More than 200 articles and texts relating to Chondrosarcoma in the general skeleton, in the head and neck area, and in extraskeletal soft tissues, were reviewed and summarized. In the interest of space conservation and avoidance to the extent possible, of distracting referential interjections in the text, the entire list of materials reviewed has not been referenced. For example, articles that contribute essentially nothing more than single case reports are not included in the list of references. Effort has been made, however, to include articles of historical significance, articles reporting series of cases, and articles exploring ideas and concepts. The authors believe this review to be a comprehensive summary of the Chondrosarcoma literature. An analysis of 37 cases of Chondrosarcoma of the jaws will be published as Part II.

Analysis was made of 37 cases of Chondrosarcoma of the maxilla or the mandible. The most common complaint at initial presentation was the presence of a mass. Pain was a presenting symptom in less than half of the patients. The usual roentgenographic appearance was an ill‐defined radiolucency with mottled areas of calcification. When teeth were involved, some cases showed a widened periodontal membrane space on periapical dental radiographs. Histologically, the tumors usually showed recognizable cartilage and a lobular growth pattern, but cellular evidence of malignancy was often subtle. Radical surgery was the treatment of choice. Prognosis appears best when such surgery is performed soon after the onset of symptoms. Also, patients presenting with mandibular tumors and with tumors of the better differentiated histologic grades enjoy a better survival time. Neck dissection is probably not warranted for Chondrosarcoma of the jaws. Contrary to the relationship between these two tumors in other skeletal sites, the overall prognosis for Chondrosarcoma of the jaws does not appear to be as good as that for osteosarcoma of the jaw

Herpes viruses are associated with many types of oral tumors and can be tumorigenicin vitro.The mechanisms by which they transform cells remain unknown, but it has been suggested that they contain insertion sequences. Two algorithms were developed to search DNA sequences for the presence of patterns that characterize such elements using either selection or scoring techniques. When randomly shuffled herpes virus sequences were examined each algorithm detected many such patterns but the scoring algorithm found fewer than the selection algorithm. In the herpes virus sequences that are known to contain such patterns the computer techniques found many more than previously reported, indicating that visual inspection alone is not adequate. The frequency of these patterns suggest that although the transforming regions of herpes virus DNA could contain transposable elements experimental confirmation is necessary.

The effect of the radioprotector 2‐mercaptopropionylglycine (MPG) in preventing the synthesis of radiation‐induced osteodentin was examined. Fifty rats were exposed to single x‐ray doses of between 3 and 19 Gy localized to the maxillary incisor growth centers. Half of the animals received an injection of MPG before irradiation while saline was administered to the other rats. Guinea pig epidermal keratinocytes were harvested and grownin vitro(20). The cells were treated with polyphenolic compounds, acetate/shikimate aromatics, that are ubiquitous in plants. In addition, the cells were exposed to tobacco glycoprotein (TCP), a plant‐derived substance prepared from cured tobacco (19). All of the polyphenolic compounds enhanced the attachment of cells to a defined substratum and inhibited cell proliferation. Tobacco glycoprotein (TOP), rutin conjugates and ellagic acid inhibited cell spreading and enhanced the apparent microviscosity of keratinocyte membrane lipids. These data suggest that the use of these compounds may be associated with alterations in cytoskeletal architecture and membrane fluid dynamics that might result in clinical alterations characterized by whitening of mucosa

Little data is available on regional blood flow in the normal primate oral mucosa and none on that in atherosclerotic animals. Three adult Rhesus monkeys were maintained on a normal diet and 4 on a high fat, high cholesterol diet for 20 months. Radiolabelled microspheres were used to measure blood flow in skin and 16 oral mucosa regions. In normal animals, blood flow ranged from 160.81 to 8.68 ml/min/100 gm tissue. Blood flow in the same regions of atherosclerotic animals showed significantly lower values than in controls, ranging from 65.90 to 1.04 ml/min/100 gms tissue. However, the relative blood flow to the different regions was not significantly different between control and atherosclerotic animals. Histologic examination of tissue from the atherosclerotic animals revealed gross intimal plaques occluding the lumina of the carotids and atherosclerotic lesions in the lingual arteries. It is concluded that the decrease in blood flow in oral mucosa in the atherosclerotic animals is not the result of local changes in the mucosal vasculature but may be related to lesions seen in the major afferent vessels.

Keratocysts of the solitary type were histologically and histometrically compared with those associated with the nevoid basal cell carcinoma syndrome (NBS). It was observed that parakeratinization, intramural epithelial remnants and satellite cysts were a more frequent finding among NBS keratocysts than among solitary keratocysts. A hypothetical model of the maturation pathway of Langerhans cells (LC) within an epithelial environment is presented. This model is based onin vitrostudies using human gingival organ culture. In this model, T6 (GD1) antigen is induced on a T6(‐) intraepithelial population by Interleukin‐1 secreted by epithelial cells. This process is abrogated by a locally produced Interleukin‐1 inhibitor, ILS. These T6(+) LC then express first HLA‐DR and subsequently HLA‐DQ surface antigens under the influence of either lipopolysaccharide or gamma interferon. The induction of these Class II antigens on LC is inhibited by prostaglandin E2. It is postulated that these Class II antigen positive LC are then available to function as antigen presenting cells. This hypothesis is consistent within vitrostudies and severalin vivoobservations.
