A case of extramedullary plasmacytoma of the left palatal tonsil in a 51‐year‐old woman is reported. The histological features of the tumor are typical, except for the appearance of crystal inclusions in plasma cells, histiocytes and matrices. The ultrastructural features of 12 cases of leukoedema were investigated and compared with clinically normal buccal mucosa. Histochemistry was undertaken to try to resolve some of the observed ultrastructural changes. The characteristic “intra‐cellular edema” of the epithelial cells in leukoedema is due to vacuolation in the cytoplasm of cells. Abnormal mitochondria were observed in these cells. The vacuoles contained a granular material somewhat like clumped glycogen granules, but histochemistry failed to identify this material as glycogen. Towards the surface of the epithelium, the vacuolated cells collapsed into a compact layer of flattened cells. The outer cells of this layer abruptly swelled again to form the characteristic superficial layer of “ballooning” cells of leukoedema. The latter cells were not vacuolated but contained remnants of organelles, membraned vesicles with remnants of organelles, keratohyalin granules and structures apparently related to keratohyalin granules. We propose that the vacuolation represents a limited reversible form of cellular degeneration resulting from cell damage and that impeded mitochondrial function may be the cause of the vacuolation. The superficial “ballooning” cells are degenerated cells. The flattening of the vacuolated cells into a compact layer and the presence of keratohyalin granules and keratohyalin‐like structures in the superficial cells are regarded as features of an aborted form of

Leukoplakia, lichen planus and carcinoma in situ of human oral mucosa were analysed histometrically by light microscopy. Leukoplakia lesions showed a greater total nuclear density than lichen planus lesions. They had a similar basal nuclear density to carcinomas in situ. The total and basal nuclear areas were notably greater in carcinomas in situ, indicating the presence of larger nuclei in comparison to those of other lesions. We believe it to be possible to make a differential diagnosis based on numerical variables.

The rehardening properties of four saliva substitutes on artificially softened human enamel have been investigated by microhardness measurements. The saliva substitutes were all based on the same formula, containing calcium, phosphate and fluoride as the main electrolytes, and mucin or carboxymethylcellulose as the main macromolecules. Histochemical methods for proteins were applied to crystals found in pleomorphic adenomas of the parotid. Positive reactions were obtained with methods for tyrosine, tryptophan, arginine and sulphydryl groups, and indicate a greater complexity of composition than has previously been acknowledged.

The effects of repeated stimulation of hamster oral mucosa and epidermis with 12‐0‐tetradecanoylphorbol‐13‐acetate (TPA), were examined usingin vitroassays of glycolysis and amino acid incorporation. The response of control and hyperplastic tissues to epinephrine and cAMP, and their rates of incorporation of histidine and leucine, were also examined. Repeated treatment with TPA in the concentration range of 1 × 10−8to 1 × 10−5M resulted in increased rates of glycolysis, amino acid incorporation and mitosis in both tissues. During the induction of hyperplasia, a loss of the normal epithelial response to epinephrine was observed at Day 8, suggesting re‐establishment of mechanisms controlling homeostasis. Four Ca(OH)2‐containing pulp‐capping medicaments were compared for their effects on protein synthesis in early subcultures of monkey‐ and human‐pulpal fibroblasts. While protein synthesis, as well as DNA synthesis, was depressed by three of the medicaments, the protein‐synthetic rate in human‐cell cultures in the presence of Life® rose to control levels when the medium was changed daily, but was depressed when the medium was not changed. An ultrastructural study was carried out to investigate the nature of an eosinophilic material, having the staining properties of amyloid, in a case of calcifying epithelial odontogenic tumour. At the electron‐microscopical level, the eosinophilic masses consisted of two types of structure which are probably related. The first type appeared as sheets of fine filaments measuring 10–12 nm in diameter. The second type was in the form of aggregates of lamina densa fragments, probably secreted by the tumour epithelium. These fragments appeared to undergo some loss of electron density and became degraded into fine filaments, having similar thickness and electron density to those forming the filamentous masses. Most reported studies of experimental oral cancer have been carried out in the hamster. This animal is not available in certain countries and there is controversy regarding its suitability as a model for experimental oral carcinogenesis. An attempt was made to reproduce an experimental oral cancer model in rats, using a protocol based on that described by Lekholm and Wallenius (1976). The carcinogen 4 nitroquinoline 1‐oxide was applied to the palates of rats for periods of up to 24 weeks. Macroscopic and microscopic examination of carcinogen‐treated palatal mucosa was carried out during the experimental period. Verrucous carcinoma‐like lesions of the mid‐palate and squamous carcinomas of the gingival mucosa were produced, beginning at 16 and 20 weeks respectively.
