Fluoride Uptake by Enamel Surfaces, Root Surfaces and Cavity Walls following Application of a Fluoride Varnishin vitro |

Twelve root surfaces and enamel surfaces adjacent to fresh amalgam restorations were treated with the fluoride varnish Fluor Protector® for 48 h in vitro. The untreated half of each buccal surface served as a control. The varnish was removed after 48 h and the teeth immersed in artificial saliva for 20 h. Electron probe microanalysis revealed that in 75% of the recordings the fluoride concentration in the treated enamel surfaces ranged from 1,500 to 4,500 ppm, while the remaining 25% showed concentrations below the detection limit (1,500 ppm). All the treated root surfaces showed concentrations greater than 1,500 ppm; values up to 25,000 ppm were recorded. In the enamel and dentin cavity walls, fluoride concentrations above the detection limit were recorded in 5 and 55% of the cases, respectively. In the dentin walls, values up to 7,000 ppm were found. In the untreated enamel surfaces, root surfaces and cavity walls, the fluoride concentration was generally below the detection limit. This study demonstrated a considerable uptake of fluoride from the varnish by enamel and root surfaces. In addition, the cavity walls had taken up fluoride ions released from varnish applied on the tooth surface adjacent to amalgam fillings

In four separate studies, 50% aqueous solutions of sugar substitutes (150 or 200 μl) were topically applied 3 or 5 times daily on molar teeth in rats receiving a low cariogenic diet (20% sucrose) ad libitum. All animals had been inoculated with Actinomyces viscosus Ny 1, Streptococcus mutans OMZ 176 and feces from caries-active rats. No diarrhea was observed and reduced weight gains were found in only one study. The topical application of sugar substitutes was noncariogenic. However, no sugar substitute significantly and consistently reduced bacterial agglomerate (rat plaque) formation, smooth surface or fissure caries incidence versus the water controls

Bovine enamel was exposed to buffered lactic acid in hydroxyethylcellulose, hydrochloric acid or orthophosphoric acid during different periods. The resulting surface roughness was determined by a laser speckle technique, by a mechanical profilometric instrument, and from SEM stereomicrographs by applying these techniques to the same samples. The methods are compared. The results correspond reasonably well. Exposure of the enamel to hydrochloric acid or orthophosphoric acid gives a large increase of the surface roughness during the first 30 sec; further exposure gives some decrease of the roughness. Exposure to buffered lactic acid for 7 days gives hardly any change in surface roughness.

Concentrations of strontium (Sr) were given preeruptively to rats at 0.0, 0.5, 1.0, 2.5, 5.0, and 7.5 μg Sr/10 g body weight by esophageal intubation. At weaning, the same rats were given 0, 5, 10, 25, 50, and 75 mg Sr/liter via the drinking water. Caries scores were recorded for buccolingual, morsal, sulcal, and proximal lesions in all rats. Reductions in caries scores were only seen for buccolingual scores. Maximum reductions in buccolingual scores were found in rats given 7.5 μg Sr/10 g body weight preeruptively and 75 mg Sr/liter posteruptively. Uptake of strontium by enamel was significantly correlated with drinking water concentration
