzyme activity by the detection of the decrease in the concentration of NADH used as the co-factor of the enzyme reaction. A glassy carbon electrode modified by adsorption of Mg++and NADH is employed for the measurement of the anodic peak current corresponding to the oxidation of NADH.

摘要:
A novel optical immunoassay system was developed and tested using human IgG as a model antigen. Following adsorption of antiserum to the surface of an optical waveguide, the immobilised antibody was then reacted with a solution containing antigen. The reaction was detected utilising the evanescent wave component of a light beam totally internally reflected within the waveguide. The growing antigeh-antibody layer resulted in an increase of scattered light which was monitored kinetically. Individual measurements were completed within 5 minutes and the resultant dose response curve had a sensitivity limit of approximately 10 nmol/L.

摘要:
A rapid, simple and reproducible method for sample pretreatment and analysis of disopyramide and its mono-N-dealkylated metabolite in serum is described. Samples for analysis were prepared by using Sep-Pak silica cartridge treated with 1 N sodium hydroxide followed by elution in 8 ml ethyl acetate. The eluate was back-extracted with acidic solution and an aliquot of the solution was injected directly onto the column.

摘要:
The method depends upon reducing papaverine with Zn/HC1 to a secondary base which could interact with fluoranil giving a charge-transfer complex with two prominent absorption maxima (350 and 530 nm). The method suits control analysis of papaverine-containing pharmaceuticals. The measured color is relatively stable, obeyed Beer's law (20-200 μg/ml° and showed % average recovery of 98.88 + 0.9.

摘要:
The well known Hurexide reaction for purine bases was modified and optimized for the quantification of caffeine in crude drugs, refreshing drinks and pharmaceuticals. The method was also applied for the detection and assay of caffeine in mixtures of illegal opiate samples. It was also applied to assay theobromine and theophylline alongside with caffeine in their natural and synthretic mixtures after a simple separating procedure depending on their different basicities.
