摘要:
An electron microscopic study of the intraoral lesions of Darier's disease demonstrated a defect of desmosomes and tonofilaments in the early development of affected mucosa. Marked cytoplasmic vacuolation, keratohyaline granule formation and nuclear pyknosis as reported in skin acantholytic cell ultrastructure was not conspicuous in the oral mucosa of the case reported. The clinical diagnosis, histopathologic diagnosis, pedigree and treatment of Darier's disease is discussed.

摘要:
In Part A, the techniques most commonly used to study epithelial cell kinetics and their suitability for use in man are reviewed. In Part B, the application of such methods to comparative studies of normal and abnormal oral epithelia of both man and experimental animals is examined. Claims regarding the possible prognostic value of LI's or MI's of biopsies of potentially or overtly malignant human oral lesions are also considered. Normal and abnormal kinetic data for other lining epithelia, e.g., skin, uterine cervix and gastrointestinal mucosa arc assessed where they provide a better illustration of some of the problems arising in such comparative investigations. Finally, the present and likely future areas in which cell kinetic studies may provide a guide to expected tumour prognosis and optimum treatment are summarized.

This paper is concerned with the relationship between eosinophilic material (EM) and amyloid‐like material in adenomatoid odontogenic tumors. In duct‐like structures and between opposing rows of tall columnar cells, EM did not stain for amyloid. Under electron microscopy, EM was composed of fibrillar and granular materials, and the fibrillar material was not amyloid. Two different kinds of EM were found in solid cell masses. Lesions from cases 2, 3, 4 and part of case 1 contained small droplet‐shaped EM and these EM did not stain for amyloid. Case 1 also contained EM that stained positively for amyloid. The structure of amyloid positive EM resembled developing enamel of human tooth germs. This material was tubular and finely granular. The tubular material resembled enamel matrix fibers rather than amyloid and the fine granular material was stippled. A new human epithelial cell line (MK—1) has been established from a well differentiated squamous cell carcinoma of the gingiva. It has been subcultured 90 times during the past 23 months. The population doubling time of the MK—1 cell line was 28 h. A colony forming ability with 18% plating efficiency was shown. Chromosomal analysis revealed an aneuploid human male karyotype with 22 marker chromosomes. The modal chromosome number was 75. Scanning electron micrographs of MK—1 cells showed numerous microvilli and intercellular bridges. Transmission electron micrographs of MK—1 cells showed epithelial characteristics with desmosomes, tonofilaments and membrane‐coating granules. Hetero‐transplantation of cultured MK—1 cells produced tumors, the histology of which was similar to that of original ones. The ultrastructure of the tumors showed the presence of abundant tonofibrils and keratohyalin granules. This MK—1 cell line is a model useful for the study of keratinization of squamous

In this study, plaque‐like lesions, epithelial hyperplasias, and papillary invasive carcinomas were produced in the tongues of hamsters by the application of DMBA combined with trauma for 16 weeks. The junction between the neoplastic epithelial cells and the connective tissue was studied ultrastructurally. In plaque‐like lesions, the basal lamina was partially decreased in both thickness and density or partially discontinuous. In epithelial hyperplasias, cytoplasmic projections were observed in the portion of the basal cell near the intercellular space, and the basal lamina was absent around these cytoplasmic projections. In papillary invasive carcinomas, however, numerous cytoplasmic projections extended from various portions of the basal surface of the basal cells into the underlying connective tissue. The basal lamina was also absent around these projections. In epithelial hyperplasias and papillary invasive carcinomas, some of the cytoplasmic projections appeared almost empty, some contained a small number of tonofilaments and ribosomes, and others were filled with a large number of ribosomes but did not contain the tonofilaments. Initial calcification in healing amputated tooth pulps was examined in a transmission electron microscope (TEM). The process is characterized by an abundance of extracellular matrix vesicles (0.1–0.2 μm in diameter) distributed between forming cells and the surface of the amputated pulp. Osmiophilic material and needle‐like crystals within the vesicles was followed by disappearance of the vesicular membrane and accumlation of crystals that aggregate into calcified fronts. Ca and P were detected in these needle‐like crystals by an electron probe X‐ray microanalyzer. This finding suggests that the crystals were produced during the calcifying process. The healing of amputated tooth pulps is essentially similar to initial clarification events that occur in other normal and pathologic calcified
