Trace amounts of molybdenum catalyze the reduction of selenium (IV) to the element by tin (II) in hydrochloric acid medium. The absorbance of the red colloidal selenium is read at 390 nm. Potential interferences are avoided by separating the molybdenum by solvent extraction. Physiological concentrations of molybdenum are determined in 10 ml. of ashed human blood or plasma and 1 ml. of ashed human urine.

Both primary and secondary amines may be readily labelled with the 9-fluorenylmethoxy carbonyl group by the reaction with 9-fluorenylmethyl chloroformate (FMOCCI). The reaction proceeds rapidly under alkaline aqueous conditions, allows for simple elimination of excess reagent by partitioning, improves chroatographic resolution of amino acid-like compounds by anion exchange liquid chromatography and introduces a fluorophore with a large quantum yield which is essentially unaffected by solvent-solute interactions.

A system for the determination of urinary polyamines by ion exchange chromatography with fluorescence detection is presented. It provides a sensitive and specific assay of putrescine, cadaverine, spermidine and spermine in urine hydrolyzates. An internal standard is used, which makes the automated determination of series of samples easy.

Optimum reaction conditions by pH-stat studies of lipase fromcandida cylindraceatowards triolein emulsions were determined. These conditions are no less than 40 mMcalcium ion and no more than 0.3Msodium chloride in 10 mM (tris-hydroxymethyl) amino methane (THAM) buffer at pH 7.45. Comparison of the enzyme activity and relative amount of product formed as a function of calcium ion concentration imply a thermodynamic as well as a kinetic role for calcium. Increasing the THAM concentration to 0.5Mappears to facilitate the deactivation of the enzyme by the free fatty acid produced in the reaction.

A recently described reaction between barbituric acid and biliverdin has been investigated for suitability as a new approach to the colorimetric estimation of total bilirubin concentrations in hyman sera. The 6eaction was found to give absobance values which correlated well with bilirubin concentrations determined by an automated diazotisation method. As a manual technique, the sensitivity was greater than most methods based upon diazotisation. The reaction is likely to be of particular value when a method is required to estimate low levels of serum bilirubin.

A simple method for the preparation of N-ethyl derivatives of secondary amino tricyclic antidepressants is described, using dimethylformamide diethyl acetal as the alkylating reagent. The GC data and electron impact and chemical ionization mass spectra of N-ethyl derivatives are presented. The use of these derivatives in the quantitation of plasma levels of these drugs in clinical studies is discussed.
