The tumor promoting effects of snuff was studied in Lewis rats initiated with 4‐nitroquinoline‐N‐oxide (4‐NQO) and Sprague Dawley rats repeatedly inoculated with herpes simplex virus type 1 (HSV‐1). The test substances were administered in a surgically created canal in the lower lips of the rats. There were 15 rats in each test group and 10 rats in the control group. In the groups treated with 4‐NQO and 4‐NQO + snuff, 8 and 12 tumors (5 and 9 malignant) were found, respectively. In the group subjected to HSV‐1 only, 3 tumors were found (2 malignant), in the group subjected to snuff only, 4 tumors were found (3 malignant) and in the group subjected to the combination of HSV‐1 and snuff, 13 tumors were found (7 malignant). In the control group only one malignancy was found. The study did not show any promoting effects of snuff in the oral cavity after initiation with 4‐NQO. Neither was there any increase in the number of oral tumors in rats treated with HSV‐1 and snuff. However, there was a marked increase in the number of malignant tumors outside the oral cavity in the group treated with HSV‐1 and snuff, underlining the importance of interactions between these two agents in the developme

A recent report has demonstrated that the avidin‐biotin‐peroxidase (ABP) technique can successfully be used on previously stained histologic sections without the loss of sensitivity. In this study the tissues had been microwave fixed and only H&E stained sections were used. This procedure was tested on formalin‐fixed tissues previously stained with H&E, PAS and van Gieson. Stained sections were prepared from selected blocks used as controls. One week after staining the sections were decolorized and together with the unstained control sections were restained with the ABP technique using IgA, IgD, IgE, IgM, kappa and lambda light chains, S100, EMA, LCA, amylase and PCK antisera. Demonstration of the tissue antigens was best with the previously stained H&E slides with little or no loss in sensitivity. Normal human oral (cheek) mucosa was studied to discover whether the oral cavity resembles the Mucosal Immune System (MIS) or the Skin Immune System (SIS). Immunophenotypes of lymphocyte subsets and Langerhans cells (LC) with their exact locations in the epithelium and papillary layer of the normal buccal mucosa were determined and compared with data of normal human skin. In a double staining procedure, the distribution of T‐lymphocytes in relation to blood and lymph vessels was determined. Immunophenotyping of LC was done with a CD la monoclonal antibody. In contrast to the skin, T‐lymphocytes in buccal mucosa are not primarily perivascular in location. They are more or less randomly distributed on both sides of the basement membrane. The epithelium of the buccal mucosa contains about 37 times as many T‐lymphocytes as the epidermis of normal skin. T‐cell numbers in the papillary layer are more or less comparable. The CD4/CD8 ratios of about 1/2 in the epithelium of buccal mucosa and 1/4 in the skin indicates preferential presence of the CDS subset in both sites, but the helper/inducer T‐lymphocytes play a much greater role in the epithelium of the buccal mucosa when compared with skin. B‐lymphocytes were not found in the epithelium and papillary layer of the buccal mucosa. Thus, immune response associated cells in buccal mucosa do not show the MIS pattern since B cells are absent. It has more in common with SIS but differences are also apparent. In the epithelium of the buccal mucosa the density of LC does not differ significantly from that of the skin, but the papillary layer of the buccal mucosa contains significantly fewer LC than the skin. Four cases of lateral cervical cysts were reported. All cyst walls were lined with keratinized stratified squamous epithelium, and contained lymph follicles with well‐defined germinal centers. Ectopic salivary gland tissue was found in one case. Histochemical examination by the immunoperoxidase method revealed the existence of carcinoembryonic antigen (CEA). However, alpha‐feto protein (AFP) and amylase were not detected in any of the cysts. The cyst fluid contained high levels of CEA and high titers of salivary type amylase, but AFP was negative. These results do not support any one of the theories concerning the etiology of cervical lymphoepithelial cysts. An epithelial cell line, RSMTx, has been established from the submandibular gland of weanling Fisher 344 rats by treatment of explanted tissue clumps with 3‐methylcholanthrene. These cells exhibit a polygonal shape on light microscopy and a polar appearance, with desmosomes, terminal bar‐like structures, surface microvilli and cytoplasmic interdigitations, when examined by electron microscopy. The cells react positively with an antiserum to cytoskeletal keratin, and a commercial monoclonal antibody to an “epithelial membrane antigen.” An antiserum, prepared against early passage cells in hamsters, reacts primarily with ductal elements in tissue sections of submandibular gland, as does an antiserum prepared in mice with late passage cells. In order to facilitate studies on human salivary glands, a method was developed for the culture of minor salivary gland duct cells from tissues obtained from oral surgery protocols. Minor salivary glands were isolated from such tissues, and a serum‐free growth medium was developed which supported the growth of the ductal component of these glands. The ductal origin of these cells was confirmed through immunohistochemical localization of replicating nuclei through incorporation of BrdU. The presence of epidermal keratin in replicating cells and the absence of smooth muscle myosin further substantiated the ductal origin of cells. Using normal growth mediun calcium concentrations (1.05 mM), these cells produced a keratinized multilayer of cells unable to undergo routine subculture procedures. A reduction in calcium ion concentration to 0.1 mM resulted in a cell monolayer, without evidence of terminal keratinization, which could undergo at least eight serial passages (1:3 ratio) under cell culture conditions. Vervet monkey attached gingiva and alveolar mucosa was used to investigate the effect of primary fixative composition and osmolarity on the scanning electron microscope appearance and epithelial cell surface feature density. Primary fixation was obtained using 12 different fixatives with osmolarities varying between 320–2010mOsm followed by further standard SEM processing procedures. All primary fixatives investigated produced acceptably fixed oral epithelium for SEM study, showing all the morphologic features characteristic of either keratinized or non‐keratinized oral tissue. Point counting revealed that the density of microvilli of attached gingiva epithelial cells when fixed at 2010mOsm was 72±8% of the cell surface area. This decreased to 40±5% when fixed at 320mOsm. Similarly the microplication density of the alveolar mucosa epithelial cells decreased from 70±5% at 2010mOsm to 43±7% at 320mOsm. Both these differences proved to be highly sign

The present paper describes the trend in incidence of oral and pharyngeal carcinomas in Denmark in the period 1943–1985, and provides an illustration of the effect of birth cohort on the overall trend. The results are discussed in relation to changes in consumption of alcoholic beverages and tobacco, and to changes in dietary habits in this century. This study examined the reaction of the local vasculature of the oral mucosa in 16 Sprague Dawley rats receiving systemic nicotine delivered (1.5 mg/kg/day) via subcutaneous minipumps for 24 h or 2wk. Control animals received saline. After treatment animals were killed and biopsies taken from palate, maxillary gingiva and buccal mucosa, frozen and cryostat sections incubated to demonstrate alkaline phosphatase, which is a capillary marker. The total length of the capillary fragments in the nicotine treated groups was significantly less than of the control group. There was also a decrease in capillary height in both of the nicotine groups when compared to the control animals. This study indicates that morphologic alterations occur in the microvasculature of the oral mucosa following systemic nicotine administration. This may have implications for the role of chronic tobacco use in the etiology of oral mucosal disease, including periodontal disease.
